    Abstract:

    The effect of culture age and various cooling rates on the survival and regrowth of cryopreserved cell cultures of Papaver Somniferum L. is described. Survival and regrowth following cryopreservation, in liquid nitrogen, occurred only with 4 and 5 day old cultures. No regrowth was observed in 3 or 6 day old cultures. Cooling rate had a significant effect on survival following cryopreservation with an optimum cooling rate of 0.5 degrees C min (-1).

    Abstract:

    Abstract Cell suspension cultures derived from Agrobacterium rhizogenes-transformed Papaver Somniferum tissue were investigated for growth and alkaloid content. Transformed cultures grown in phytohormone-free medium, and cultures derived from non-transformed tissue, showed little difference in growth. Both transformed and untransformed cell lines accumulated high quantities of benzylisoquinoline alkaloids, of which the major species was sanguinarine. These cell lines also produced a variety of minor alkaloids, including the sanguinarine analogues, oxysanguinarine, norsanguinarine and dihydrosanguinarine, and the protopine-type alkaloids, protopine and cryptopine. Although these transformed suspension cultures were derived from axenic root tissue, a rich source of morphinan alkaloids, there was no evidence of stable production of morphinan alkaloids in the cultures.
