Rainbow Smelt (Osmerus mordax) Genomic Library and EST Resources

Ewart, William R DriedzicAbstract:Genomic resources in rainbow smelt ( Osmerus mordax ) enable us to examine the genome duplication process in salmonids and test hypotheses relating to the fate of duplicated genes. They further enable us to pursue physiological and ecological studies in smelt. A bacterial artificial chromosome library containing 52,410 clones with an average insert size of 146 kb was constructed. This library represents an 11-fold average coverage of the rainbow smelt ( O. mordax ) genome. In addition, several complementary deoxyribonucleic acid libraries were constructed, and 36,758 sequences were obtained and combined into 12,159 transcripts. Over half of these transcripts have been identified, several of which have been associated with cold adaptation. These basic resources show high levels of similarity (86%) to salmonid genes and provide initial support for genome duplication in the salmonid ancestor. They also facilitate identification of genes important to fish and direct us toward new technologies for other studies in fish biology.

These loci were screened among a minimum of 79 individuals and the average number of alleles per locus was 16 with an average heterozygosity of 0.86. These microsatellites are being used to examine population structure and life-history evolution among natural populations of rainbow smelt.

Veterinary Parasitology, 2011Co-Authors: Chaoqun Yao, Donal Otoole, Mike Driscoll, Warner Mcfarland, Jonathan H Fox, Todd E Cornish, W R JolleyAbstract:Infections of domesticated dogs by a worldwide parasitic nematode Filaroides osleri (Oslerus osleri) lead to verminous tracheobronchitis that are often misdiagnosed clinically as kennel cough, due to infection with the bacterium Bordetella bronchiseptica. Diagnosis of two canine cases in Wyoming, USA prompted a search of the literature of canid infections in North America. Infections of domestic dogs are reported in nine US states and four Canadian provinces. Dogs of multiple breeds and both sexes were infected. Most were two years old or younger at diagnosis. Anthelmintic treatments were effective in relieving clinical symptoms, as well as causing resolution of tracheobronchial nodules. Other canid species, including coyotes (Canis latrans) and wolves (Canis lupus), have been infected across North America with a prevalence of 23% and 4%, respectively. Infection with F. osleri should be included in the differential diagnosis of infectious tracheobronchitis of dogs. It can be confirmed most readily by endoscopic detection of distinctive submucosal parasite-filled nodules, combined with histological examination of endoscopic biopsies.

We estimated prey selectivity by comparing prey numerical abundance estimates from spring bottom trawl surveys and lake trout diet information in similar areas from spring gill net surveys con-ducted annually in Lake Superior. The model simulates food-web interactions among linked populations of O. mordax, Coregonus artedi and Sander vitreus. Simulations indicated that O. mordax dominate where: (1) adult O. mordax prey on young-of-the-year (YOY) C. artedi, (2) YOY O. mordax negatively affect YOY S. vitreus through competition and (3) adult S. vitreus experience moderate fishing mortality. Osmerus mordax dominated simulations across a broad range of variable values that regulated competition and predation, and displayed threshold responses to increasing angler harvest. Consequently, angler harvest should be carefully managed in lakes susceptible to O. mordax invasions because the alternative could lead to fishery collapse.
