Apoptosis in Human Osteosarcoma Cell Line SOSP-9607 Induced by Caspase-6

Methods: The expression level of caspase-6 in the Osteosarcoma Cell Line SOSP-9067 was examined by RT-PCR method. The adeno-virus adv5 vector with caspase-6 gene was constructed and transferred into the Osteosarcoma Cell Line SOSP-9607 by lipo-fection. The Cell survival rate after transfection was assayed by MTT method. The Cell morphological changes were observed by microscope and electron microscope, the apoptosis of transferred Cells were examined by gel electrophoresis. Results:No expression of caspase-6 was examined in the Osteosarcoma Cell Line SOSP-9607. A high expression of caspase-6 was identified by RT-PCR after the transfection. The Cell growth curve decLined after transferring caspase-6. Electrophoresis of DNA displayed the apoptosis ladder. Conclusion: Transferring caspase-6 into the Osteosarcoma Line SOSP-9607 may inhibit the growth of the Osteosarcoma Cell Line SOSP-9607 and this effect might be achieved by inducing apoptosis.

The gene silencing mediated by RNAi on survivin expression in Osteosarcoma Cell Line MG-63
China Oncology, 2007Co-Authors: Zhang WeiAbstract:Background and purpose:Osteosarcoma is the most common malignancies with high degree of malignancy and poor prognosis in young people.Survivin is a member of the inhibitor of apoptosis(IAP) protein gene family and its expression is related to the degree of malignancy and prognosis in Osteosarcoma.The experiment was to study the inhibition of expression of survivin gene by synthesized short survivin-siRNA in Osteosarcoma Cell Line MG-63. Methods:Chemically synthesized short survivin-siRNA was transfected into the Osteosarcoma Cell Line MG-63.The survivin mRNA and protein level were detected by reverse transcription-polymerase chain reaction(RT-PCR) and flow cytometry(FCM),the biological morphology and growth inhibition of MG-63 were observed with light microscopy and MTT assay,respectively.Results:Short siRNA targeting survivin down-regulated the transcription of survivin oncogene and protein level.The proliferation of Osteosarcoma Cell Line MG-63 was inhibited after transfection.Conclusions:Chemically synthesized short survivin-siRNA can effectively inhibit survivin expression and Cell proliferation in Osteosarcoma Cell Line MG-63.Short survivin-siRNA may be a useful therapeutic strategy for cancer through the regulation of gene expression.

Effects of exogenous wild type p53 on behaviors of Osteosarcoma Cell Line

Methods:DNA transfection technique was used to introduce exogenous recombinant wild type p53 plasmid,named as PM47,into Osteosarcoma Cell Line HOS 8603. After culturing in selection medium,survivors were selected.PCR was used to detect the existence of PM47 in transfected tumor Cells.The morphology of untransfected and transfected tumor Cells was observed under contrast microscope and light microscope,and growth features were assessed. In situ Cell death detection method was used to detect apoptotic Cells. Flow cytometry was used to analyze the DNA contents and Cell cycle of the cancer Cells. Then,immuno histochemical technique was used to detect expression of p53,p21/WAF1,bax,bcl 2 proteins in untransfected and transfected tumor Cells.Results:Phenotype of transfected tumor Cells changed into long spindle shaped from untransfected epithelial like Cells.The transfected tumor Cells grew slowly,detached easily from the dishes and some were going into apoptosis.The transfected tumor Cells mostly arrested in G 1 phase by analysis of Flow Cytometry. The immunohistochemical results indicated that the p21/WAF1,bax proteins were detected stronger positive,but expression of bcl 2 protein was suppressed in trasnsfested tumor Cells.Conclusion:Wild type p53 induces expression of p21/WAF1,acts as an inhibitor of Cell cycle progession,inhibits expression of bcl 2,to result in growth inhibition,induces expression of bax to trigger apoptosis.

Effect of kappa-selenocarrageenan inducing apoptosis of Osteosarcoma Cell Line in vitro

METHODS:Human Osteosarcoma Cell Line was cultured with kappa-selenocarrageenan. Thiazolyl blue (MTT )test, double fluorescence staining of acridine orange (AO) and ethidium bromide (EB), transmission electron microscopy (TEM) and flow cytometry were applied to detect the apoptosis rate and Cell cycle.RESULTS:Osteosarcoma Cells affected by 0,30,60 and 120 μg /mL Kppa-Selenocarrageenan after 1,2,4 and 6 d respectively had obvious descending growth curve,P0.05. The morphologic examination showed early and late apoptosis Cells. Moreover, the Cell cycle analysis revealed Kappa-Selenocarrageenan blocked S phase of Osteosarcoma Cell cycle.CONCLUSIONS:Kappa-Selenocarrageenan can induce apoptosis of human Osteosarcoma Cell in vitro.
