    Abstract:

    Background and objectivesWithania coagulans Dunal (Solanaceae) fruits are recommended to treat asthma, dyspepsia, biliousness, flatulent colic, liver complaints, intestinal infections, Strangury, wounds and as diuretic, emetic and sedative agent in Indian traditional system of medicine. The objective of this study was to describe the systematic fatty acid composition of the petroleum ether extract of W. coagulans fruits.MethodsPetroleum ether extract of W. coagulans fruits was prepared by maceration. Components of the W. coagulans petroleum ether extract were identified by gas chromatography-mass spectrometry and their concentrations were determined.ResultsThe fixed oil of W. coagulans fruits constituted of twenty nine components including unsaturated (52.36%) and saturated (22.15%) fatty acids, alkenes (5.65%), phytosterols (4.39%), fatty alcohols (4.14%), aromatic acid (3.56%), monoterpenes (3.22%), triterpenoids (1.83%) and alkanes (2.7%). Most of the saturated and unsaturated fatty acids were identified as their methyl esters.ConclusionsPalmitoleic and 11-eicosenoic acids have been reported for the first time in petroleum ether extract of the W. coagulans fruits. The present study has illustrated the chemical nature of W. coagulans fruit and described its fatty acids composition.

    The fruits of Withania coagulans Dunal (family: Solanaceae) are sweet, sedative, emetic, alterative and diuretic; used to treat asthma, biliousness, Strangury, wounds, dyspepsia, flatulent colic, liver complaints and intestinal infections in the indigenous system of medicine. Phytochemical investigation of the methanolic extract of W. coagulans fruits led to the isolation of a new fatty acid, an aromatic ester and a monoterpenic benzyl glucoside characterised as n-octatriacont-17-enoic acid (3), geranilan-10-olyl dihydrocinnamoate (4) and geranilan-8-oic acid-10-olyl salicyloxy-2-O-β-d-glucofuranosyl-(6″→1‴)-O-β-d-glucofuranosyl-6‴-n-octadec-9‴′,11‴′-dienoate (5) along with two known fatty acids, n-dotriacont-21-enoic acid (1) and n-tetratriacontanoic acid (2). The structures of isolated phytoconstituents were established on the basis of 1D and 2D NMR, FT-IR, UV, and MS data and chemical means.

Phytochemical investigation of the fruits yielded a withanolide tetraglucoside identi- fied as (20S, 22R)-5α, 20α-diacetoxy-6β-hydroxy-1-oxowitha-2,24-dienolide-6-β-D-glucopyranosyl- (6i→1ii)-β-D-glucopyranosyl-(6ii→1iii)-β-D-glucopyranosyl-(6iii→1iiii)-β-D-glucopyranoside), a capry- loyl hexaglucoside formulated as n-octanoyl-β-D-glucopyranosyl-(6a→1b)-β-D-glucopyranosyl-(6b→1c)-β- D-glucopyranosyl-(6c→1d)-β-D-glucopyranosyl-(6d→1e)-glucopyranosyl-(6e→1f)-glucopyranoside and a menthyl tetraglucoside characterized as menthol-O-α-L-glucopyranosyl-(2a→1b)-O-α-L-glucopyranosyl- (2b→1c)-O-α-L-glucopyranosyl-(2c→1d)-O-α-L-glucopyranoside along with three fatty acid esters, n-nona- cosanyl linolenate, n-octacosanyl linolenate and n-heptacosanyl linolenate.     Abstract:

    Strychnos potatorum Linn. seeds are used in the Indian traditional system of medicine for the treatment of hepatopathy, nephropathy, gonorrhoea, leucorrhoea, gastropathy, bronchitis, chronic diarrhoea, Strangury, renal and vesicle calculi, diabetes and eye diseases. The present study describes the hepatoprotective and antioxidant activities of the seed powder (SPP) and aqueous extract (SPE) of Strychnos potatorum seeds against CCl4-induced acute hepatic injury. Hepatic injury was achieved by injecting 3 ml/kg, s.c. of CCl4 in equal proportion with olive oil. Both SPP and SPE at the doses 100 and 200 mg/kg, p.o. offered significant (p < 0.001) hepatoprotective action by reducing the serum marker enzymes like serum glutamate oxaloacetate transaminase (SGOT) and serum glutamate pyruvate transaminase (SGPT). They also reduced the elevated levels of ALP and serum bilirubin. Reduced enzymic and nonenzymic antioxidant levels and elevated lipid peroxide levels were restored to normal by administration of SPP and SPE. Histopathological studies further confirmed the hepatoprotective activity of SPP and SPE when compared with the CCl4 treated control groups. The results obtained were compared with Silymarin (50 mg/kg, p.o.), the standard drug. In conclusion, SPE (200 mg/kg, p.o.) showed significant hepatoprotective activity similar to that of the standard drug, Silymarin (50 mg/kg, p.o.).

  • Vaccinium macrocarpon (cranberry) reduces intake of antibiotics in the treatment of non-severe lower urinary tract infections: A drug monitoring study

    Abstract:

    Urinary tract infections (UTIs) are among the most common bacterial infections affecting women. Due to the growing problem of antibiotic resistances there is an urgent need for alternative herbal medicinal products for the treatment of non-severe lower UTIs. Vaccinium macrocarpon Aiton is effective through various mechanisms of action without inducing resistances in microorganisms [1]. Thus, a drug monitoring study (n = 48) was conducted on women older than 18 years with an increase in leukocytes and clinical symptoms typical for UTIs like dysuria, imperative Strangury, increased or new onset of incontinence, hematuria and suprapubic pain. All subjects received one tablet of a standardized cranberry extract (67 mg proanthocyanidines) combined with Nasturtium officinale W.T. Aiton, Armoracia rusticana G. Gaertn., B. Mey & Scherb., and vitamins, twice daily (morning and evening) during the first week and one tablet per day for the following four weeks. After 14 days (t1) and after 35 days (t2) taking the tablets a further survey and control check-up with a urine test have been conducted. Patients with persistent clinical symptoms received an antibiotic, for the others the intake of cranberry tablets was continued. 34 patients (70.8%) could abstain from taking antibiotics within the first 14 days (t1) and 32 patients (66.7%) within 35 days (t2). 28 patients (87.5%) were without symptoms at t2. In conclusion, this drug monitoring study was the first study performed with a combination product (cranberry, nasturtium, armoracia), which furthermore supports the concept of using alternative medicine treating non-severe lower UTIs.
