Tianjin Journal of Traditional Chinese Medicine, 2020Co-Authors: Coe ProgramAbstract:In part I,we reported that the repetitive administration of Shaoyao-Gancao-Tang(SGT)increased the bioavailability of glycyrrhizin(GL)in SGT.In clinic,SGT is sometimes used together with some synthetic drugs.In this partⅡ,the influences of co-administered synthetic drugs on the pharmacokinetic fate of GL from SGT were investigated.The results showed that the co-administration of either a histamine2-receptor antagonist(cimetidine)or an anticholinergic drug(Scopolamine Butyl Bromide,SBB)did not clearly influence the bioavailˉability of GL in SGT.However,the co-administration of a triple therapy OAM consisting of a proton pump inˉhibitor(omeprazole,OPZ)and two antibacterial drugs(amoxicillin,AMPC,and metronidazole,MET)markedly reduced the plasma GA concentration.The reduction of plasma GA by OAM was shown to be due to the anˉtibacterial drugs AMPC-MET,not the proton pump inhibitor OPZ.The mechanism of the reduction by AMˉPC-MET involved the severe decrease of the GL-metabolizing activity of intestinal bacteria by AMPC-MET.These results suggest that it is not appropriate to simultaneously administer AMPC-MET with SGT.

The influence of commonly prescribed synthetic drugs for peptic ulcer on the pharmacokinetic fate of glycyrrhizin from Shaoyao-Gancao-tang.
Biological & Pharmaceutical Bulletin, 2001Co-Authors: Ju-xiu He, Teruaki Akao, Takao Nishino, Tadato TaniAbstract:: The influence of synthetic drugs prescribed for peptic ulcer on the pharmacokinetic fate of glycyrrhizin (GL) from Shaoyao-Gancao-tang (SGT, a traditional Chinese formulation, Shakuyaku-Kanzo-to in Japanese) was investigated in rats. Co-administration of histamine H2-receptor antagonist (cimetidine) and anticholinergic drug (Scopolamine Butyl Bromide) with SGT didn't influence the area under the plasma concentration-time curves (AUC) of glycyrrhetic acid (GA), an active metabolite derived from GL in SGT. The AUC of GA from SGT were significantly reduced by co-administration of synthetic drugs commonly used for peptic ulcer in a triple therapy (OAM), a combination of a proton pump inhibitor (omeprazole) and two antibiotics (amoxicillin and metronidazole). We found that the reduction of AUC in OAM treatment was due to the antibacterial effect of amoxicillin and metronidazole on intestinal bacteria in rat which lead to the decrease of GL-hydrolysis activity. The present study suggests that it may not be a proper way to use triple therapy containing antibiotics simultaneously with SGT for healing of chronic ulcers.

Determination of Hyoscine Butyl Bromide in Liquid Oral Solutions by Using HPLC-DAD

Al-fakhoryAbstract:A simple and rapid analytical method for routine quantification of Hyoscine Butyl Bromide (Scopolamine Butyl Bromide) in liquid oral formulations by high- performance liquid chromatography was developed and validated. The oral solution samples can be directly analyzed by reversed-phase HPLC on Intersil ODS-3 column (4.6 mm x 250 mm, i.d., 5-micron particle size) with acetonitrile- phosphate buffer of pH= 6.80± 0.05 (30:70 v/v) respectively as a mobile phase that has a flow rate of 1.5 mL /min. UV Detection was carried out at 210 nm. The method was linear over the selected concentration and ranged from 0.02 to 0.14 mg/mL (r 2 = 0.9992) and the detection limit was 0.0012 mg/mL. The mean percent recoveries ranged from 99.0% to 100.09%. The results demonstrated good linearity, accuracy, and precision. The method was applied to determine commercial liquid oral formulations.

Comparison of anti-cholinergic drug phencynonate and Scopolamine on the evoked potential and the long-term potentiation in the dentate gyrus of hippocampus in anesthetic rats
Ningxia Medical Journal, 2020Co-Authors: Peng Xiao-dongAbstract:Objective To study the effect of the anti-cholinergic drug Phencynonate and Scopolamine on extracellular field evoked potentials and its LTP in the dentate gyrus of hippocampus in urethane-anaesthetized rats.Methods Single pathway recording of evoked field potentials was made from the dentate granule cells of hippocampal hemisphere in response to stimulation of the ipsilateral medial perforant path(MPP).The amplitude of population spike(PS amplitude),was employed to evaluate the effects of drug on the overall changes in cellular responses.Results Phencynonate hydrochloride(0.2-17.4 mg/kg,i.p.) could potentiate basal synaptic transportation by increasing the PS amplitude.Scopolamine Butyl Bromide(0.3-20 mg/kg,i.p.) could not affect the basal PS amplitude.Phencynonate hydrochloride and Scopolamine Butyl Bromide did not alter either the PS amplitude LTP by conditioned tetanus(60Hz,30 pulses).Conclusion Anti-cholinergic drug Phencynonate can potentiate the excitability of the cells in dentate gyrus of hippocampus,while Scopolamine could not potentiate the excitability in the same test situation.
