    Abstract:

    Osteoporosis is a chronic skeletal disease characterized by low bone mass and microarchitectural deterioration with a consequent increase in bone fragility and fracture risk. The aim of the study was to evaluate the effects of honey (Apis dorsata) supplements on increased bone strength in ovariectomized rat as animal models of osteoporosis. Twenty female rats at 3 months of age, weighing 150-200 g were used in the study. The rats were divided into five groups (n=4) : Sham operation group (SH); Ovariectomy group no treatment(OVX); Ovariectomy with treatment Apis dorsata 1g/Kg BW (AD-1); Ovariectomy with treatment Apis dorsata 2g/Kg BW (AD-2); Ovariectomy with treatment Apis dorsata 4g/Kg BW (AD-3). The treatment started to be given the next day after Ovariectomy operation for 12 weeks. The Rats were sacrified within 12 weeks, and then the right femur were taken bone strength test. Based on the statistical analysis of the bone strength test, the greatest score belongs to the Sham operation group (SH) that have significant difference (p 0,05). In conclusion, honey (Apis dorsata) supplements has the effect of increasing bone strength in ovariectomized rat as animal models of osteoporosis, so that honey (Apis dorsata) supplements has the potential to be used as an alternative treatment for osteoporosis.Osteoporosis is a chronic skeletal disease characterized by low bone mass and microarchitectural deterioration with a consequent increase in bone fragility and fracture risk.     This study aims to determine the potential of the Cissus quadrangularis extract to decreased the expression of IL-1 β so that it can to inhibit bone resorption and osteoporosis fracture healing is not delayed union. Forty rats adapted for 1 week, and then divided into four groups with 10 replications. P0 (sham-operated + Osteotomy + CMC Na); P1 (Ovariectomy + Osteotomy + CMC Na); P2 (Ovariectomy + Osteotomy + Raloxifene 5.4 mg /kg); P3 (Ovariectomy + Osteotomy + CQ 750 mg / kg). Osteoporosis induced is made by bilateral Ovariectomy in rats, then to make sure that osteoporosis is already happened, 8 weeks post-Ovariectomy performed radiology examinations on the femur, then the osteotomy action. IL1-β expression observations performed in at the 2nd week and 6th week after osteotomy through the immunohistochemistry examination.     We measured the effects of Ovariectomy on the bone mass of mice that lacked type I interleukin-1 receptor (IL-I R1 -/- mice) in two genetic backgrounds (C57BL/6 x 129/Sv and C57BL/6) to investigate the role of interleukin-1 in the actions of estrogen on bone. At three weeks after surgery, ovariectomized wild-type mice decreased trabecular bone volume in the proximal humerus by 70% in a C57BL/6 x 129/Sv background and 48% in a C57BL/6 background compared to sham-operated controls. In contrast, there was no significant decrease in trabecular bone mass in IL-1 R1 -/- mice after Ovariectomy. The estrogen status of all groups was confirmed by measurement of uterine wet weight. These results demonstrate that a functional IL-1 response pathway is required for mice to lose trabecular bone mass after Ovariectomy in this model and they imply that IL-1 is an important mediator of the effects of Ovariectomy on bone mass. Hence, therapeutic interventions that block the effects of IL-1 on bone may be beneficial for treating diseases of rapid bone loss such as post-menopausal osteoporosis.

    : Bone loss induced by Ovariectomy is due to the direct activity on bone cells and mesenchymal cells and to the dysregulated activity of bone marrow cells, including immune cells and stromal cells, but the underlying mechanisms are not completely known. Here, we demonstrate that Ovariectomy induces the T-cell co-stimulatory cytokine LIGHT, which stimulates both osteoblastogenesis and osteoclastogenesis by modulating osteoclastogenic cytokine expression, including TNF, osteoprotegerin, and the receptor activator of nuclear factor-κB ligand (RANKL). Predictably, LIGHT-deficient (Tnfsf14-/- ) mice are protected from Ovariectomy-dependent bone loss, whereas trabecular bone mass increases in mice deficient in both LIGHT and T and B lymphocytes (Rag -/- Tnfsf14 -/- ) and is associated with an inversion of the TNF and RANKL/OPG ratio. Furthermore, women with postmenopausal osteoporosis display high levels of LIGHT in circulating T cells and monocytes. Taken together, these results indicate that LIGHT mediates bone loss induced by Ovariectomy, suggesting that patients with postmenopausal osteoporosis may benefit from LIGHT antagonism. © 2020 Pathological Society of Great Britain and Ireland.
