    Atherosclerosis is characterized by endothelial dysfunction, lipid deposition, fibro-proliferative reactions and inflammation. Octacosanol is a high-molecular-weight primary aliphatic alcohol. As the main component of a cholesterol-lowering drug, Octacosanol could inhibit lipids accumulation and cholesterol metabolism. To explore the indication of Octacosanol on endothelial protection, we evaluated its effects on the proliferation and migration of human umbilical vein endothelial cells (HUVEC). Cell viability assay using methyl thiazolyl tetrazolium and 5-ethynyl-2′-deoxyuridine revealed that 3.125 μg/ml Octacosanol promoted the proliferation of HUVEC. A cell migration assay indicated that 0.781 and 3.125 μg/ml Octacosanol increased the migration of HUVEC. Moreover, the phosphorylation levels of Akt and Erk1/2 were significantly elevated under exposure to Octacosanol. Blocking the activation of Akt and Erk with their potent inhibitors LY294002 and PD98059, respectively, markedly attenuated the Octacosanol-induced proliferation and migration of HUVEC. These findings demonstrated for the first time that Octacosanol enhanced the proliferation and migration of HUVEC and mediated these effects through activation of the PI3K/Akt and MAPK/Erk1/2 signaling pathways.

    Determination of the levels of 1-Octacosanol is important in food stuff for the study of its pharmacological activities and health benefits. In this study, a novel, simple and fast internal standard method for the non-derivatization ultra-performance liquid chromatographic determination of 1-Octacosanol in raw materials and health products was developed and validated based on evaporative light scattering detection. The linearity (r2 > 0.998), recovery (99.1–100.2%, RSD <2.7%), intra- and inter-day precision (RSD <3.8%), limit of detection (1.0 mg/L), limit of quantification (2.2 mg/L) of the 1-Octacosanol were determined. The method was successfully applied to nine real 1-Octacosanol products. The results of analyses had close agreement with the labeled claims of 1-Octacosanol content in these products. Compared with the classical gas chromatography method, the developed method was simpler, faster and more environmentally friendly due to avoiding any derivatization step. This protocol represents a rapid and feasible method for quality control of 1-Octacosanol products.

  • Purification process of Octacosanol extracts from rice bran wax by molecular distillation

    Abstract Molecular distillation (MD) was used to increase the purity of Octacosanol extracts from transesterified rice bran wax (RBW), and the effects of MD parameters on the purification process and efficiency were studied. The Octacosanol content, triacontanol content and the ratio of Octacosanol content ( C o ) to triacontanol content ( C t ) in distillates and residues, the yields of distillates and residues, and split ratio (yield of distillates to the yield of residues) were investigated in the circumstance of temperature and pressure changes. Results indicated that distilling temperature and distilling pressure are very important factors in purification process. The increase of distilling temperature or the decrease of distilling pressure could increase the mean free path (MFP) of molecules; therefore, the components with larger molecular weight could be separated and concentrated onto the condenser, and the yield of distillates increased and the split ratio was higher, but the content of Octacosanol in distillates did not increase accordingly. Purified by MD at 150 °C and 0.5 Torr, the Octacosanol content in distillates can be up to 37.6%, increasing by 48.0% over that in the raw Octacosanol extracts. The purification processes of Octacosanol extracts with MD were further explained by comparing the components of distillates with residues at 0.5 Torr, and the components of distillates at 0.5 Torr with those at 2.0 Torr as well.

    Abstract The purification of crude Octacosanol extract from transesterified rice bran wax was carried out by molecular distillation (MD). A central composite rotate design (CCRD) was used in order to optimize the experimental parameters: distilling temperature and vacuum degree. Through the response surface methodology (RSM), the optimal MD conditions were determined and the quadratic response surfaces were drawn from the mathematical models. The results suggest that the distillng temperature and vacuum degree significantly affected both Octacosanol content and yield of purified products, while the interaction of distilling temperature and vacuum degree was not significant in the two models. The optimum conditions of the purification of crude Octacosanol extract were: distilling temperature 176.1 °C and vacuum degree 1.29 Torr. Optimal values predicted by RSM for the Octacosanol content and yield of purified product were 25.93% and 51.09%, respectively. Close agreement between experimental and predicted values was obtained.

    UNLABELLED: Reportedly policosanols (PCs) have various beneficial functionalities on health. A gas chromatography-tandem mass spectrometry (GC-MS/MS) with a low limit of detection (LOD), and high specificity, recovery, and precision was successfully established for the PC analysis in vegetable oils. The LODs for the PCs were in the range of 0.002 to 0.016 μg/mL. The relative standard deviation (RSD) for the repeated analysis of PCs was less than 3.356%. The mean recoveries for spiked heptacosanol and Octacosanol in vegetable oil were 102.3% and 106.3%, respectively. The total PC contents in the vegetable oils varied from 3.01 to 427.83 mg/kg oil. Perilla seed, grape seed, and rice bran oils were found to be highly rich sources of PCs, containing 427.83, 245.15, and 171.17 mg PCs/kg oil, respectively. Corn, sesame, and soybean oils contained only a negligible quantity of PCs. The PC composition in vegetable oils was greatly source dependent. In perilla seed oil, Octacosanol was the single most predominant component, representing 55.93% of the total PC. In grape seed oil, however, hexacosanol is the most abundant PC, followed by Octacosanol, tetracosanol, and triacontanol in a decreasing order. The major PCs in rice bran oil were triacontanol, Octacosanol, hexacosanol, and tetracosanol, which constituted over 87.3% of the total PC. This represents the 1st report on the composition and contents of PC in most vegetable oils analyzed here. PRACTICAL APPLICATION: The information might be used for the development of vegetable oil products with beneficial functionality.
