    Abstract:

    Mast cell activation releases the mediators associated with type I allergy. As such, the study of mast cell activation is critical for understanding the allergic reaction, and for developing methods to control it. Importantly, another ligand receptor pair (compound 48/80 and MRGPRX2) that activates mast cells in addition to allergen-IgE-FceRI has been identified. As mast cells mature in tissue from hematopoietic stem cells, their physiology and pathophysiology is difficult to study. Mast cell lines and mast cells cultured from stem cells are often studied instead of tissue mast cells. There has been some progress in the description of the mechanism of the activation of mast cells, substances limiting mast cell activation and in the catalogue of proteases that mast cells express. Basophil Granulocytes express FceRI, bind IgE and respond to allergen crosslinking in a very similar fashion to mast cells. In the recent literature, Basophils were mistakenly described as antigen-presenting cells; this has convincingly been disputed in a number of subsequent publications. Their function in physiology and pathophysiology is not known, but they are frequently used to document allergic sensitisation in the Basophil activation test. Significant progress has been made in documenting the relevance of Basophil activation as a second-line test in allergy diagnosis. Basophil reactivity and sensitivity may reflect symptom severity and allergen threshold, and are used to document and monitor allergy. The physiology and pathophysiology of allergic effector cells remain an important area of research.

    Abstract Despite being first described in humans nearly 130 years ago, the Basophil Granulocyte has received little recognition other than being the least common leukocyte circulating in blood. Even after its identity as the source of histamine released by blood cells in response to reaginic IgE, its role in allergic disease has largely been viewed as redundant to that of the tissue mast cell. This line of thought, however, is changing with evidence that has emerged during the last 15 years. Not only have these rare cells been shown to constitute a significant source of cytokines (IL‐4 and IL‐13) vital to the pathogenesis of allergic disease, but by doing so, may very well modulate T‐helper 2‐type inflammation at the level of T‐cell/dendritic cell interactions. This novel concept combined with the fact that Basophils selectively infiltrate allergic lesion sites has sparked greater interest in this once overlooked immune cell, both in adaptive as well as in innate immunity.

    Bakalářská práce se zaměřuje na využití testu aktivace bazofilních granulocytů v rámci specifické alergenové imunoterapie. Teoretická část shrnuje charakteristiku bazofilů, alergické reakce, diagnostiku a terapii alergických onemocnění. V praktické části byl využit test aktivace bazofilů za pomoci průtokové cytometrie ke zhodnocení senzitivity pacientů v průběhu specifické imunoterapie. Srovnáním výsledků ve formě procenta aktivovaných bazofilů, CD-sens a plochy pod titrační křivkou s hodnotami alergen specifických protilátek IgE byla potvrzena validita testu aktivace bazofilů s použitím rekombinantních alergenů jako metody vhodné k posouzení citlivosti pacientů k alergenu.ObhájenoThis bachelor thesis is focused on employing Basophil activation test in the context of specific allergen immunotherapy. Theoretical part summarizes Basophil characteristics, allergic reactions, diagnostics and therapy of allergic diseases and disorders. In practical part, flow cytometric Basophil activation test was used to evaluate sensitivity of patients in the course of specific immunotherapy. By comparing the results in the percentage of activated Basophils, CD-sens and areas under the titration curves with IgE-specific allergen values, the validity of the Basophil activation test using recombinant allergens was confirmed as a method suitable for assessing patients' sensitivity to allergen

    Abstract:

    Food allergy is one of the most important food safety problems that has attracted increasing attention. The food allergy experimental models provide not only the accurate allergen detection and evaluation methods but also the powerful approaches for mechanism investigations. In this paper, we reviewed the common food allergy cell models including mast cell, Basophil Granulocyte and Basophil, as well as the animal models of BALB/c mouse, C3H/HeJ mouse, and BN rat. We also introduced zebrafish, a promising model organism for investigating immunity though lacking direct applications in food allergy to date, and focused on traumatic inflammation, bacterial infection and viral infection models. In addition, we also summarized the clinical diagnostic research methods for food allergy. The elucidation of these topics will help researchers to understand the characteristics and mechanisms of various models and thus select the proper models for particular study, so as to support further investigations of food allergy.
