Age-related protein transformations have heterochronous character. The slowest ontogenetic development lasting 4-6 years is peculiar to one of antigens of Beta-Globulins. Analysis of variability of Beta-Globulins in the age diapason beginning from the 4-month-old fry and including adult spawned individuals indicates different functional significance of their individual components. The Beta A-globulin absent at early postembryonic stages becomes the second, after albumin, quantitatively predominant antigen in the adult fish blood. The amount of Beta B-globulin, like antigens of alpha 1- and alpha 2-Globulins preserving relative ontogenetic stability, can decrease significantly in freshwater sturgeons due to their preparation for spawning.

The deepest changes occurred in individual components of albumins and Beta-Globulins (transferrins) and were mostly related to an increased proportion of the protein accounting for these antigens. Transformation of the main component of albumins A1 into adult antigens was completed in 5-month old fry. The main component of Beta-Globulins A (component of transferrins) appeared in the blood flow much later than other proteins and could retain the fry features until the age of 3-4 years. Other antigens belonging to alpha1- and alpha2-Globulins and the second component of transferrins were more stable and did not undergo substantial changes. The direction of ontogenetic variability of serum antigens in sturgeon fry did not depend on the habitat of adult fish in fresh or sea water.

Most widely used, depending on media, are SPEP on cellulose acetate, SPEP on agarose and capillary electrophoresis. The aim of our study was to evaluate the transferability of reference intervals (RI) for SPEP determined on cellulose acetate to agarose for the urban population of Zagreb, Croatia. METHODS: The RI for SPEP on cellulose acetate were determined on the 2246 randomly selected urban adults from population of Zagreb, Croatia in the period from 1998 to 2000 using automated system Olympus Fractoscan Junior. The transferability studies were performed for the separation of serum proteins on agarose using semiautomated system Sebia Hydrasys 2 Scan on the reference sample group of 120 healthy adults with standard laboratory tests within RI determined for the Croatian population. RESULTS: RI for adult population for SPEP on cellulose acetate in accordance to agarose were for albumin 57, 0-69, 0% vs. 59, 2–70, 5% ; alpha-1-Globulins 2, 0–4, 0% vs. 1, 9–3, 2% ; alpha-2-Globulins 5, 0–9-5% vs. 7, 9– 12, 6% ; Beta-Globulins 7, 5–12, 3% vs. 7, 5–12, 0% and gamma-Globulins 12, 7– 22, 1% vs. 9– 16, 3%, respectively. CONCLUSIONS: Our results showed that RI obtained for SPEP on cellulose acetate in accordance to agarose are slightly higher for albumin using agarose, do not differ for alpha-1-Globulins and Beta-Globulins, but there is a great difference for alpha-2-Globulins and gamma-Globulins.The reason for this could be in fact that in an electric field each protein migrates with a speed which is the result not only of his net charge at given pH buffer, size and shape, but also evaporation current, electroendosmosis and porosity of the media, because of which the RI for SPEP have to be produced for each media separately. The difference in the results of SPEP depending on the media is also apparent in the RI for capillary electrophoresis, which, according to literature, are more like those on cellulose acetate than those on agarose. Therefore, it is important that report of SPEP is standardized, containing data of media used and corresponding RI. It is expected that this would reduce variation between laboratories and the risk of misinterpretation of results.

Zhabilov serum fractions in the zone of alpha 1, alpha 2 and Beta-Globulins from healthy people form a characteristic precipitation curve in terms of structure and density with the TNP (thymus nuclear protein) and CNP (carcinoma nuclear protein) as antigens when tested in two-dimensional agarose-gel electrophoresis. The observed immune phenomenon has not so far, to the best of our knowledge, been reported in the scientific literature and has profound implications for fundamental and applied immunology. The object of the present study was to confirm Zhabilov's data and study the ontogenetic aspect of the problem. We used 45 sera of children from different age groups, of healthy adult controls, HIV-infected and cancer patients. The sera were tested against the following exoantigens: TNP (thymus nuclear protein), CNP (carcinoma nuclear protein-Viral Genetics Inc, LA), as well as against ST (staphylococcal toxin), SL (streptolysin) and LPS (lipopolysaccharide) from Gram-negative bacteria (National Center for Parasitic and Infectious Diseases, Sofia, Bulgaria). Two-dimensional electrophoresis was used modified after the protocol of UCLA chemical labs. The results of our study show that immediately following birth the precipitation curve is barely discernible reaching the normal shape following 1 month-5 years of age. The precipitation curve from HIV-infected sera and those of cancer patients is similar to that of newborns. The results from our unpublished observations with sera from other biologic species tested against the same antigens show similar results: no curve was observed in fish whereas the reaction was positive with frogs, birds, rabbits and other mammals. This confirmation of the basic biologic law of ontogenesis being a repetition of phylogenesis gives us reason to consider those fractions as participating in the immune maturation of organisms for acquired immune response. There exists a possibility that they might be part of the already known immunomediators or a stage in the transition from non-specific to specific immunoGlobulins which have not lost their importance in immunogenesis. The lack of previous studies in this respect and the observation and confirmation of this reaction in two different labs underscore the importance of this new immune phenomenon.
