    Abstract:

    Fibronectin isoforms are generated by the alteRNAtive splicing of a primary transcript derived from a single gene. In rat at least three regions of the molecule are involved: EIIIA, EIIIB, and V. This study investigated the splicing patterns of these regions during development and aging, by means of ribonuclease protection analysis. Between fetal and adult rat, the extent of inclusion of the EIIIA and/or EIIIB region in fibronectin mRNA varied according to the type of tissue analyzed; but the inclusion of the V region, and in particular the V25 alteRNAtive variant, was significantly higher in all fetal than in adult tissues. These data suggest a crucial role of the V25 variant, possibly related to its interaction with the alpha 4 beta 1 integrin receptor during development. On the other hand, during aging, the only significant change observed in the splicing pattern was a decrease in the EIIIA variant in brain. The high inclusion levels of the EIIIA and EIIIB regions in young adult brain suggest that these segments may play an important role in differentiated brain tissue. The decreasing levels of inclusion of the EIIIA segment in brain fibronectin mRNA during aging may be an age-related marker with functional consequences.

    Abstract Messenger RNA of mouse L cells is methylated in both base and ribose moieties. On the average there are about 2.2 methyl groups per 1000 nucleotides in mRNA, a proportion which is about one-sixth that of mammalian ribosomal RNA. The variety of methylated bases in mRNA is more limited than in ribosomal RNA. A very low level of methylation is detected in heterogeneous nuclear RNA, suggesting that methylation, like polyadenylation, may constitute a post-transcriptional modification of Messenger RNA Precursor in eucaryotic cells.

    Abstract:

    The Sex-lethal (Sxl) protein of Drosophila melanogaster regulates alteRNAtive splicing of the transformer ( tra ) Messenger RNA Precursor by binding to the tra polypyrimidine tract during the sex-determination process. The crystal structure has now been determined at 2.6 Å resolution of the complex formed between two tandemly arranged RNA-binding domains of the Sxl protein and a 12-nucleotide, single-stranded RNA derived from the tra polypyrimidine tract. The two RNA-binding domains have their β-sheet platforms facing each other to form a V-shaped cleft. The RNA is characteristically extended and bound in this cleft, where the UGUUUUUUU sequence is specifically recognized by the protein. This structure offers the first insight, to our knowledge, into how a protein binds specifically to a cognate RNA without any intramolecular base-pairing.
