    Abstract:

    Objective: A new simple and precise stability-indicating bioanalytical reverse-phase high-performance liquid chromatography (RP-HPLC) method was developed and validated for the simultaneous estimation of prazosin and Polythiazide in their formulation and in human plasma. Materials and Methods: The developed method was successfully used for assaying drug contents in plasma. Isocratic elution mode was carried at Agilent C18 column (150 mm × 4.6 mm, 5 μm particle size) using 0.01 N disodium hydrogen phosphate pH 5.0:acetonitrile (55:45) as mobile phase at flow rate 1.0 ml min at detection wavelength 265 nm. Celecoxib was taken as an internal standard. The method was validated as per ICH guidelines. Results and Discussion: The present validated method can be successfully applied for the estimation of prazosin and Polythiazide in human plasma over the concentration range of 12.5–500 ng/m of prazosin and 6.25–250 ng/ml of Polythiazide. The method for the determination of prazosin and Polythiazide in human plasma using HPLC detection met the acceptance criteria with respect to selectivity, precision, accuracy, linearity, and recovery. Conclusion: The proposed method is simple, rapid, accurate, precise, and appropriate for pharmacokinetic and therapeutic drug monitoring in the clinical laboratories.

    . Thirty-eight men with untreated essential hypertension in WHO stage 1 or II, aged 19–57 years, all working, have been studied ambulatorily. Oxygen consumption, heart rate, cardiac output (Cardiogreen) and intra-arterial brachial pressure were recorded at rest in supine, sitting and standing position and during steady state work. At rest plasma volume and serum electrolytes were measured. The subjects were divided into four groups: I, untreated 7; II, chlorthalidone 100 mg every second day 9; III, Polythiazide 1 mg every second day 7; and IV, hydrochlorothiazide 50 mg twice daily 15. The subjects were restudied after 8–12 months. In the untreated group none of the results at the second examination were significantly different from those at the first. In the treated groups the causal resting blood pressure and intra-arterial pressure at rest and during work were significantly lower after treatment than before. At rest when sitting, the mean arterial pressure was reduced 21, 15 and 18% in groups II, III and IV and during work a little less. Serum-K dropped significantly in all groups, the mean drop being about 0.7 mEq/1 in all groups. Plasma volume (hydrochlorothiazide group only) was 0.24 1 lower after therapy, but the difference was not significant. In the thiazide groups the pressure drop was usually associated with a drop in peripheral resistance, both at rest and during exercise. In the chlorothalidone group the pressure drop was usually caused by a reduction in cardiac index, with little or no effect on the peripheral resistance.

  • Sustained Antihypertensive Response with Minizide: Long-Term Follow-Up in a Multicentre Study

    Abstract:

    The safety and effectiveness ofprazosin hydrochloride combined with poly-thiazide for the treatment of hypertension was studied in four European countries by ten investigators. One-hundred and seventy-sevenadult male and female patients. 91 % with essential hypertension, were treatedfor an average of 7 months. Reductions from placebo-controlled baseline blood pressure exceeded 15 % over the 8-week short-term phase of the study with further reductions ofup to 22 % observed at the end ofthe study. Ninety-six per cent of all patients completed therapy with a diastolic blood presssure of 90 mm Hg or less. Most patients were adequately controlled on 2 tablets or less once daily. Each tablet contained 0·5 mg prazosin hydrochloride and 0·25 mg Polythiazide. Long-term toleration was excellent. Side-effects were typical of those expected with antihypertensive medication and nearly all were reported during the first 8 weeks of treatment with no interruption or discontinuation ofdrug. Nearly all patients werefollowed throughout for laboratory changes. There were minimal changes consistent with the therapy and there was no pattern indicative oftoxic potential. It is concluded that the drug combination, Minizide, is effective and well tolerated as initial and long-term therapy in hypertension and that the convenient dosage regimen will lead to enhanced treatment compliance for this chronic condition

    Abstract A second-derivative spectroscopic method for the simultaneous determination of Polythiazide and prazosin in tablets was developed. Solutions of these tablets in 0.2 N methanolic hydrochloric acid were analyzed by measurement of the amplitudes of the positive peak at 356 nm with respect to the negative peak at 346 nm ( 2 D 346 ,356) for prazosin, and the amplitude of the positive peak at 236 nm with respect to the baseline ( 2 D 236 ) for Polythiazide. Confirmation of the technique was established using HPLC. The use of derivative spectroscopy in this manner provides a specific, rapid and accurate determination of the binary mixture in the tested concentration range of 1–10 μg/ml for Polythiazide and 1–20 μg/ml prazosin.
