    Abstract:

    Nine randomized, double-blind, placebo-controlled studies of major depressive disorder were pooled to evaluate the effects of desvenlafaxine 50- and 100-mg/d on energy and lassitude in adults with major depressive disorder ( n=4279). Changes from baseline to endpoint in 17-item Hamilton Rating Scale for Depression (HAM-D17) Work and Activities, Retardation, and Somatic Symptoms General items, HAM-D17 psychomotor Retardation Factor, and Montgomery-Asberg Depression Rating Scale Lassitude item were analyzed with a mixed model for repeated measures analysis of variance. Associations between residual energy measures and functional impairment, based on the Sheehan Disability Scale, were modeled using stepwise multiple linear regression. Improvement from baseline was significantly greater for both desvenlafaxine doses versus placebo on all energy symptom outcomes at week 8 (all p⩽0.005). Both early improvement in HAM-D17 psychomotor Retardation at week 2 and residual energy symptoms at week 8 were associated with Sheehan Disability Scale total score at week 8 (all p⩽0.001). Among Sheehan Disability Scale remitters and responders, the HAM-D17 psychomotor Retardation score at week 8 was significantly lower with desvenlafaxine (both doses) than placebo. Desvenlafaxine 50 and 100 mg/d significantly improved energy and lassitude symptoms in patients with major depressive disorder. Both early improvement in energy and fewer residual energy symptoms were associated with functional improvement.

    Abstract:

    A new, simple, sensitive, precise and robust high performance thin layer chromatography (HPTLC) method was developed for the estimation of eflornithine hydrochloride (DFMO) in pharmaceutical dosage forms. Estimation was performed on TLC aluminum plates precoated with silica gel 60F254 as stationary phase. Linear ascending development was carried out in twin trough glass chamber saturated with mobile phase consisting of methanol: chloroform: acetic acid: 1% triethylamine (4:6:0.1:0.5 v/v/v) at room temperature (25±2°C). Analysis of the plate in absorbance mode at 220nm was carried out. The system was found to give compact spots for DFMO with Rf (Retardation Factor) value of 0.55 respectively. The data for calibration plots showed good linear relationship with correlation coefficient of 0.999 in the concentration range of 300–800ng mL-1 for DFMO respectively. The values of limit of detection (LOD) were 0.6238ng mL-1 and limit of quantification (LOQ) were 1.8903ng mL-1 for DFMO respectively. The accuracy of the method was 100.44%. The precision demonstrated a relative standard deviation of less than 2.5%. The results were satisFactory when compared with the literature. This new method was validated according to the International Conference on Harmonization (ICH) guidelines which include linearity, precision, accuracy, specificity, robustness, detection and quantitation limits. The developed methods found to be sufficiently precise and reproducible for established conditions and after validation may be used for routine analysis of eflornithine hydrochloride in pharmaceutical formulations.

Amongst all four Factors, volume of methanol in mobile phase appeared to have a possibly significant effect on retention Factor of miconazole nitrate compared with the other two drugs nadifloxacin and mometasone furoate, and therefore it was important to be carefully controlled. In summary, a novel, simple, accurate, reproducible, and robust high-performance thin-layer chromatographic method was developed, which would be of use in quality control of these cream formulations.

    Although the removal of arsenic(V) (As(V)) from solution can be improved by forming metal-bearing coatings on solid media, there has been no research to date examining the relationship between the coating and As(V) sorption performance. Manganese-coated bone char samples with varying concentrations of Mn were created to investigate the adsorption and desorption of As(V) using batch and column experiments. Breakthrough curves were obtained by fitting the Convection-Diffusion Equation (CDE), and Retardation Factors were used to quantify the effects of the Mn coatings on the retention of As(V). Uncoated bone char has a higher retention Factor (44.7) than bone char with 0.465 mg/g of Mn (22.0), but bone char samples with between 5.02 mg/g and 14.5 mg/g Mn have significantly higher retention Factors (56.8–246). The relationship between Retardation Factor (Y) and Mn concentration (X) is Y = 15.1 X + 19.8. Between 0.2% and 0.6% of the sorbed As is desorbed from the Mn-coated bone char at an initial pH value of 4, compared to 30% from the uncoated bone char. The ability of the Mn-coated bone char to neutralize solutions increases with increased amounts of Mn on the char. The results suggest that using Mn-coated bone char in Permeable Reactive Barriers would be an effective method for remediating As(V)-bearing solutions such as acid mine drainage.
