The present study examines the effects of a fixed combination of valerian, passion flower, lemon balm, and butterbur extracts (Ze 185) on Biological and affective Responses to a standardized psychosocial Stress paradigm. Purpose The aim of the present study was to investigate the efficacy of Ze 185 on cortisol and anxiety Stress Responses to acute psychosocial Stress in healthy subjects. Study Design This study was a randomized, placebo-controlled, double blind study with 3 parallel groups. Methods 72 healthy male participants were randomized to 3 groups (Ze 185, placebo or no treatment) during 4 days prior to a standardized psychosocial Stress paradigm. Principle outcomes were salivary cortisol and self-reported anxiety Responses to Stress assessed at the fourth day. Results The Stress paradigm induced significant and large cortisol and self-reported anxiety Responses. Groups did not differ significantly in their salivary cortisol Response to Stress, but participants in the Ze 185 condition showed significantly attenuated Responses in self-reported anxiety in comparison to placebo (F(3, 41) = 3.33, p = 0.03) and no treatment (F(3, 43) = 2.77, p = 0.05). Conclusion The results show that Ze 185 significantly attenuated the subjective emotional Stress Response during an acute Stress situation, without affecting Biological Stress Responses. Given that a circumscribed Biological Stress Response is to be considered as an adaptive mechanism, Ze 185 reduces self-reported anxiety Response to Stress without affecting assumingly adaptive Biological Stress Responses.

Materials and methods: 72 healthy male participants were randomized to Ze 185 (fixed combination of 4 herbal extracts), placebo or no treatment. 4 days post-baseline, all participants underwent a standardized psychosocial Stress paradigm (Trier Social Stress Test) [1]. Participants took either tablets of Ze 185 or placebo or received no treatment during 4 days prior to and the day of the Stress exposure. PsychoBiological Stress Responses were repeatedly assessed with the salivary cortisol, heart rate, heart rate variability and self-reported anxiety Responses. Key findings: The standardized psychosocial Stress paradigm had a significantly impact on all three psychoBiological parameters in conditions. Groups did not differ significantly in their salivary cortisol, heart rate and heart rate variability Responses over time, but participants in the Ze 185 group showed a significantly decreased Stress-induced elevation of subjective anxiety Response in comparison to placebo (p = 0.04) and a faster return to baseline.\ud \ud Conclusion: The results show that Ze 185 significantly attenuates the subjective emotional Stress Response during an acute Stress situation, without affecting Biological Stress Response systems. Some methods commonly used to collect blood samples may alter the Stress Response. As such, noninvasive means to analyze cortisol levels are increasingly being developed. We adapted an existing collection method to simultaneously obtain saliva from multiple socially living hamadryas baboons (Papio hamadryas hamadryas) and validated an enzyme-immunoassay kit to quantify cortisol within the saliva samples. Over a period of 12 months, saliva samples were regularly collected from approximately half of the 18-member colony, representing younger monkeys who were more willing to participate. The assay met the four criteria typically used to assess the effectiveness of a new analytical technique: parallelism, precision, accuracy, and sensitivity. Cortisol levels were also proportional to those expected given published plasma levels of cortisol in baboons. Further, salivary cortisol levels increased in individuals following significant Stress-related events, such as removal from the group, indicating Biological validation. The technique provided a reliable and effective means to assess a physiological indicator of Stress in a social group without initiating a Stress Response owing to handling or sedation, and provided a real-time assessment of cortisol levels and reactivity. 2244 – Maternal prenatal anxiety and epigenetic modification glucocorticoid receptor gene: the shaping of the Biological Stress Response

This study investigates both the association between maternal cortisol and emotional state during pregnancy and the methylation state of the promoter region of NR3C1 gene. Methods We examined 83 pregnant women. Psychological data and diurnal cortisol data were assessed to evaluate maternal Stress once each trimester. DNA methylation at different loci of the NR3C1 gene, including exon 1B, 1D and 1F, was analyzed in genomic DNA from cord blood mononuclear cells. Results Univariable analyses indicated pregnancy related anxiety to be the strongest psychological parameter throughout pregnancy. Most significant findings concerned 1F. Particularly the methylation state of CpG9 was significantly associated with maternal emotional wellbeing. In a multivariable model the proportion of variance in methylation state of F9 explained (PVE) by pregnancy related anxiety was 7.8% (p=0.023) during T1. Furthermore different CpG-units located at the nerve growth factor inducible protein A (NGFI-A) binding sites of 1F were associated with maternal anxiety [(F20.21: PC PRAQ and fear of integrity in T1: respectively PVE:8.9% and PVE:9.0%; Fear of delivery T2: PVE:8.0%, Fear of integrity T2: PVE:6.0% and STAI T2: PVE:5.9%) - (F12.13: PC PRAQ T1: PVE:6.9%, fear of integrity T2: PVE:6.0% and fear of delivery T2: PVE:8.0%)] and cortisol (F38.39: PVE: 8.9%) in T3. Conclusion These data indicate that prenatal maternal emotional state, particularly pregnancy related anxiety, are associated with the methylation state of the NR3C1 gene in the child.
