However, the mechanism of action of ghrelin is not fully understood. In the present study, we attempted to elucidate the role of histamine in ghrelin-induced acid secretion in rat Stomach. Intravenous administration of ghrelin at 0.8 to 20 μg/kg dose dependently increased gastric acid secretion, as measured by the gastric lumen perfusion method. The maximum response was almost equal to that of gastrin (20 μg/kg). These actions were abolished by bilateral subdiaphragmatic vagotomy. Famotidine (0.33 mg/kg) also completely inhibited the effects of ghrelin. Furthermore, ghrelin increased histidine decarboxylase (HDC) messenger RNA (mRNA) levels, as measured by real-time reverse transcription-polymerase chain reaction using LightCycler. The action of ghrelin on HDC mRNA was abolished by vagotomy. Ghrelin did not affect histamine release from isolated vascularly perfused rat Stomach. Taken together, these results suggest that ghrelin stimulates gastric acid secretion via a mechanism involving activation of vagal efferent nerve and histamine release from gastric enterochromaffin-like cells.

Physiological and Biochemical Zoology, 1995Co-Authors: Susanne Holmgren, Anncathrine JonssonAbstract:Neurotensin-like material was found throughout the gut of the amphibian Necturus maculosus, in both muscle layers and mucosa, and with the highest levels in the intestine. Ion-exchange high-performance liquid chromatography (HPLC) and reversed-phase HPLC revealed several peaks of immunoreactive material, with one closely related to neurotensin 1-6 occurring by both methods. Exogenous neurotensin was excitatory on longitudinal Stomach muscle; this excitation was unaffected by tetrodotoxin, indicating a direct effect on the smooth muscle. An inhibitory effect of neurotensin on circular muscle preparations was abolished by tetrodotoxin, revealing an excitatory effect. Noradrenaline and adrenaline were in many cases inhibitory on the circular preparations, an effect that was blocked by the β-adrenergic anztagonist propranolol. Propranolol also blocked the inhibitory effect of neurotensin in approximately 50% of the tests, which suggests an involvement of an adrenergic link in the inhibitory effect of neuroten...

evaluation of residual Stomach Motility after proximal gastrectomy for gastric cancer by electrogastrography

Based on fast Fourier transformation, recorded slow waves could be analyzed to obtain the following parameters: dominant frequency (DF), percentage normal frequency (% 3 cycles per minute [cpm]), and power ratio (PR). EGG parameters, the length of the greater curvature of the residual Stomach (LGC), were recorded in 18 gastrectomized patients. Compared to 12 healthy controls, the gastrectomized patients had abdominal EGG parameters including lower %3cpm (43 ± 21% vs 83 ± 7%; P 20 cm and controls in PR (2.3± 0.9 vs 2.5± 0.8; n.s.). In conclusion, the Motility of the residual Stomach would be equal to that of the nonresected Stomach as if the volume of the residual Stomach was more than half.
