Volatile fingerprinting of the Plum brandies produced from different fruit varieties

In this work, Plum brandies distillated by the same technology from 25 Plum cultivars were distinguished by comprehensive two-dimensional gas chromatographic analysis and sensory evaluation. The number of identified compounds in studied samples varied between 91 and 195 depending on the Plum variety. Enriched volatile organic compounds (VOC) profile was identified for the samples received from “Chrudimer”, “Cacak Fruitful” and “Hanita” Plum varieties, whereas in the case of “Gabrovská”, “Presenta”, Elena” and “President” Plum varieties, the VOC profile was significantly reduced. From qualitative point of view, the particular Plum brandies showed differences in the presence of unsaturated fusel alcohols (e.g. 3-methyl-3-buten-1-ol, trans -3-hexenol), unsaturated aldehydes (2-butenal, 2-nonenal), monoterpene derivatives (e.g. linalool acetate, geraniol acetate) and lactones, which were mainly detected at the trace level.

In addition, genetic diversity and genetic structure was studied in three Plum species (P. domestica L., Prunus cerasifera Ehrh. and Prunus spinosa L.) using chloroplast DNA (cpDNA) markers and five single sequence repeat (SSR) loci. Based on phenotypic traits, some varieties, such as mirabelle Plums, grouped together. Bayesian structure analysis was used to identify different genetic groups, whereby damson Plums were clearly distinguished from greengage Plums. When examining the three species together, a higher level of cpDNA allelic richness was found in P. cerasifera and in P. spinosa than in P. domestica where only five cpDNA haplotypes were detected in the national Plum collection, with one main haplotype that accounted for 80% of the varieties studied. P. domestica cpDNA haplotypes tended to group together with P. cerasifera haplotypes whereas most of P. spinosa haplotypes formed a separate cluster. SSR markers were somewhat able to distinguish the three species. These results provide some clues as to the origin of Plum and the various Plum varieties. Our results also provide useful information for the management of Plum genetic resources.

Plum sectorial resistance to Plum pox virus is graft transmissible

Sectorial resistance is the resistance which is manifested in a sector or portion of infected plant, where virus is not present. Sectorial resistance to PPV is exhibited in the crown or in primary shoots of the Plum tree. Double budding method was applied. One bud was carrying sectorial resistance to PPV, while the other one was infected with the virus. Infection on shoots was evaluated by symptoms appearance and serological test for PPV presence.Among three Plum cultivars the best results were obtained with cv. “Crvena ranka“. Sectorial resistance to PPV has been successfully transmitted by buds of “Crvena ranka“. Plum cv. “Čačanska rodna“ failed to transmit sectorial resistance. Plum cv. “Stanley“ was between those two above-mentioned Plum cultivars

The elimination of Plum pox virus in Plum cv. Bluefree and apricot cv. Hanita by chemotherapy of in vitro cultures

Hanita infected with Plum pox virus (PPV) were used for the virus elimination by chemotherapy. Low ribavirin concentrations of 5 and 10 mg/l in Murashige-Skoog medium were applied in the treatment. Plum pox virus was completely eliminated by 5 mg/l of ribavirin in Plum cv. Bluefree within twenty weeks and in apricot cv. Hanita in twelve weeks of the application. Plum pox virus was completely eliminated by 10 mg/l of ribavirin both in Plum cv. Bluefree and apricot cv. Hanita within twelve weeks. The presence of PPV was not proved by RT-PCR. Clones of Plum cv. Bluefree and apricot cv. Hanita were re-tested by RT-PCR one year after the termination of the ribavirin treatment and negative results confirmed the elimination of Plum pox virus.
