Ingénierie écologique pour la biorémédiation des systèmes aquatiques: effets du couplage de la bioturbation avec la phytoremediation sur le cadmium et l'atrazine.

The accuracy of phytoremediation processes has been primarily demonstrated by individual applications on soils or water sediments. The present Ph.D. aims to demonstrate the interest of additional bioturbation combined to phytoremediation processes for the improvement bioremediation efficiency of aquatic sediments. This strategy benefits are tested experimentally in controlled laboratory conditions with a serie of microcosms reproducing each a portion of water/sediment interface such as in wetland areas. In our experiments, bioturbation was carried out by a conveyor-belt invertebrate population, the tubificidae oligochaetes Tubifex tubifex, well known as an active ecological engineer. The phytoremediation was conducted by the riparian plant Typha latifolia known for its ability to remove organic and inorganic pollutants from sediments by accumulation into its biomass. The experiments were managed to demonstrate the effects of this Biological Influence (plant and inveterbrate) on the mass balances and fluxes of one metal and one pesticide as models of pollutants. Cadmium as a heavy, inorganic and conservative metal pollutant was introduced as a pulse input in the overlying water of the contaminated microcosms, with a cadmium concentration of 20 µg.L-1 in at the initial time of the experiment that lasted one month. In a second experiment, atrazine was mixed in the whole sediment column at the initial time in order to reach a concentration of 5 µg.g-1 of fresh sediment as a source of organic micropollutant and herbicide in the microcosms. The pesticide was radiolabeled with 14C. Fluxes from water to sediment, and from sediment to plants were assessed in experimental conditions with several treatments (+/- plants, +/- bioturbation, +/- pollutants) allowing to demonstrate the effects of the Biological Influence.Our results indicated that the tubificids and the related bioremediation Influences are still efficient under cadmium and atrazine contaminations in aquatic systems. Biotransport due to tubificids changed the distribution of cadmium across the sediment column and enhanced the pumping of cadmium from the water to surface sediment and then to the anoxic underlying sediment surrounding the plant roots. Thereby this biotransport increased the bioaccumulation of cadmium in the root system of Typha latifolia as demonstrated by the roots enrichment coefficients (ECR). Combining Cd contamination and bioturbation optimized the transport of the metal (flux) from sediment to plants with fluxes estimated to 0.02+/-0.00 and 0.07+/-0.03 µg Cd per day without and with bioturbation respectively. In the case of atrazine contamination, bioturbation Influence on the chemical properties of sediment (pH, porosity, Organic matter) is explaining the adsorption-desorption behavior of atrazine in sediment, resulting in the acceleration of atrazine mobility and bioavailability of the pesticide. The transfert of pollutant that passed from attached forms onto sediment particles into a free fraction in pore water in the sediment column was enhanced under bioturbation. Consequently higher values of atrazine were measured in the roots system with ECs of 7.16 ± 0.66 with bioturbation compared to 5.99 ± 0.64 in the treatment without bioturbation. The metabolization of the pesticides was also significantly increased under the effects of bioturbation on the sediment microbiome as demonstrated by enhanced number and quantity of metabolites in Typha latifolia roots in this treatments.Our study, therefore, highlights the potential of bioturbation addition of the phytoextraction for integrated bioremediation strategies of metallic and organic polluted sediments of aquatic ecosystems. Further researches need to take into account the bioturbation Influence on the microorganism communities in the relationship with organic compounds degradation.Ce sujet de recherche vise mieux comprendre les processus de bio remédiation qui participent à la réduction des charges en polluants dans les écosystèmes aquatiques du type zones humides, en tant que question d’actualité en ingénierie écologique. L’efficacité des processus de phytoremédiation a été largement démontrée par des applications individuelles sur le sol et les sédiments. Cette thèse a pour objectif de démontrer la participation d’une population d’invertébrés dans l’efficacité de la réduction de polluants des sédiments aquatiques en combinant le processus de bioturbation avec la phytoremédiation. Les hypotheses de recherche ont été testée expérimentalement en conditions de laboratoire à l’aide d'une série de microcosmes reproduisant chacun une portion d’interface eau/ sédiments similaire aux conditions en zones humides. Dans nos expériences, la bioturbation est réalisée par une population d’oligochètes Tubificidae bien connue comme un ingénieur écologique. L’Influence de cette biodiversité sur les flux et bilan de masse de polluants modèles, a été démontré à l’aide de 2 expériences de laboratoire mettant en oeuvre des séries de microcosmes contaminées avec du Cadmium en tant que métal trace avec une concentration initiale de 20 µg.L-1, dans l’eau surnageante, et de l’atrazine marquée avec une concentration de 5 µg.g-1 de sédiment frais en tant que micropolluant organique persistant et herbicide. Les résultats de ces expériences démontrent que le bio-transport créé par la population de tubificidae ainsi que la bioremédiation sont toujours actifs en présence de contamination ce qui confirme le potentiel de dévelopement de ces organismes en ingénierie écologique. La bioadvection du sédiment et des contaminants par les tubificidae est quantifiée grâce à l’utilisation de luminophores (traceurs particulaires). Le processus de bioconvoyage engendre l’ensevelissement et le renouvellement de la quantité de polluant autour du système racinaires des Typha latifolia. Les coefficients d’enrichissement des racines (ECR) pour le Cadmium sont plus élevés en présence de bioturbation. L’interaction entre l’activité des tubificidés et la contamination par le Cd depusi la colonne d’eau Influence le bilan de masse du cadmium. Les flux de métaux du sédiment vers les racines sont estimés de l’ordre de 0.02+/-0.00 et 0.07+/-0.03 µg Cd par jour sans et avec bioturbation, respectivement. Dans le cas de l’atrazine, l’Influence de la bioturbation sur les propriétés chimiques du sédiment (pH, porosité, matière organique, etc.) explique les modifications observées au niveau du processus d’adsorption-désorption de l’atrazine. Il est démontré que la bioturbation par les tubificidae accélère la mobilité de l’atrazine et favorise sa biodisponibilité en passant de la forme adsorbée sur les particules de sédiment vers la forme libre dans l’eau interstitielle. Les coefficients d’enrichissement de l’atrazine dans les racines de typha sont également plus élevés en présence de bioturbation (7.16 ± 0.66) que sans bioturbation (5.99 ± 0.64). La biodégradation de l’atrazine est significativement plus avancée en présence de bioturbation comme le démontre un nombre plus élevé de métabolites, et une quantité de métabolites plus importante mesurés dans les racines Typha latifolia.Notre étude démontre le potentiel apporté par la bioturbation comme facteur d’amélioration des performances de la phytoremédiation, pour un métal lourd comme pour un micro-polluant organique. Les recherches futures sur la base de ces résultats devraient s'intéresser à mettre en évidence l’Influence de la bioturbation sur les communautés de micro-organismes en charge de la dégradation des composés organiques persistants

The functional role of emergent macrophytes in nature-based solutions (NBS) aiming to mitigate nutrient loading in freshwater ecosystems

Freshwater degradation in combination with water scarcity are the reason of severe stress on water resources globally. As one third of the world’s population does not have access to clean drinking water, nutrient pollution is imperative to be addressed. Nature-based solutions (NBS) is a recent concept to address several societal challenges, using techniques inspired by nature, or by nature itself, while preserving ecosystem sustainability. Water security is among the essential societal challenges that NBS can address, with nutrient pollution of freshwater systems being one of its primary aspects. While NBS projects aiming to address nutrient loading in freshwater systems, conspicuously include emergent macrophytes in their design, there is a lack in mechanistic understanding of how these aquatic plants enhance water treatment performance. In the present doctoral thesis, we focused on bridging this knowledge gap by investigating the plants’ physical, chemical and Biological Influence on the subsurface aquatic environment and the implications for nutrient mitigation. Specifically, we examined subsurface solute transport and nutrient retention under the presence of three emergent macrophyte species; Iris pseudacorus L., Phragmites australis L., and Scirpus lacustris L., while we also explored the Influence of dissolved organic carbon (DOC) quality on nitrogen cycling. To this aim, we performed a series of three studies in a setup of 12 artificial flumes, where we used traditional methods of stream ecology (i.e. pulse additions of conservative and reactive tracers) to characterize subsurface solute transport and to estimate nutrient spiraling metrics. Likewise, we experimentally modified DOC lability in the flumes, and we characterized root system architecture of the used emergent macrophytes. Regarding the physical effect of macrophytes, our results showed that macrophytes root architecture can substantially Influence subsurface solute transport. Specifically, a dense root system of fine roots results in increased hydraulic retention, while an architecture of thicker roots leads to inverse results, facilitating infiltration. Further, we found that the chemical and Biological Influence of emergent macrophytes on the subsurface aquatic environment is species specific, and depends on physiological differences between the species, and on the plants’ capacity to create long residence times in the subsurface. Additionally, the presence of emergent macrophytes was found to facilitate N removal, while the availability of a labile C source mostly increased microbial respiration, rather than denitrification. Finally, in the general discussion of the thesis, we discuss the above results and further synthesized them with information from the literature, within the context of NBS. Overall, the present doctoral thesis elucidates some unclear aspects of the functional role of emergent macrophytes in NBS aiming nutrient mitigation, highlight the importance of suitable macrophyte species selection in NBS systems to optimize treatment performance, and provides valuable guidelines to NBS practitioners for successful macrophyte species identification.La degradació dels ecosistemes aquàtics s’ha atribuït, en gran mesura, a l’excés de concentracions de nutrients d’origen antropogènic. Aquesta degradació, combinada amb l’escassetat d’aigua, és la principal causa de l’alarmant problemàtica dels recursos hídrics a nivell global. Donat que un terç de la població mundial no té accés a aigua potable, es fa imprescindible abordar la problemàtica de la contaminació per nutrients. El concepte de solució basada en la natura (SBN) ha aparegut recentment per designar aquelles tècniques inspirades en la natura que aborden diversos reptes socials tot preservant la sostenibilitat dels ecosistemes. La seguretat de poder disposar d'aigua es troba entre els desafiaments socials més importants que avui dia afronten les SBNs, essent la contaminació per nutrients en ecosistemes aquàtics un dels seus principals objectius. Si bé els SBN destinats a abordar la problemàtica de la càrrega de nutrients en sistemes aquàtics incorporen macròfits emergents en el seu disseny de forma rutinària, hi ha una manca de coneixement dels mecanismes que expliquen com aquestes plantes aquàtiques milloren el tractament de les aigües. En aquesta tesi doctoral, ens hem proposat reduir aquesta mancança del coneixement investigant com els trets físics, químics i biològics de les plantes influeixen en el medi aquàtic subsuperficial i com afecten la mitigació de nutrients. Concretament, hem examinat el transport de soluts a la subsuperficie i la retenció de nutrients sota la presència de tres espècies de macròfits emergents; Iris pseudacorus L., Phragmites australis L. i Scirpus lacustris L.. Addicionalment, hem explorat la influència de la qualitat del carboni orgànic dissolt en el cicle del nitrogen. Per portar a terme aquest objectiu, es van realitzar una sèrie de tres treballs dissenyats amb una mateixa configuració de 12 canals artificials experimentals, i on es van aplicar mètodes tradicionals de l’ecologia fluvial (és a dir, addicions sobtades de traçadors conservatius i d’elements reactius) per poder caracteritzar el transport subsuperficial de soluts i estimar les mètriques de l’espiral de nutrients. Igualment, es va modificar experimentalment el grau de labilitat del carboni en el sistema de canals, i es va caracteritzar l'arquitectura del sistema radicular d’aquests tres macròfits emergents. Quant a l’efecte físic dels macròfits, els resultats demostren que l’arquitectura radicular dels macròfits influeix substancialment en el transport subsuperficial dels soluts. Concretament, un sistema dens d’arrels fines dóna lloc a una major retenció hidràulica, mentre que una arquitectura d’arrels més gruixudes condueix a resultats inversos, donat que faciliten la infiltració a nivell intersticial. A més, hem constatat que la influència química i biològica dels macròfits emergents en el medi subsuperficial aquàtic és específica de cada espècie, i depèn de les diferències fisiològiques entre elles, i de la capacitat de les plantes per crear major temps de residència en el medi intersticial. Addicionalment, hem provat que la presència de macròfits emergents facilita l’eliminació de nitrogen, mentre que la disponibilitat d’una font de carboni làbil fa augmentar la respiració aeròbica microbiana en lloc de la desnitrificació. Finalment, a la discussió general de la tesi, es discuteixen els resultats obtinguts i es sintetitzen dins del context de les SBN amb informació extreta de la bibliografia. En general, la present tesi doctoral posa de manifest aspectes poc coneguts sobre el paper funcional dels macròfits emergents quant a la reducció de nutrients en les SBNs. També posa en relleu la importància de fer una selecció adequada d’espècies de macròfits per a sistemes on s’apliquen SBN a fi d’optimitzar el tractament d’aigües; i proporciona valuoses pautes per a professionals que apliquen SBNs per tal d’identificar les espècies de macròfits més apropiades per la reducció de nutrients

El concepte de solució basada en la natura (SBN) ha aparegut recentment per designar aquelles tècniques inspirades en la natura que aborden diversos reptes socials tot preservant la sostenibilitat dels ecosistemes. La seguretat de poder disposar d'aigua es troba entre els desafiaments socials més importants que avui dia afronten les SBNs, essent la contaminació per nutrients en ecosistemes aquàtics un dels seus principals objectius. Si bé els SBN destinats a abordar la problemàtica de la càrrega de nutrients en sistemes aquàtics incorporen macròfits emergents en el seu disseny de forma rutinària, hi ha una manca de coneixement dels mecanismes que expliquen com aquestes plantes aquàtiques milloren el tractament de les aigües. En aquesta tesi doctoral, ens hem proposat reduir aquesta mancança del coneixement investigant com els trets físics, químics i biològics de les plantes influeixen en el medi aquàtic subsuperficial i com afecten la mitigació de nutrients. Concretament, hem examinat el transport de soluts a la subsuperficie i la retenció de nutrients sota la presència de tres espècies de macròfits emergents; Iris pseudacorus L., Phragmites australis L. i Scirpus lacustris L.. Addicionalment, hem explorat la influència de la qualitat del carboni orgànic dissolt en el cicle del nitrogen. Per portar a terme aquest objectiu, es van realitzar una sèrie de tres treballs dissenyats amb una mateixa configuració de 12 canals artificials experimentals, i on es van aplicar mètodes tradicionals de l’ecologia fluvial (és a dir, addicions sobtades de traçadors conservatius i d’elements reactius) per poder caracteritzar el transport subsuperficial de soluts i estimar les mètriques de l’espiral de nutrients. Igualment, es va modificar experimentalment el grau de labilitat del carboni en el sistema de canals, i es va caracteritzar l'arquitectura del sistema radicular d’aquests tres macròfits emergents. Quant a l’efecte físic dels macròfits, els resultats demostren que l’arquitectura radicular dels macròfits influeix substancialment en el transport subsuperficial dels soluts. Concretament, un sistema dens d’arrels fines dóna lloc a una major retenció hidràulica, mentre que una arquitectura d’arrels més gruixudes condueix a resultats inversos, donat que faciliten la infiltració a nivell intersticial. A més, hem constatat que la influència química i biològica dels macròfits emergents en el medi subsuperficial aquàtic és específica de cada espècie, i depèn de les diferències fisiològiques entre elles, i de la capacitat de les plantes per crear major temps de residència en el medi intersticial. Addicionalment, hem provat que la presència de macròfits emergents facilita l’eliminació de nitrogen, mentre que la disponibilitat d’una font de carboni làbil fa augmentar la respiració aeròbica microbiana en lloc de la desnitrificació. Finalment, a la discussió general de la tesi, es discuteixen els resultats obtinguts i es sintetitzen dins del context de les SBN amb informació extreta de la bibliografia. En general, la present tesi doctoral posa de manifest aspectes poc coneguts sobre el paper funcional dels macròfits emergents quant a la reducció de nutrients en les SBNs. També posa en relleu la importància de fer una selecció adequada d’espècies de macròfits per a sistemes on s’apliquen SBN a fi d’optimitzar el tractament d’aigües; i proporciona valuoses pautes per a professionals que apliquen SBNs per tal d’identificar les espècies de macròfits més apropiades per la reducció de nutrients

p2 01 21 the Biological Influence of brain derived neurotrophic factor bdnf on the aggressiveness of human breast cancer cells
Cancer Research, 2011Co-Authors: M X Yang, T A Martin, W G JiangAbstract:Introduction: Brain derived neurotrophic factor (BDNF) is a member of the neurotrophin superfamily and has been indicated in the pathophysiology of the nervous system and is important in a number of neurological and psychological conditions. Recently, BDNF has also been shown to play a role in the development and progression of solid tumours myeloma, lung cancer and prostate cancer. It has been recently reported that BDNF is also aberrantly expressed in human breast cancer and that a raised level of BDNF is associated with poor clinical outcome and reduced survival. The present study aimed to investigate the Biological role of BDNF in human breast cancer. Materials and methods: A panel of human breast cancer cells were used in the present study. The expression profile of BDNF was evaluated using RT-PCR. We constructed a set of anti-BDNF transgenes which were used to transfect breast cancer cells in order to generate BDNF knockdown cells (MCF ΔBDNF ). The impact of BDNF knockdown on growth and cellular migration were evaluated using standard growth assays and ECIS (electronic cell impedance sensing) technology. Results: BDNF gene transcripts were successfully detected in the breast cancer cell lines, MCF-7, MDA MB-231 and ZR 75-1. MCF-7 wild type cells were subject to transfection of anti-BDNF transgenes, followed by the establishment of BDNF knockdown sub-lines and empty vector control cells (MCF pef ). Loss of BDNF in breast cancer cells resulted in reduction of cell growth rate (growth rate in MCF ΔBDNF by day 3 1.60+/−0.11, compared with 2.06+/−0.04 in MCF pef , p=0.006). Using electric cell-substrate impedance sensing, it was found that loss of BDNF in breast cancer cells resulted in a significant increase in the rate of cellular attachment and migration (migration rate over 14 h in ohms: MCF ΔBDNF 6310.125+/−595.183 compared with MCF pef 5115.5 +/−422.394, p=0.0003). Conclusion: When BDNF was stably knocked down in MCF-7 cell lines, the growth and proliferation were decreased. The ECIS results also showed that cell migration was decreased in MCF-7 cells stably transfected with ribozymes for BDNF compared with the vector control. It is concluded that BDNF, a neurotrophic growth factor aberrantly expressed in cancers such as breast cancer, has a profound impact on the cellular behaviour of breast cancer cells and that loss of BDNF is associated with a reduction of the aggressiveness of breast cancer. BDNF is therefore a potential therapeutic target in breast cancer and its effect in human breast cancer requires further investigation. the effect of warming on grassland evapotranspiration partitioning using laser based isotope monitoring techniques

Studies addressing the effects of climatic warming on the ecosystem total water balance are scarce, and measured warming effects on the T/ET ratio in field experiments have not been seen in the literature. In this study, we quantified T/ET ratios under ambient and warming treatments in a grassland ecosystem using a stable isotope approach. The measurements were made at a long-term grassland warming site in Oklahoma during the May–June peak growing season of 2011. Chamber-based methods were used to estimate the δ2H isotopic composition of evaporation (δE), transpiration (δT) and the aggregated evapotranspiration (δET). A modified commercial conifer leaf chamber was used for δT, a modified commercial soil chamber was used for δE and a custom built chamber was used for δET. The δE, δET and δT were quantified using both the Keeling plot approach and a mass balance method, with the Craig–Gordon model approach also used to calculate δE. Multiple methods demonstrated no significant difference between control and warming plots for both δET and δT. Though the chamber-based estimates and the Craig–Gordon results diverged by about 12‰, all methods showed that δE was more depleted in the warming plots. This decrease in δE indicates that the evaporation flux as a percentage of total water flux necessarily decreased for δET to remain constant, which was confirmed by field observations. The T/ET ratio in the control treatment was 0.65 or 0.77 and the ratio found in the warming treatment was 0.83 or 0.86, based on the chamber method and the Craig–Gordon approach. Sensitivity analysis of the Craig–Gordon model demonstrates that the warming-induced decrease in soil liquid water isotopic composition is the major factor responsible for the observed δE depletion and the temperature dependent equilibrium effects are minor. Multiple lines of evidence indicate that the increased T/ET ratio under warming is caused mainly by reduced evaporation.

Efficience alicamentaire des plantes à tanins pour une production animale agroécologique
HAL CCSD, 2019Co-Authors: Marie-magdeleine CarineAbstract:Nutraceutical efficiency of tropical tannin rich plants for agroecological livestock production In view of the global challenges associated with food security and climate change by 2050, agro-ecology is an innovative option which seeks to improve in a greener way, the efficiency of production and use of resources in livestock systems. Livestock production is one of the mainstays of the world's diet, sheep and goats dominating in the tropics in terms of numbers and purposely raised for their milk and meat. However, unfavorable climatic conditions, threatens the production of livestock owing to unavailability of food resources and to increase in diseases inception. The risk of the latter is increased by globalization and human behavior, as illustrated by the high prevalence of gastrointestinal pathogens in farmed small ruminants, which, in addition, have developed significant resistance to most synthetic products worldwide. In order to enhance animal productivity, in the face of new challenges of global agriculture, alternative feed resources, prevention of disease resistance and natural resources, should be given the needed attention. Agroecology relies on fundamentals, among which, the crop-livestock synergy, found in more or less integrated in mixed farming systems (MFS). These MFS are based on certain agro-ecological principles contributing to the greater autonomy of the farm, such as the reduction of inputs on the farm, the recycling of co-products of harvest, the valorization of unconventional resources for animal feed, and the choice of local breeds for livestock. In tropical and subtropical PBS, sheep and goats of tropical breeds have a unique adaptation to tropical climates and local food resources. In addition, the presence of biomass with nutritional properties allows both to feed and care for animals. It is in this context that the valorization of plant secondary metabolites is important, given their food and / or health values.Condensed tannins (CT), which are part of plant secondary metabolites, have bio-activities that do not only Influence animal's use of plant resource, but also its health, and gas emissions into the environment. The effects include toxicity, nematicidal activity (anthelmintic), rumen nitrogen fermentation and urinary nitrogen losses, microbial ruminal fermentation orientation and CH4 emissions, and intestinal protein digestibility. These polyphenolic multi-active compounds are present in many feed resources utilized in animal production. The Biological effects of CTs depend on their concentration, structure, and molecular weight. The knowledge, and therefore the characterization of CT is an important criterion for the choice of the plant resource to be utilized. The study of the bioactive potential of plant resources rich in CT, for animal production, must therefore focus on the qualitative and quantitative evaluation of the compounds in plant materials.The research project, presented in this HDR, was conducted as part of the INRA-URZ research unit scientific project (which I have been part of since 2007), with the support of 44 students trainees (from Bac + 2 ; to Master degree) and 1 PhD thesis. The study sought to ascertain the chemical structures, nutritional value and anthelmintic characteristics of CTs from tropical resources and the optimal utilization of plant resources, while reducing the environmental footprint (CH4 emissions). The chemical and bio-active (anthelmintic, fermentation, nutraceutical) characterizations of CTs was concurrently studied with the small ruminant farming systems, before moving towards a finalized section for application to the breeding system (pellet test).Studies on CT have been done, partly, in support of our Biological experiments, in particular for the evaluation of the CT content in the tested plant species. However, current assay methods do not allow for the comparison of CT concentrations of samples that do not belong to the same plant species. Based on the analytical database that generated, the current study focuses on the development of a common standard and a calculation model (equation) to estimate the CT content of plants, and to allow the comparison of samples, independently of the internal standards of each plant species. In addition, the results obtained during the chemical characterization of the CTs highlighted the presence of specific galloyls and epi-afzelechins groups in some plant species. These functional groups, rarely found in such high proportions, could Influence the Biological effect of CTs. On the other hand, the database, derived from the wide phytochemical screening of plant resources containing CT, supplemented with data obtained by near-infrared spectroscopy (NIRS), was evaluated during a first calibration study, for the prediction of levels and the discrimination of CT of the 3 model resources Manihot esculenta, Leucaena leucocephala, and Cajanus cajan. A NIR database has thus been created and can now be used to evaluate these 3 model resources, in terms of CT (3 plant species, 3 botanical families, 481 PIR spectra and CT contents: 246 with Manihot esculenta, 64 with Leucaena leucocephala and 81 with Cajanus cajan). A quantitative method for the rapid prediction of CT contents using the NIRS method was developed for Manihot esculenta (cassava) and Cajanus cajan (pigeon pea). The coefficients of correlations of the calibration straight lines for the estimation of the CT contents and standard errors of cross-validation (SECV) were 0.64 and 3.16% for Manihot esculenta, and 0.87 and 2.95% for Cajanus cajan. Standard Errors of Prediction (SEP) were 3.89% and 1.58% respectively for Manihot esculenta and Cajanus cajan.On the other hand, the study of fifteen tropical plants with CT, carried out for their Biological Influence on animal productions, validated their nutritional, fermentative and anthelmintic effects. The CTs of all tested resources had significant depressive effects on CH4 emissions (eg 4.7%, 29.3% and 28.9% for Glyricidia sepium, Leucaena leucocephala and Manihot esculenta compared to the control, gCH4 / kg of digestible organic matter ingested ) and on the fecal excretion of gastrointestinal nematode eggs by the animals (from 33 to 80% reduction of parasite fecal egg count, for a ≥50% quantity of ingested DM). In addition, the fertility of female worms and the eggs of the gastrointestinal parasitic nematode H. contortus appeared to be weakened. The fermentation profiles and the effects on the parasitic stages being variable according to different CT, the structure-activity link was thus highlighted. Similarly, the differences in CT reactivity at the different parasite stages (from 0 to 60% efficiency on the larval development of L1 to L3 stages and from 0 to 99% efficiency on the exsheathment of the L3 infesting stage, at the concentration of 2.5 mg / ml CT extract, relative to the negative control) are explained, depending on the dose, but are also related to the chemical structure. For example, according to our results, prodelphinidin CTs would be more effective on parasite development, while prodelphinidin monomers would be more effective on L3 stage larval exsheathment. The analysis of the chemical and Biological databases established, allow us, in part, to strengthen the structure-activity link thereafter. In addition, a more in-depth study of the structural differences in CTs should allow a better understanding of the differences in action and in the CT-protein interactions involved.For the use of the 3 forages with CT, namely, Manihot esculenta, Leucaena leucocephala and Cajanus cajan, selected for their dual nature (presence of CT and proteins) and their availability on the farm, drying and granulation of the forages promoted the nutraceutical effect on Manihot esculenta, Cajanus cajan and Leuacaena leucocephala. This process preserved dietary and anthelmintic effects. In addition, the mixture of the 3 resources showed a synergistic in vitro effect against the nematode H. contortus, and substantially increased by granulation (+ 3% inhibition of larval exsheathment of H. contortus). Moreover, quantities of plants with CT were determined (75 to 150 g of fresh product, i.e. 15 to 30 g of dry product, per kilo of live weight) making it possible to treat the animals against gastrointestinal parasitism (reduction of the parasite infestation of animals at 67% relative to the alfalfa control), while ensuring good growth (increase in post-infestation growth of 6.8, 31.1, 45.8 and 21.7 g / day respectively for Manihot esculenta, Cajanus cajan, Leucaena leucocephala, and the combination of the three forages). It is now a matter of refining the values by studying a dosing regimen.For the future, an exploration of the potentials of the NIR technology and chemometrics, should be considered by coupling databases from phytochemical and Biological works on plant resources with CT, with those obtained from the NIR study. A high-throughput qualitative and quantitative analysis of CT rich plants could thus contribute to developing, more rapidly, the knowledge of the anthelmintic, nutritional and environmental values of tropical CT rich plants, for small ruminant breeding. It may also lead to a tool for predicting the bio-activity of the resources on the health, production and emissions from animals.Face à l’enjeu mondial de sécurité alimentaire dans un contexte de changement climatique à horizon 2050, l’agro-écologie est une option innovante, permettant d’améliorer de manière plus écologique, l’efficacité de la productivité et l’utilisation des ressources dans les systèmes d’élevage. Les productions animales, sont l’un des supports de l’alimentation mondiale, avec en zone tropicale, une prédominance pour l’élevage de moutons et de chèvres, qui sont associés à la production de lait et de viande. Cependant, du fait des aléas climatiques, la production de protéines d’origine animale est menacée par la diminution de la disponibilité des ressources alimentaires, et par l’augmentation du développement des maladies. Le risque de ces dernières est augmenté par la mondialisation et les comportements humain, comme l’illustre la forte prévalence des pathogènes nématodes gastro-intestinaux chez les petits ruminants d’élevage, qui de surcroit, ont développé mondialement une résistance importante à la plupart des produits anthelminthiques de synthèse. Afin d’accroitre les productions animales, il s’agit donc, dans un contexte de changement climatique, de protéger les animaux des maladies et de produire des aliments alternatifs, tout en valorisant les ressources naturelles. L’agro-écologie s’appuie sur des fondamentaux parmi lesquels, la synergie cultures-élevage, déclinée dans les systèmes de type polyculture élevage (SPE) plus ou moins intégrés. Ces SPE reposent sur certains principes agro-écologiques contribuant à la plus grande autonomie de l’exploitation, comme la réduction des intrants à la ferme, le recyclage des co-produits de récolte, la valorisation de ressources non conventionnelles pour l’alimentation animale, et le choix de races locales pour le bétail. Dans les SPE tropicaux et subtropicaux, les moutons et chèvres de races tropicales présentent une adaptation unique aux climats tropicaux et aux ressources alimentaires locales. De surcroit, la présence de biomasse aux propriétés alicamentaires permet à la fois de nourrir et de soigner les animaux. C’est dans ce contexte que la valorisation des métabolites secondaires des plantes prend toute son importance, au regard de leurs valeurs alimentaires et/ou santé.Les tanins condensés (TC), qui font partie des métabolites secondaires des plantes, ont des bio-activités qui impactent l’utilisation de la ressource végétale par l’animal, sa santé, et différentes émissions dans l’environnement, notamment en termes de toxicité, d’activité nématicide, de fermentescibilité de l’azote dans le rumen et émission d’azote urinaire, d’orientation des fermentations microbiennes du rumen et des émissions de CH4, et de digestibilité intestinale des protéines. Ces composés polyphénoliques multi-actifs, sont présents dans de nombreuses ressources végétales valorisables pour les productions animales. Les effets biologiques variables des TC dépendent de leur concentration, leur structure et de leur poids moléculaire. La connaissance, et donc la caractérisation des TC est un critère important pour le choix de la ressource végétale à valoriser. L’étude du potentiel bio actif des ressources végétales riches en TC, pour une utilisation en productions animales, doit par conséquent être liée à une évaluation qualitative et quantitative des composés dans les matières végétales. Le projet de recherche, présenté dans le cadre de cette HDR, a été mené dans le cadre du projet d’unité de l’INRA – URZ, dont je fais partie depuis 2007. Avec l’appui de 43 étudiants stagiaires (de Bac+2 à Bac+5) et d’1 thèse, sur la période 2010-2019, il s’est agi par mes travaux, d’acquérir une meilleure connaissance (structures chimiques, modes d’action alimentaire et anthelminthique) des TC issus de ressources tropicales, afin de fournir des éléments permettant d’optimiser l’utilisation des ressources végétales en élevage tropical, pour améliorer les productions et santé animales, tout en réduisant l’empreinte environnementale (émissions de CH4). Les travaux de recherche ont donc été menés de manière parallèle, selon les volets de caractérisations chimique et bio-active (anthelminthique, fermentaire, alicamentaires) des TC, en lien avec les systèmes d’élevage de petits ruminants, avant d’évoluer vers un volet finalisé pour une application au système d’élevage (test de granulés alicamentaires). Les travaux sur la connaissance chimique des TC ont été développés, pour partie, en support à nos expérimentations biologiques, notamment pour l’évaluation des teneurs en TC dans les espèces végétales testées. Cependant, les méthodes de dosage actuelles ne permettent pas de comparer les concentrations en TC d’échantillons n’appartenant pas à la même espèce végétale. En prenant appui sur la base de données analytiques que nous avons générée, nous travaillons actuellement à la mise au point d’un standard commun et d’un modèle de calcul (équation), afin d’estimer la concentration en TC de plantes, et de permettre la comparaison d’échantillons, indépendamment des standards internes à chaque espèce végétale. De plus, les résultats obtenus lors de la caractérisation chimique des TC mettent en évidence la présence de groupements galloyls et epi-afzelechines particuliers dans certaines espèces végétales. Ces groupes fonctionnels, rarement retrouvés dans des proportions aussi élevées, pourraient influer sur l’effet biologique des TC. D’autre part, la base de données, issue du large screening phytochimique de ressources végétales à TC, complétée de données obtenues par spectroscopie dans le proche infrarouge(SPIR), a été valorisée lors d’une première étude de calibration, pour la prédiction des teneurs en TC, et la discrimination des 3 ressources modèles Manihot esculenta, Leucaena leucocephala, et Cajanus cajan. Une base de données PIR a ainsi été constituée, et est désormais mobilisable pour l’évaluation de ces 3 ressources modèles, sur le plan des TC (3 espèces de plantes, 3 familles botaniques, 481 spectres PIR et teneurs en TC : 246 Manihot esculenta, 64 Leucaena leucocephala, 81 Cajanus cajan). Une méthode quantitative applicable aux Manihot esculenta (manioc) et Cajanus cajan (pois d’angole), pour la prédiction rapide des teneurs en TC à l’aide de la méthode SPIR a été développée. Les coefficients de corrélations des droites de calibration pour l’estimation des teneurs en TC et les erreurs-standards de validation croisée (SECV) sont respectivement de 0,64 et 3,16% pour Manihot esculenta, et de 0,87 et 2,95% pour Cajanus cajan ; et les erreurs de prédictions (SEP) de 3.89% et 1.58% respectivement pour Manihot esculenta et Cajanus cajan.D’autre part, l’étude d’une quinzaine de plantes tropicales à TC, pour leurs effets biologiques applicables en productions animales, a validé des effets alimentaires, fermentaires et anthelminthiques. Les TC de toutes les ressources testées ont des effets dépressifs significatifs sur les émissions de CH4 (ex : respectivement – 4.7% ; - 29.3% et – 28.9% pour Glyricidia sepium, Leucaena leucocephala et Manihot esculenta par rapport au témoin, gCH4/kg de matière organique digestible ingérée (MODI) et sur l’excrétion d’œufs de nématodes gastro-intestinaux par les animaux (de 33 à 80% de réduction d’OPG pour une quantité de MSI ≥50%). De plus, la fécondité des vers et les œufs du nématode parasite gastro-intestinal H. contortus semblent fragilisés. Les profils fermentaires et les effets sur les stades parasitaires étant variables en fonction des TC, le lien structure-activité a donc été mis en évidence. De même, les différences de réactivités des TC sur les différents stades parasitaires (de 0 à 60% d’efficacité sur le développement larvaire des stades L1 à L3 ; et de 0 à 99% d’efficacité sur le dégainement au stade L3 infestant, à la concentration de 2.5 mg/ml d’extrait de TC, relativement au témoin négatif) s’expliquent, selon la dose, mais sont également liées à la structure chimique. Par exemple, d’après nos résultats, les TC à prodelphinidines seraient plus efficaces sur le développement du parasite, tandis que les monomères à prodelphinidines seraient davantage efficaces sur le dégainement larvaire. L’analyse de la base de données chimiques et biologiques, que nous avons constituée nous permettra, en partie, d’approfondir le lien structure-activité par la suite. En complément, une étude plus poussée des différences structurales des TC devra permettre de mieux appréhender les différences d’action, et les interactions TC-protéines en cause.Pour l’utilisation des 3 plantes alicamentaires à TC, Manihot esculenta, Leucaena leucocephala et Cajanus cajan, sélectionnées pour leur caractère dual (présence de TC et de protéines) et leur disponibilité sur l’exploitation, nous avons mis au point un procédé de séchage des plantes, et démontré que le procédé de séchage-granulation, que nous utilisons, est adapté à un usage alicamentaire du Manihot esculenta, du Cajanus cajan et du Leuacaena leucocephala. De plus ce procédé préserve des effets alimentaires et anthelminthiques. En outre, le mélange des 3 ressources montre un effet synergique contre le parasitisme par le nématode H. contortus in vitro, et sensiblement augmenté par la granulation (+3% d’inhibition du dégainement larvaire d’H. contortus). Des quantités de plantes à TC (75 à 150g de produit frais, soit 15 à 30g de produit sec, par kilo de poids vif) permettant de traiter les animaux contre le parasitisme gastro-intestinal (réduction de l’infestation parasitaire des animaux de 33 à 67% par rapport au témoin luzerne), tout en assurant une bonne croissance (augmentation de la croissance post infestation, de 6.8 ; 31.1 ; 45.8 et 21.7 g/j respectivement pour Manihot esculenta, Cajanus cajan, Leucaena leucocephala, et le mélange de ces 3 dernières plantes), ont été déterminées. Il s’agit maintenant d’affiner les valeurs en étudiant un schéma posologique. Pour l’avenir, une exploration des potentiels de la technologie SPIR et de la chimiométrie doit être envisagée, en couplant les bases de données issues des travaux phytochimiques et des recherches sur la bio-activité alicamentaires et fermentaires des ressources végétales à TC, avec celles issues de la SPIR. Une analyse qualitative et quantitative haut débit des plantes à TC pourra ainsi contribuer à développer, de manière plus rapide, la connaissance des valeurs antiparasitaire, alimentaire et environnementale des plantes tropicales à TC pour les petits ruminants d’élevage. De plus, cela pourra également aboutir à un outil de prédiction de la bio-activité des ressources à TC sur la santé, la production et les émissions des animaux
