Recent advances in molecular biotechnology (e.g., next generation genomics) have increased the range of collection material employable for DNA-based analyses to unprecedented levels. Nevertheless, the value of museum specimens strictly depends on reliability of data associated with them. We report on investigations of ancient DNA from specimens of smooth-coated otter ( Lutrogale perspicillata , Mustelidae), the largest otter species living in Asia, in US and European mammal collections. Mitochondrial DNA Cytochrome-b gene sequencing proved that the studied specimens were not the expected taxon. Indeed, they actually belonged to three different species, namely the Asian small-clawed ( Aonyx cinereus ), Eurasian ( Lutra lutra ) and African clawless ( Aonyx capensis ) otters. This represents the first record of mustelid misidentification from museum collections. Detection of errors can be extremely difficult when based only on collectors’ notes and data. Hence, we warn scientists involved in otter research about potential challenges when dealing with museum specimens. We recommend curators pursue a multidisciplinary approach, including DNA analyses, to accurately catalogue the resources under their management and uphold the value of biodiversity information.

Fifty-eight modern and 11 archival (dated 1882–1970) otters sampled from Iraq to Malaysian Borneo were genotyped (mtDNA Cytochrome-b, 10 microsatellite DNA loci). Moreover, 16 Aonyx cinereus (Asian small-clawed otter) and seven Lutra lutra (Eurasian otter) were sequenced to increase information available for phylogenetic reconstructions. As reported in previous studies, we found that L. perspicillata, A. cinereus and A. capensis (African clawless otter) grouped in a clade sister to the genus Lutra, with L. perspicillata and A. cinereus being reciprocally monophyletic. Within L. perspicillata, we uncovered three Evolutionarily Significant Units and proved that L. p. maxwelli is not only endemic to Iraq but also the most recent subspecies. We suggest a revision of the distribution range limits of easternmost L. perspicillata subspecies. We show that smooth-coated otters in Singapore are L. perspicillata x A. cinereus hybrids with A. cinereus mtDNA, the first reported case of hybridization in the wild among otters. This result also provides evidence supporting the inclusion of L. perspicillata and A. cinereus in the genus Amblonyx, thus avoiding the paraphyly of the genus Aonyx.

Aonyx capensis is more widespread than Lutra maculicollis, which is adapted to feed on fish, and hence is limited to areas which can support them, whereas clawless otters are adapted to eat the more widespread crabs and frogs. Otters are protected in reserves and national parks. Hunting outside of these areas is not a serious threat, but habitat degradation and loss due to rapid expansion of the human population. Two otter species occur in South Africa: the Cape clawless otter Aonyx capensis and the spotted-necked otter Lutra maculicollls. L'uso degli ambienti ripariali è simile per entrambe le specie che selezionano in particolare tratti di rive caratterizzati dalla presenza di rocce emergenti sia coperte sia prive di vegetazione.
