Given the dynamic changes in immune cell subsets and tissue organization, which occur in GVHD, localization and timing of critical immunological events in vivo may reveal basic pathogenic mechanisms. To this end, we transplanted luciferase-labeled allogeneic splenocytes and monitored tissue distribution by in vivo bioluminescence imaging. High-resolution analyses showed initial proliferation of donor CD4+ T cells followed by CD8+ T cells in secondary lymphoid organs with subsequent homing to the intestines, liver, and Skin. Transplantation of purified naive T cells caused GVHD that was initiated in secondary lymphoid organs followed by target organ manifestation in gut, liver, and Skin. In contrast, transplanted CD4+ effector memory T (TEM) cells did not proliferate in secondary lymphoid organs in vivo and despite their in vitro alloreactivity in mixed leukocyte reaction (MLR) assays did not cause acute GVHD. These findings underline the potential of T-cell subsets with defined trafficking patterns for immune reconstitution without the risk of GVHD.

Higher intensity of itching has been associated with depth of the wounds and specific body locations but these differences are not well understood. Our aim was to investigate the intensity of post burn pruritus in grafted and non-grafted burns across anatomic areas and to report on itch-inducing factors and applied treatments. The study included 226 patients prospectively followed for 18 months. Results showed that grafted patients and non-grafted patients reported similar overall itch intensity in-hospital. At 3 months post burn, grafted patients had higher overall itch scores, a difference that was found robust across the study period. Grafted wounds were found to produce higher mean itch intensity at 3 months post burn but this difference disappeared at 12

Skin grafting may be necessary to close nonhealing Skin wounds. This report describes a fast and minimally invasive method to produce minced Skin suitable for Transplantation to Skin wounds. The technique was evaluated in an established porcine Skin wound healing model and was compared to split-thickness Skin grafts and suspensions of cultured and noncultured keratinocytes. Materials and methods. The study included 90 wounds on 3 pigs. Fluid-treated full-thickness Skin wounds were grafted with minced Skin, split-thickness Skin grafts, noncultured keratinocytes, or cultured keratinocytes. Controls received either fluid or dry treatment. The wound healing process was analyzed in histologies collected at Days 8 to 43 postwounding. Wound contraction was quantified by photoplanimetry. Results. Wounds transplanted with minced Skin and keratinocyte suspension contained several colonies of keratinocytes in the newly formed granulation tissue. During the healing phase, the colonies progressed upward and reepithelialization was accelerated. Minced Skin and split-thickness Skin grafts reduced contraction as compared to keratinocyte suspensions and saline controls. Granulation tissue formation was also reduced in split-thickness Skin-grafted wounds. Conclusions. Minced Skin grafting accelerates reepithelialization of fluid-treated Skin wounds. The technique is faster and less expensive than split-thickness Skin grafting and keratinocyte suspension Transplantation. Minced Skin grafting may have implications for the treatment of chronic wounds. (C) 2002 Elsevier Science (USA).

However, their role in Skin Transplantation is still unknown. In this study, we investigated the trafficking of intra-graft donor T cells to the recipient’s lymphoid organs after Skin Transplantation in mice. To test this C57BL/6 (B6) mice received a Skin transplant from a double transgenic B6 donor whose T cells expressed DSred while FoxP3 expression was associated with GFP (Tregs being double positive). At different time points after Transplantation, cell suspensions of recipients’ lymph nodes, spleen and thymus were prepared. The presence and characterization of fluorescent donor cells was investigated using imaging flow cytometry analysis. First, the presence of T cells (DSred+ FoxP3-) was detected only at d4-d7 post-Transplantation i.e. after Skin graft revascularization and essentially in the host’s thymus. On the other hand, double positive (DSred+ FoxP3+) Tregs were found in all recipient’s lymphoid organs. Surprisingly, high frequencies of single positive GFP+ cells were found in both ipsilateral lymph nodes and thymus. The nature of these cells is being investigated. In summary, our results show that most T cells leaving the graft are Tregs spreading through all recipient’s lymphoid organs while other intra-graft T cells migrate preferentially to the host’s thymus.

The intrinsic apoptosis pathway is differently regulated among conventional and regulatory T cells. Targeted pharmacological modulation of this pathway with a small molecule Bcl-2/Bcl-xL inhibitor (ABT-737) caused a selective depletion of effector T cells and a relative enrichment of regulatory T cells in vivo. Treatment with ABT-737 resulted in a tolerogenic milieu, which was exploited to alleviate graft-versus-host disease, to prevent allograft rejection in a stringent fully MHC-mismatched Skin Transplantation model and to induce immunological tolerance in combination with bone marrow Transplantation. This concept has the potential to find various applications for immunotherapy, since it allows pharmacologic exploitation of the immunomodulatory properties of regulatory T cells without the need for cell manipulation ex vivo.
