Guangzhou Chemical Industry, 2012Co-Authors: Pan Hai-yaAbstract:2-(4-carbosulfonylphenyl)-1-(6-methyl-3-pyridinyl)ethanone,a key intermediate of Etoricoxib,was synthesized from thioanisole via Friedel-Crafts acylation,Oxidation,Willgerodt-Kindler Rearrangement,Claisen condensation with an overall yield of 39% and HPLC purity of 92.3% with further purification.4-(Methanesulfonyl)phenylacetic acid and 2-(4-Mesylphenyl)-1-(6-methylpyridin-3-yl)-ethan-1-one as key products were characterized with IR and NMR.

Fine chemicals, 2012Co-Authors: Pan XianhuaAbstract:8-Chlorodibenzothiepin-10(11H)-one,a key synthetic intermediate of Zotepine,was synthesized by metal-catalyzed coupling reaction,Willgerodt-Kindler Rearrangement,hydrolysis and intermolecular cyclization reaction.The coupling reaction was catalyzed by CuI/L-proline,so that the reaction temperature could be cooled down to 100 ℃ with a higher yield of 85%.Willgerodt-Kindler Rearrangement and hydrolysis was carried out in a one-pot-reaction.The mole yield of all four steps was 40%.The product structure was identified by means of IR,MS,and 1HNMR.
