Anti-inflammatory effects in muscle injury by Transdermal Application of gel with Lychnophora pinaster aerial parts using phonophoresis in rats

Anti-inflammatory activity of Lycnophora pinaster was investigated for extracts, fractions, and isolated compounds of their aerial parts. The hexane extract (HE) provided α -amyrin, lupeol, mixture of α -amyrin and lupeol, mixture of 3- O -acetyl-lupeol and 3- O -acetyl-pseudotaraxasterol, and mixture of the steroids stigmasterol and sitosterol. The aqueous extract (WE) provided a fraction containing alkaloids (AF) and another one containing phenolic compounds (PF). Methods The crude hexane extract obtained from aerial parts of L. pinaster was submitted to chromatographic fractionation. The fractionation of PF was performed by preparative HPLC analysis, providing the flavonoid quercetin. The extracts, fractions, and compounds isolated from L. pinaster were tested to evaluate the anti-inflammatory activity by experimental model of impact injury, followed by Transdermal Application of gels with these samples. The Application of the gels was performed using phonophoresis in rat paws after induction of muscle injury. Histological analysis was based on scores assigned by the capacity of decreasing the lesion. Results HE and WE exhibited anti-inflammatory activity. Some fractions, triterpenes, and steroids also reduced the inflammatory infiltrates caused by muscle injury. Lupeol promoted a significant reduction of inflammation. Quercetin also provided significant results, promoting the greatest decreases in muscle injury. Conclusion The results of this work suggest that topical Application of triterpenes, steroids and flavonoid significantly decreases the inflammatory process generated by muscle injury. The Transdermal Application using phonophoresis in rat paws of gel with lupeol and quercetin attenuates the inflammation.

time related immunomodulation by stressors and corticosterone Transdermal Application in toads

However, time-changes related to immunomodulation associated with glucocorticoid (GC) alterations following stressors and GC treatment are complex. The present study describes time-related changes in immune response and corticosterone (CORT) plasma levels following restraint challenge, short, mid and long-term captivity, and CORT exogenous administration by Transdermal Application (TA) in Rhinella ornata toads. We observed increased neutrophil: lymphocyte ratios after restraint challenge and CORT TA, without changes following short and mid-term captivity. Plasma bacterial killing ability was sustained in all treatments, except long-term captivity, with decreased values after 90 days under such conditions. Phagocytic activity of peritoneal cells increased after mid-term captivity, and the phytohemagglutinin swelling response was impaired in those animals treated with CORT TA for 20 consecutive days. Plasma CORT levels increased or were sustained after restraint challenge (depending on initial values), decreased following mid and long-term captivity (for those animals showing high CORT in the field) and increased after 20 days of CORT TA. By performing assessments of time-changes in immune processes and CORT plasma levels in R. ornata, we demonstrate immuno-enhancing effects following restraint, short and mid-term stressors, while long-term stressors and CORT TA promoted immunosuppression in these toads.

In anurans, testosterone and corticosterone are correlated to sexual behavior, reproduction, and immune function. Male toads (Rhinella jimi) were treated with acute doses of testosterone (T) and corticosterone (CORT) and immune variables (plasma bacterial killing ability [BKA], swelling after phytohemagglutinin [PHA] injection and the time point of maximum PHA swelling response) were measured. Transdermal T and CORT Application increased androgen (T-DHT) and CORT plasma levels after 1 h of treatment, respectively, without a dose-specific effect. Transdermal T treatment did not affect BKA or PHA swelling response. Individuals treated with Transdermal CORT showed an earlier maximum PHA swelling response and a tendency of lower BKA 15 h after treatment. Our results indicated that an acute experimental increase of CORT plasma levels diminished time for inflammatory resolution and suppressed non-cellular innate response.

Chronically elevated glucocorticoid levels are mostly associated with immunosuppression. However, there is still a paucity of knowledge regarding the amphibian physiological reaction to short- and long-term stress conditions, including glucocorticoid effects on the immune response. In order to better clarify the relationship between glucocorticoids and immune response, newly captured Brazilian toads (Rhinella icterica) (1 week in captivity) were subjected to a daily Transdermal Application (TA), of corticosterone or vehicle, for 30 consecutive days. Measures were made on the first day (acute stressor) and last day (chronic stressor), at timepoints 1, 6, and 12 hr post TA. A number of variables were analyzed: corticosterone plasma levels (CORT); neutrophil/lymphocyte ratio (N:L); testosterone plasma levels (T); innate immune response, as indicated by bacterial killing ability (BKA); and whole blood phagocytosis. The corticosterone TA only simulated hormonal changes associated with the acute stress response, even after 30 consecutive days of the treatment, with the increased CORT in response after corticosterone TA being evident only 1 hr postApplication and not thereafter. The general responses to corticosterone TA included increased CORT and N:L at first day of the treatment, and increased CORT and phagocytosis on the last day of the treatment. A decrease in T and BKA associated with the time in captivity was also evident, suggesting that captivity may be a chronic stressor for these toads.

Dermal scatter reduction in human skin: a method using controlled Application of glycerol.

McnicholsAbstract:Background and Objective Previous studies in a hairless Guinea pig model showed that Transdermal Application of glycerol effected a temporary reduction in dermal scatter of light. This study focuses on the Application of this protocol on human patients. Study Design/Materials and Methods After stratum corneal removal, glycerol was applied to human subjects using a low pressure Transdermal Application device. Optical coherence tomography imaging showed increased intensity of radiation reaching deeper regions in the skin and photographs showed enhanced visualization of dermal structures. Results/Conclusion Topically applied glycerol increased light penetration of in vivo corneal-stripped skin. This minimally invasive approach to temporary dermal scatter reduction has the potential to improve the efficacy of light-based diagnostic or therapeutic devices. Lasers Surg. Temporary dermal scatter reduction: Quantitative assessment and implications for improved laser tattoo removal

Bell, Massoud MotamediAbstract:Background and Objectives Temporary dermal clearing, i.e., reduction in the attenuation coefficient of the dermis and epidermis, may lead to improved laser tattoo removal by providing increased efficiency of laser delivery to embedded ink particles and enabling the use of shorter wavelength visible lasers more effective on certain inks. Study Designs/Materials and Methods In a hairless guinea pig model of human tattoo, we tested both intradermal and Transdermal Application of glycerol, using visual inspection, spectral analysis, and optical coherence tomography techniques to assess effectiveness. In controlled experiments, we compared the outcomes of single laser treatment sessions for both cleared and uncleared tattoo sites using Q-switched 755 and 532 nm lasers on three different inks. Results Intradermal injection of clearing agents induced dermal clearing but resulted in necrosis and scar. Transdermal Application of clearing agents resulted in moderate reversible clearing, which was localized to the superficial layers of the skin and did not result in complications. Statistically significant differences in laser treatment outcome were observed relative to a number of treatment parameters including the treatment of certain tattoos by short wavelength lasers. Conclusions Temporary clearing of superficial skin layers may be performed in an apparently safe and reliable manner. Clearing should lead to increased penetration of laser light to tattoos and should, therefore, increase treatment efficiency.
