Development and large scale application of cryopreservation techniques for shoot and somatic embryo cultures of Tropical Crops

Escobar, Florent EngelmannAbstract:Shoot-tips and somatic embryos are the explants of choice for the in vitro long-term storage of ex situ plant genetic resources in liquid nitrogen. Cryopreservation of organized structures has significantly progressed, especially for species of Tropical origin, with the development of several vitrification-based procedures such as encapsulation-dehydration, vitrification and droplet-vitrification approaches. They have allowed improvements in survival and recovery after cryopreservation compared with conventional crystallization-based protocols, proving their effectiveness for large scale application with embryos and shoot-tips of different plants. This review addresses the main physical and technological aspects involved in plant cryopreservation methods, illustrating the development of research with three cases: citrus, cassava and potato. These studies demonstrate how cryopreservation strategies are increasingly applied for their successful employment in the genebanks.

Finally, the collection and conservation situation of Tropical Crops in China was predicted.

Protection of Tropical Crop Germplasm Resources in China
Chinese Journal of Tropical Crops, 2011Co-Authors: Tian XinminAbstract:The protection of Chinese Tropical crop germplasm resources,which plays an important role in biodiversity and constitutes the material basis of the sustainable development of domestic Tropical Crops,is essential to the development of Tropical crop industry.Since 1978,several research studies and collections have been carried out,and 47 000 copies in the form of germplasm nursery,in vitro library,and seed bank have been saved.The identification and evaluation of Tropical crop germplasm focused on quality traits,stress tolerance,pest resistance and cytological traits,etc,which has established a standardized database that can be publicly shared through the Tropical crop germplasm resources information network.But there are still problems such as unclear resource detail,unsubstantial foundation of resource research,lack of resource innovation,inadequate storage facilities,lack of storage techniques,faint consciousness of sharing and lack of mechanisms for sharing,etc..It is suggest to carry out a comprehensive survey and collection of domestic resources while introducing and studying foreign resources.The identification and evaluation need to be strengthened.Furthermore,to promote the sharing of resources and cultivate new varieties with independent intellectual properties,creation of germplasm resources needs to be increased and the sharing mechanism has to be established.
