  • Determination of the Immune Activity of Sheep Placenta Extracts by Erythrocyte Rosette Test

    Abstract:

    The immune activity and the best active concentration of sheep placenta extract were assayed by erythrocyte Rosette Test.Three concentration gradients were set up to use erythrocyte Rosette method to assay the optimal concentrations of thymus lymphocytes and sheep red blood cells.The lymphocytes were put into the 45 ℃ water bath for 45min to destroy its receptor,and then compared with the lymphocytes without de-receptor by determining the concentration of sheep placenta extract by erythrocyte Rosette Test.Meantime,four concentration gradients of thymus lymphocytes were set up to assay the concentration when the immune activity of sheep placenta extract is the highest.The results showed that the sheep placenta extracts had high immune activity and it reached the highest when the cell concentration of basic solution was 10-2 /ml.

    To improve the conditions of T lymphocyte-E-Rosette Test for examining the biological activity of thymosin,the effects of different doffed E receptor methods,the ratio of lymphocytes to sheep red blood cells,the pH value of Hank's solution,and the centrifugal speed on the E-rose formation were compared and the dyeing or counting time that influence the observation in the E-rose formation was studied.The E-rose formed from different sources of lymphocytes and different red blood cells was also observed.It was found that the optimied doffed E method could reflect the biological activity of thymosin,and the pH value of Hank's solution between 7.0~7.5 was the best result of rose formation.The best ratio of lymphocytes to sheep red blood cells was 1∶20,the centrifugal speed should be set at 800r/min,the sample should be put at 4℃ after dying,and it should be observed in 16~20h.The thymosin of guinea pig and the rabbit red blood cell,the thymosin of calf and sheep red blood cell both have the pertinency to form E-rose.The improved conditions can increase the accuracy rate when examinind the biological activity of thymosin by T lymphocyte-E-Rosette Test.
