This strain, whose genome is described here, was isolated from the fecal flora of an asymptomatic patient. A. senegalensis is an anaerobic Gram-negative rod-shaped bacterium. Here we describe the features of this organism, together with the complete genome sequence and annotation. The 4,017,609 bp long genome (1 chromosome, but no plasmid) contains 3,113 protein-coding and 50 RNA genes, including 5 rRNA genes.

    Abstract Here we describe the basic characteristics of Corynebacterium pacaense strain Marseille-P2417T (= CSUR P2417), Alistipes megaguti strain Marseille-P5997T (= CSUR P5997) and Alistipes provencensis strain Marseille-P2431T (= CSUR P2431 = DSM 102308). The phenotypic criteria, the 16S ribosomal RNA sequencing and MALDI-TOF MS spectra analysis were used to identify and characterize these new bacteria species, which were isolated from fresh human stool specimens.

    BACKGROUND: Management of blood cholesterol is a major focus of efforts to prevent cardiovascular diseases. The objective of this study was to investigate how the gut microbiota affects host cholesterol homeostasis at the organism scale. RESULTS: We depleted the intestinal microbiota of hypercholesterolemic female Apoe-/- mice using broad-spectrum antibiotics. Measurement of plasma cholesterol levels as well as cholesterol synthesis and fluxes by complementary approaches showed that the intestinal microbiota strongly regulates plasma cholesterol level, hepatic cholesterol synthesis, and enterohepatic circulation. Moreover, transplant of the microbiota from humans harboring elevated plasma cholesterol levels to recipient mice induced a phenotype of high plasma cholesterol levels in association with a low hepatic cholesterol synthesis and high intestinal absorption pattern. Recipient mice phenotypes correlated with several specific bacterial phylotypes affiliated to Betaproteobacteria, Alistipes, Bacteroides, and Barnesiella taxa. CONCLUSIONS: These results indicate that the intestinal microbiota determines the circulating cholesterol level and may thus represent a novel therapeutic target in the management of dyslipidemia and cardiovascular diseases.

    Abstract:

    Lessonia nigrescens inhabits rocky intertidal and shallow subtidal zone, being a crucial brown algae to ecology and economy. L. nigrescens contains a broad range of bioactive compounds, such as fucans and phlorotannins. In this study, the antidiabetic activity of Lessonia nigrescens ethanolic extract was investigated in streptozotocin (SZT)-induced type 2 diabetic mice fed with a high-sucrose/high-fat diet. The components of the intestinal microflora in diabetic mice were analyzed by high-throughput next-generation 16S rRNA gene sequencing. Fasting blood glucose levels in type 2 diabetic mice were significantly decreased after administration of the extract. The histology revealed that the extract could protect the cellular architecture of liver and kidney. The extract significantly increased Bacteroidetes bacteria and decreased Firmicutes populations in intestinal microflora. Specifically, the extract can increase Barnesiella and reduce Clostridium and Alistipes. The increased gene and protein expression levels of PI3K in liver were observed in the extract groups, while the expressions of JNK1 were significantly downregulated by the extract.
