    INTRODUCTION Synthetic Colourants are subject to various allergies. As a result, the use of plant-based additives has proved promising. OBJECTIVE It is in this perspective that our objective was undertaken to investigate Natural Colourants from edible cactus. Opuntia species populations were single-sequence repeat (SSR) fingerprinted, the analysis of betalains and phenolic compounds and the evaluation of their biological potentials were also characterised. RESULTS Photometric quantification of betalains and phenols showed an interspecific variation across Opuntia species. Opuntia ficus indica fruits showed the highest betalain [betaxanthins; 843.67 and betacyanins; 1400 mg/100 g dry weight (DW)] and polyphenol contents. Reversed-phase high-performance liquid chromatography (RP-HPLC) analysis showed that the variation of individual phenols profile was influenced by interspecific and genetic factors. Isorhamnetin-O-(di-deoxyhexosyl-hexoside) was the major compound and its content varied according to Opuntia species, while catechol was the predominant phenolic compound in O. humifusa with 1.88 μg/g DW. Concerning cactus species, Opuntia Colourants exhibited a potent antiradical activity [half maximal inhibitory concentration (IC50 ) up to 1 μg/mL]. Opuntia species were effective against Gram-positive and Gram-negative bacterial strains [inhibition zone (IZ) up to 27 mm]. A high genetic diversity within Opuntia genotypes based on SSR markers was revealed. UPGMA (unweighted pair group method with arithmetic mean) dendrogram and PCoA (principal coordinate analysis) based on Natural pigments and antimicrobial profiles indicated significant variation. The correlation approach proved the presence of a probably metabolic relationship between genetic markers, pigments and their biological activities. CONCLUSION A possible association between molecular approach and metabolic profile analysis of Opuntia allows tracing the relationship among species for its genetic conservation.

    Abstract Anthocyanins are widely distributed, glycosylated, water-soluble plant pigments, which give many fruits and flowers their red, purple or blue colouration. Their beneficial effects in a dietary context have encouraged increasing use of anthocyanins as Natural Colourants in the food and cosmetic industries. However, the limited availability and diversity of anthocyanins commercially have initiated searches for alternative sources of these Natural Colourants. In plants, high-level production of secondary metabolites, such as anthocyanins, can be achieved by engineering of regulatory genes as well as genes encoding biosynthetic enzymes. We have used tobacco lines which constitutively produce high levels of cyanidin 3- O -rutinoside, delphinidin 3- O -rutinoside or a novel anthocyanin, acylated cyanidin 3- O -(coumaroyl) rutinoside to generate cell suspension cultures. The cell lines are stable in their production rates and superior to conventional plant cell cultures. Scale-up of anthocyanin production in small scale fermenters has been demonstrated. The cell cultures have also proven to be a suitable system for production of 13 C-labelled anthocyanins. Our method for anthocyanin production is transferable to other plant species, such as Arabidopsis thaliana , demonstrating the potential of this approach for making a wide range of highly-decorated anthocyanins. The tobacco cell cultures represent a customisable and sustainable alternative to conventional anthocyanin production platforms and have considerable potential for use in industrial and medical applications of anthocyanins.

    Anthocyanins are water-soluble pigments giving the red, purple and blue colours of many flowers and fruit. In addition to their physiological roles in plants, to attract pollinators and seed dispersers, dietary anthocyanins are associated with protection against certain cancers, cardiovascular diseases and other chronic human disorders. Enhanced supplies of pure anthocyanins would service the demands of research to investigate these health-promoting effects and would also prove a valuable resource for the Colourants and cosmetic industries to investigate the effects of chemical modifications, co-pigments, and pH on colour and stability for developing new plant sources of Natural Colourants, and new Natural colours.

    Maintaining colour strength and fastness of the fabrics dyed with Natural Colourants had been the major constraint of utilizing plant based dyes in modern textile practices. The present study was concerned with the extraction of lutein dye from marigold (Tagetes erecta L.) flowers and role of gamma radiation in improving colour strength and fastness characteristics of the extracted dye. The investigation of dyed fabric in spectraflash showed that gamma ray treatment of 30 kGy was the optimum absorbed dose for surface modification to improve its dye uptake ability. Good colour strength was obtained when irradiated cotton (RC, 30 kGy) was dyed with extract of radiated marigold flower powder (RP) at 70 °C for 85 min, keeping M:L of 1:50 using dye bath of pH 5.0. The results from mordanting experiments revealed that 7% of tannic acid as pre-mordant and 5% of Cu as post-mordant were the best treatments to improve colour strength. It was found that gamma ray induced extraction of lutein from marigold flowers had a potential to be utilized as Natural dyes in textile sector to produce yellowish green shades.

    The present study was concerned with extraction of Natural Colourants from pomegranate (Punica granatum L.) rind powder and characterization of extracted dye under the influence of gamma radiations. Both dyeing powder and silk fabric were exposed to different absorbed doses (20, 25, 30, 35 and 40. kGy) of high energy radiations using Cs-137 gamma irradiator. Aqueous, alkali and alcohol solubilized extracts were obtained from un-irradiated and irradiated pomegranate rind powders. Aluminium, copper, chromium and tannic acid (2-10% each) were employed to improve colour strength of extracted dye. Exposure of pomegranate peel colourant and silk fabric to gamma radiation greatly improved the extraction and dyeing process by reducing material to liquor ratio, dyeing time, dyeing temperature and salt concentration. Gamma ray treatments also had strong influence on colour strength and fastness characteristics of silk fabrics dyed with irradiated pomegranate peel. Gamma ray treatment of 40. kGy was the most effective dose for colourant extraction from pomegranate peel and surface modification of silk fabric. The good colour strength was obtained by dyeing silk with pomegranate peel fabrics at 50. °C for 40. min, keeping dye bath pH at 6.0 with salt concentration of 10. g/L. The result from mordanting experiments indicated that 10% of aluminium as pre-mordant and 10% of tannic acid as post-mordant were the best to enhance colour strength of extracted dye. It was also observed that gamma ray treated pomegranate rind had a good potential to alternate yellow and golden synthetic dyes in textile industry. © 2014 Elsevier B.V.

    Abstract Roots of three unique Chinese radish cultivars were evaluated as potential sources for anthocyanin-type Colourants or value-added products. These cultivars showed high variation in anthocyanins (63.77–160.74 mg/100 g FW). Seventeen pigments were tentatively identified by mass spectroscopy as pelargonidin-3-sophoroside-5-glucoside derivatives with multiple acylation of hydroxycinnamic acids. A bright colour (CIELab) of radish anthocyanins has been shown at a wide pH range, comparably stable at pH   3.6, respectively. In a radish juice model (pH 5.8/2.5), thermal degradation of anthocyanins or glucosinolates was associated closely with media pH values. In conclusion, cultivar selection, and thermal and pH conditions during processing or storage should be taken into account for quality, stability, and health benefits of radish derived Natural Colourants or nutraceutical products.
