The aim of this study was (i) to screen Arctium lappa and Tussilago farfara extracts for activity against Mycobacterium tuberculosis and (ii) to isolate and identify the compound(s) responsible for this reputed anti-TB effect. Materials and methods The activity of extracts and isolated compounds was determined against Mycobacterium tuberculosis H37Rv using a high throughput spot culture growth inhibition (HT-SPOTi) assay. Results The n-hexane extracts of both plants, the ethyl acetate extract of Tussilago farfara and the dichloromethane phase derived from the methanol extract of Arctium lappa displayed antitubercular activity (MIC 62.5 μg/mL). Further chemical investigation of Arctium lappa led to the isolation of n-nonacosane (1), taraxasterol acetate (2), taraxasterol (3), a (1:1) mixture of β sitosterol/stigmasterol (4), isololiolide (5), melitensin (6), trans-caffeic acid (7), kaempferol (8), quercetin (9), kaempferol-3-O-glucoside (10). Compounds isolated from Tussilago farfara were identified as a (1:1) mixture of β sitosterol/stigmasterol (4), trans-caffeic acid (7), kaempferol (8), quercetin (9), kaempferol-3-O-glucoside (10), loliolide (11), a (4:1) mixture of p-coumaric acid/4-hydroxybenzoic acid (12), p-coumaric acid (13). All compounds were identified following analyses of their physicochemical and spectroscopic data (MS, 1H and 13C-NMR) and by comparison with published data. This is the first report of the isolation of n-nonacosane (1), isololiolide (5), melitensin (6) and kaempferol-3-O-glucoside (10) from Arctium lappa, and of loliolide (11) from Tussilago farfara. Amongst the isolated compounds, the best activity was observed for p-coumaric acid (13) (MIC 31.3 μg/mL or 190.9 μM) alone and in mixture with 4-hydroxybenzoic acid (12) (MIC 62.5 μg/mL). Conclusions The above results provide for the first time some scientific evidence to support, to some extent, the ethno-medicinal use of Arctium lappa and Tussilago farfara as traditional antitubercular remedies.

Effect of aqueous extract of Arctium lappa L. (burdock) roots on the sexual behavior of male rats

It is used to treat impotence and sterility in China, and Native Americans included the root in herbal preparations for women in labor. However, its use has not been scientifically validated. The present study therefore investigated the effects of aqueous extract of Arctium lappa L. roots on sexual behavior in normal male rats. Seventy-five albino male rats were randomly divided into five groups of 15 rats each. Rats in group 1 (control) were administered 10 mL⁄kg body weight distilled water (vehicle), group 2 received 60 mg/kg body weight sildenafil citrate (Viagra), while those in groups 3, 4, and 5 were given 300, 600, and 1,200 mg/kg body weight, respectively, of aqueous extract of Arctium lappa L. roots in the same volume. Female albino rats were made receptive by hormonal treatment. Sexual behavior parameters in male rats were monitored on days 3, 7 and 15 by pairing with receptive females (1:3). Male serum testosterone concentrations and potency were also determined. Oral administration of Arctium lappa L. roots extract at 600 and 1,200 mg/kg body weight significantly increased the frequencies of mount, intromission, and ejaculation frequency (p < 0.05). The latencies of mount and intromission were significantly reduced and ejaculation latency was prolonged. Administration of the extract also reduced the post-ejaculatory interval. The standard drug (Viagra) was more effective than the extract. The extract significantly increased the frequencies of all components of penile reflexes as well as serum testosterone levels, compared with the distilled water controls. The results of this study demonstrate that aqueous extract of Arctium lappa L. roots enhances sexual behavior in male rats. The aphrodisiac effects of the plant extract may be related to the presence of flavonoids, saponins, lignans and alkaloids, acting via a multitude of central and peripheral mechanisms. These results thus support the traditional use of Arctium lappa L. root extract for treating impotence and sterility.

GC-MS analysis of volatile oil from slices of Arctium lappa L.by supercritical CO_2 extraction
2012Co-Authors: Chen Kao-shanAbstract:Objective: To verify the feasibility of obtaining the volatile oil from slices of Arctium lappa L.by supercritical carbon dioxide(CO2) extraction and determine the chemical components of the extract.Methods: The volatile oil was obtained by supercritical CO2 and the chemical components were isolated and identified by gas chromatography-mass spectrometry(GC-MS).Results: The extraction rate was 0.84 %,and 31 compounds were identified from 41 extraction peaks.Conclusion: The technology is competent for extracting the volatile oil from slices of Arctium lappa L.by supercritical CO2,and the oil was identified as a mixture of multiple chemical components.

Chemical Constituents of the Root of Arctium lappa L.

Optimal extracting processes of chlorogenic acid from Arctium lappa
The Chinese Pharmaceutical Journal, 2003Co-Authors: Chen Kao-shanAbstract:OBJECTIVE To optimize the extracting processes of chlorogenic acid from Arctium lappa L.leaves.METHODS The extraction of chlorogenic acid in Arctium lappa L.leaves was investigated with the changes of temperature,pH of extracting solutions,extracting time and frequency and Arctium lappa L.leaves-treated methods.The contents of chlorogenic acid were measured by means of RP-HPLC.RESULTS Either methanol (pH 2~3) at 65℃ or ethanol (pH 2~3) at 80℃ was beneficial for extracting chlorogenic acid.The better extracting time was 4 h,which included 2 h for each time or 2 h for the 1 st time and 1 h for the 2 nd and 3 rd time respectively.CONCLUSION The better extracting processes should be as following:taking Arctium lappa L.leaves dried at normal temperature as the raw materials,extracting at 65℃ with methanol or 80℃ with ethanol,pH=2~3,for 4 h which included 2 h for each time or 2 h for the 1st time and 1 h for the 2nd and 3rd time respectively.
