    Abstract:

    [ES] Esta tesis doctoral se enmarca en uno de los retos demandados por la sociedad relativo a la seguridad y calidad alimentaria, establecido en los actuales planes estatales y europeos de investigación científica, técnica y de innovación. En particular, la investigación realizada en esta tesis doctoral, contribuye a la validación de una técnica de análisis novedosa para la detección de pesticidas y antibióticos de interés en la miel. Esta técnica está basada en el uso combinado de la tecnología de sensores acústicos de alta resolución (High Fundamental Frequency Quartz Crystal Microbalance - HFF-QCM) funcionalizados con anticuerpos monoclonales, como Elemento de reconocimiento biológico, para la detección de dos pesticidas (carbaril y DDT) y un antibiótico (sulfatiazol), en la matriz alimentaria miel. La metodología desarrollada se presenta como una técnica de análisis, alternativa y/o complementaria a las ya existentes, que resuelve algunos inconvenientes de las mismas; con la ventaja de requerir bajo consumo de muestra, ser fiable, simple, de bajo coste y de fácil manejo, por lo que no necesita de personal altamente cualificado. Además, es una técnica rápida que proporciona los resultados en pocos minutos, y está basada en instrumentación portátil, lo que facilita su implantación en la línea de trabajo. Los límites de detección alcanzados en miel por los inmunosensores HFF-QCM para carbaril, DDT y sulfatiazol fueron: 7, 24 y 0.10 µg/kg, respectivamente. Por su parte, los límites de cuantificación para los mismos analitos fueron: 17, 52 y 2 µg/kg, respectivamente. Los valores obtenidos con esta nueva tecnología son del mismo orden de magnitud que las técnicas de referencia actuales (cromatografía, SPR y ELISA), y confirman que cumple con los LMRs establecidos en el marco normativo actual. Respecto a la validación de esta nueva tecnología, el inmunosensor HFF-QCM desarrollado para la detección de sulfatiazol cumplió los requisitos de precisión y exactitud establecidos por la guía SANCO 12571/2013 para concentraciones de sulfatiazol en miel iguales o superiores a 10 ¿g/kg. Los inmunosensores HFF-QCM desarrollados para la detección de carbaril y DDT se aproximaron a los criterios de validación recomendados por la guía SANCO 12571/2013. Los resultados obtenidos con la técnica propuesta para el análisis de mieles reales han sido corroborados mediante cromatografía. En este sentido, ninguna de las muestras de miel reales analizadas mediante cromatografía y HFF-QCM reveló la presencia de pesticidas o de antibióticos en la miel, lo que confirma la fiabilidad de la técnica para la detección de estos contaminantes. Los hallazgos obtenidos en esta tesis son prometedores y establecen un punto de partida para futuros desarrollos encaminados a lograr la detección simultánea de varios analitos en un único análisis y muestra. Esto podría lograrse con un array de sensores HFF-QCM. In particular, the research carried out in this doctoral thesis contributes to the validation of a novel analysis technique for the detection of pesticides and antibiotics of interest in honey. This technique is based on the combined use of high resolution acoustic sensor technology (High Fundamental Frequency Quartz Crystal Microbalance - HFF-QCM) functionalized with monoclonal antibodies, as a Biological Recognition Element, for the detection of two pesticides (carbaryl and DDT ) and an antibiotic (sulfathiazole), in the honey food matrix. The developed methodology is presented as an analysis technique, alternative and / or complementary to those that already exist, which solves some of their drawbacks; with the advantage of requiring low sample consumption, being reliable, simple, low cost and easy to use, so it does not require highly qualified personnel. In addition, it is a fast technique that provides results in a few minutes, and is based on portable instrumentation, which facilitates its implementation in the industry. The detection limits reached in honey by the HFF-QCM immunosensors for carbaryl, DDT and sulfatiazole were: 7, 24 and 0.10 µg / kg, respectively. On the other hand, the quantification limits for the same analytes were: 17, 52 and 2 µg / kg, respectively. The values obtained with this new technology are of the same order of magnitude as the current reference techniques (chromatography, SPR and ELISA), and confirm that it meets the MRLs established in the current regulatory framework. Regarding the validation of this new technology, the HFF-QCM immunosensor developed for the detection of sulfathiazole meet the precision and accuracy requirements established by the SANCO 12571/2013 guideline for concentrations of sulfathiazole in honey equal to or greater than 10 ¿g / kg. The HFF-QCM immunosensors developed for the detection of carbaryl and DDT approached the validation criteria recommended by the SANCO guide 12571/2013. The results obtained with the proposed technique for the analysis of real honeys have been verified by chromatography. In this sense, none of the real honey samples analyzed by chromatography and HFF-QCM revealed the presence of pesticides or antibiotics in honey, confirming the reliability of this technique for the detection of these contaminants. The findings obtained in this thesis are promising and establish a starting point for future developments aimed at achieving the simultaneous detection of several analytes in a single analysis and sample. This could be accomplished with an array of HFF-QCM sensors.     Abstract:

    The environmental copper pollution seriously threatens the health of organisms and the safety of ecosystem. Therefore, the development of a simple and sensitive method to detect copper ion is very important. In this study, we have developed a fluorescent biosensor based on Biological Recognition Element pyoverdine to selectively detect copper ion. The fluorescence of pyoverdine is quenched obviously after binding with copper ion. A good linearity within the range of 0.2-10 mu M (R= 0.997) is attained and the detection limit is 50 nM. The biosensor has been successfully utilized for the detection of copper ion in drinking water, seawater and bio-samples and the results agree well with those obtained by the inductively coupled plasma mass spectrometry. Therefore, the established biosensor is a creditable method to detect copper ion with high sensitivity and selectivity, which can be utilized as a powerful tool to monitor copper pollution in the environment.     Abstract:

    Biosensors have gained immense acceptance in the field of medical diagnostics, besides environmental, food safety and biodefence applications due to its attributes of real-time and rapid response. This synergistic combination of biotechnology and microelectronics comprises a Biological Recognition Element coupled with a compatible transducer device. Diabetes is a disease of major concern since the ratio of world population suffering from it is increasing at an alarming rate and therefore the need for development of accurate and stable glucose biosensors is evident. There are many commercial glucose biosensors available yet some limitations need attention. This review presents a detailed account of the polypyrrole based amperometric glucose biosensors. The polymer polypyrrole is used extensively as a matrix for immobilization of glucose oxidase enzyme owing to its favourable features such as stability under ambient conditions, conductivity that allows it to be used as an electron relay, ability to be polymerized under neutral and aqueous mild conditions, and more. The simple one-step electrodeposition on the electrode surface allows easy entrapment of the enzyme. The review is structured into three categories (a) the first-stage biosensors: which report the studies from the inception of use of polypyrrole in glucose biosensors during which time the role of the polymer and the use of mediators was established. This period saw extensive work by two separate groups of Schuhmann and Koopal who contributed a great deal in understanding the electron transfer pathways in polypyrrole based glucose biosensors, (b) the second-stage biosensors: which highlight the shift of polypyrrole from a conventional matrix to composite matrices with extensive use of mediators focused at improving the selectivity of response, and (c) third-stage biosensors: the remarkable properties of nanoparticles and carbon nanotubes and their outstanding ability to mediate electrontransfers have seen their indispensable use in conjugation with polypyrrole for development of glucose biosensors with improved sensitivity and stability characteristics which is accounted in the review, which thus traces the evolution of polypyrrole from a conventional matrix, to composites and thence to the form of nanotube arrays, with the objective of addressing the vital issue of diabetes management through the development of stable and reliable glucose biosensors.

    Abstract Biosensor technology is based on a specific Biological Recognition Element in combination with a transducer for signal processing. Since its inception, biosensors have been expected to play a significant analytical role in medicine, agriculture, food safety, homeland security, environmental and industrial monitoring. However, the commercialization of biosensor technology has significantly lagged behind the research output as reflected by a plethora of publications and patenting activities. The rationale behind the slow and limited technology transfer could be attributed to cost considerations and some key technical barriers. Analytical chemistry has changed considerably, driven by automation, miniaturization, and system integration with high throughput for multiple tasks. Such requirements pose a great challenge in biosensor technology which is often designed to detect one single or a few target analytes. Successful biosensors must be versatile to support interchangeable bioRecognition Elements, and in addition miniaturization must be feasible to allow automation for parallel sensing with ease of operation at a competitive cost. A significant upfront investment in research and development is a prerequisite in the commercialization of biosensors. The progress in such endeavors is incremental with limited success, thus, the market entry for a new venture is very difficult unless a niche product can be developed with a considerable market volume.

  • Development of a smart bandage by detection of metalloproteinases

    Abstract:

    En 2017, il a été estimé que 425 millions de personnes souffraient de diabète. Les raisons de ce boom, liées au contexte socio-économique actuel, représentent une dépense globale de 1.3 milliard de dollars par an avec des estimations atteignant jusqu’à 2.1 milliards de dollars en 2030 (Bommer et al., Diabetes Care, 2018). Au-delà de dérèglements physiologiques, cette maladie peut engendrer de nombreux effets secondaires si elle n’est pas traitée correctement, notamment la formation de plaies chroniques, plaies ne suivant pas le schéma de cicatrisation habituel. Parmi les différentes plaies chroniques se trouvent les plaies du pied diabétique avec une prévalence globale de 6,3% (Sen, Advances in Wound Care, 2019) et une probabilité de développement chez les personnes diabétiques de 25% (Boulton et al., The Lancet, 2005). Ces plaies possèdent de nombreux facteurs participant à ce phénomène de pérennisation, dont les métalloprotéases matricielles ou MMP, endopeptidases à zinc aux rôles variés.Ces travaux de thèse s’intéressent à l’élaboration d’un dispositif de pansement connecté pour le suivi de l’évolution des plaies du pied diabétique. Ce suivi se fait par mesure de la concentration d’une enzyme clé dans les phénomènes d’inflammation de ce type de plaie, la métalloprotéase matricielle 9 (MMP9), également appelée Gelatinase B. Ce système se base sur l’utilisation d’alumine poreuse fonctionnalisée avec un Élément de Reconnaissance Biologique (ERB) capable d’interagir avec la cible, MMP9.Dans un premier temps, une recherche d’ERB affine pour la cible est étudiée. Cette recherche se base sur un screening de banques d’anticorps de camélidé (VHH) et de banques peptidiques par la méthode de phage display.La suite du projet se focalise sur l’élaboration de deux prototypes de biocapteurs capacitifs, l’un avec l’étude de la fonctionnalisation d’un anticorps conventionnel anti-MMP9, l’autre avec l’utilisation d’un VHH tiers, affine pour la GFP. This monitoring is done by measuring the concentration of a key enzyme in the inflammation process, MMP9 or Gelatinase B. This system is based on the use of porous alumina functionalized with a Biological Recognition Element (BRE) able to detect MMP9.The first part of the project is the search for BREs based of the screening of camelid antibodies (VHHs) or peptide libraries by phage display. Then, the project focuses on the development of two prototypes of capacitive biosensors, one for the study of the functionalization of a conventional anti-MMP9 antibody, the other with the use of a third-party VHH and its target, GFP.
