This project evaluated the effectiveness of AFEX pretreatment for converting moso bamboo (Phyllostachys heterocycla var. pubescens) to fermentable sugars, both with and without pre-soaking in hydrogen peroxide. Pretreatment conditions including temperature, water loading, residence time, ammonia loading, and hydrogen peroxide loadings were varied to maximize hydrolysis yields. The optimal conditions for AFEX were 150°C, 0.8 or 2.0 (w/w) water loading, 10-30 min residence time, and 2.0-5.0 (w/w) ammonia loading. The optimal conditions for H-AFEX were same AFEX conditions with 0.7-1.9 (w/w) 30% (wt) hydrogen peroxide solutions loading. Using 15 FPU/g glucan cellulase and under optimal conditions, AFEX pretreatment achieved a theoretical sugars yield of 64.8-72.7% and addition of hydrogen peroxide Presoaking increased the yield to 83.4-92.1%. It is about 5-fold and 7-fold increase in sugars yield for AFEX-treated and H-AFEX-treated bamboo respectively.

The absolute reason for this remains unclear, with contamination after harvesting and preparation of the graft being the most accepted hypothesis.Using the rationale that a contaminated graft could be the main factor in postoperative septic arthritis and in an effort to maximize the antibiotic efficacy of the graft, the Vancomycin Presoaking technique was developed. It has shown success in decreasing the infection rate in ACL reconstruction. In recent years, an important number of research articles using this protocol have appeared, but the technique is still not widely implemented.Recent literature shows that Vancomycin Presoaking of the graft has shown a successful decrease in the infection rate after hamstring autograft ACL reconstruction. It has also shown efficacy decreasing the infection rate in other types of grafts (patellar tendon, quadriceps tendon, allograft) and also in patients with concomitant ligament procedures or open surgeries.Despite the positive effects of Vancomycin Presoaking reducing the infection rate after ACL reconstruction, the lack of prospective randomized control trials and the heterogeneity of the different studies mean it is not feasible to recommend Vancomycin Presoaking of the graft universally for every ACL reconstruction patient. Our group started Presoaking grafts with Vancomycin in November 2015 to decrease this risk.Purpose:The objective of the study is to compare the success of this protocol against the immediate previous period in which we did not use the protocol.Study design:Retrospective cohort. Level III.Methods:Consecutive periods were studied: April 2013-October 2015 (pre Vancomycin protocol) and November 2015- May 2018 (Vancomycin protocol). All patients that underwent a hamstring autograft primary ACL reconstruction during the periods studied were included. The final outcome was the presence of postoperative septic arthritis in both groups.
