    Abstract:

    The purpose was to determine the effects of acute toxicity of glucomannan standardized Porang powder (Amorphophallus onchophyllus) and konjac powder (Amorphophallus konjac) as well as the lethal dose for 50% of the white rats (Rattus novergicus) population tested. Porang powder (Amorphophallus onchophyllus) and konjac powder (Amorphophallus konjac) both contain glucomannan fiber which has the effect of lowering cholesterol and blood sugar, reducing weight, and improving digestive and immune health. However, in porang powder and konjac powder there are calcium oxalate crystals which may form kidney stones and cause itching as well as irritation, therefore a toxicity study is necessary. The method used in this research was randomized post control group method. The experimental animals were male white rats (Rattus novergicus). Nine treatment groups, each consisting of six experimental animals, were administered different doses (175; 542,5; 1681,8; and 5213,4 mg / kg body weight) of each porang and konjac powder. The observed parameters were LD50 value (lethal dose) or mortalty in experimental animals. The LD50 values were determined using three methods, namely Thomson and Weil method, the linear equation method, and the Graph Miller and Tainter method. The results of acute toxicity tests for 24 and 48 hours did not cause mortality in white rats, showed no toxicity signs, with each LD50 value greater than 5213,4 mg / kg body weight

    Abstract:

    Toxicity tests were carried out to ensure the safety of porang powder (Amorphophallus oncophyllus) and konjak powder (Amorphophallus konjac) which standardized glucomannan. Glucomannan is a water-solube polysaccharide consisting of D-glucose and D-mannose that linked by β-1,4 glycosidic bonds. This study aimed to investigate toxicity of porang and konjak glucomannan using experimental animals white rats. The method used is a randomized post control group. The experimental animals used in white rats were 35 and were grouped into seven different groups. The glucomannan dosage used is 500 mg / kg BW ; 1800 mg / kg BW; and 6.480 mg / kg BW. In this toxicity test, observations were made on the number of death of white rats and toxicity symptoms that might appear after 24 hours and 48 hours after preparation. Then the LD50 value is calculated using the Miller and Tainter graph method as well as the Reed and Muench arithmetic methods. From the observations made, the calculation of the LD50 value for each group cannot be calculated. This is because the distribution of porang powder and konjak powder in white rats does not cause death and no toxic symptoms appear. So, can be said that porang powder and konjac powder have an LD50 value greater than 6.480 mg / kg BB. Based on the classification table of the test preparation with a dosage range of 5000-1500 g / kg BB if there is no death in experimental animals, the test preparation is declared practically non-toxic

The glucomannan levelsinporang and konjac powder were 52,33 ± 0,74% (w/w)and 61,24 ± 0,61% (w/w), respectively. Glucomannan contained in porangtuber functions as water soluble hemicellulose fiber whichhas low calories. The experimental animals used in this study were white rats (Rattus norvegicus) induced with alloxan monohydrate. Blood samples were taken from the tail end of male white rat (Rattus norvegicus) aged 2-2,5 months with the weight of 100-250 grams. The treatment group was divided into 8 groups, which included negative control group (CMC Na 0.3%), positive control group (glibenclamide 0,455 mg / kg white rat), porangpowderdosage group I; II; III (with the dosage of 59,5 mg / kg white rat; 115,5 mg / kg white rat; 175 mg / g white rat BB calculated as glucomanan, respectively), and konjac powder dosage group I; II; III (with the dosage of 59,5 mg / kg white rat, dose 115,5 mg / kg white rat, dose 175 mg / kg white rat calculated as glucomanan, respectively). The treatment was done every day for 21 days with a measurement of blood sugar levels performed every 3 days and daily body weight performed every day.     Abstract:

    Porang (Amorphophallus oncophyllus) and konjak (Amorphophallus konjac) is a type of tuber plant that contains glukomanan Polysaccharide, which glukomanan can be used to lower blood sugar levels. In this research is to know the effect of decreasing blood sugar level given by porang powder and konjak powder standard glucomannan. Glucomannan concentration in porang powder was 52,33 ± 0,74 % (w/w) and glucomannan content on konjak powder was 61,24 ± 0,61 % (w/w). The sample of this study wese eight group of mice as treatment groups with six replications on 2-2,5 months of male mice strain BALB/C which had been induced by alloxan monohydrate (200 mg/ kg body weight of mice). The treatment group consist of negative control (CMC sodium 0,3%), positive control (glibenclamide 0,013 mg / 20g body weight of mice), and treatment group I, II, III, IV, V, VI with the administration of porang powder standard glukomannan dose of 85; 165; 250 mg/ kg body weight of mice and konjak powder standard glukomannan dose of 85; 165; 250 mg/ kg Body weight of mice. Treatment is given daily for 21 days with measurement of blood glucose level every three days and weight every day. The result was analyzed by One Way ANOVA with significant value of 5%. The result indicated that konjak powder standard glucomannan dose of 250 mg / kg body weight of mice can decrease blood sugar level within 13 days
