    Blackberry viruses are pervasive, decreasing growth, yield, and plant longevity. In a quest to identify viruses associated with Blackberry yellow vein, a disease caused by virus complexes, a new double-stranded DNA virus, referred to as Blackberry virus F (BVF), a putative member of the genus Badnavirus, family Caulimoviridae, was identified. The virus was found in both cultivated and wild Blackberry samples collected from several states in the southern United States. Population structure, host range, and association with disease symptoms were assessed. As BVF integrates into the plant genome, it affects the production of virus-free propagation material, the cornerstone for certification programs.

    Abstract Blackberry yellow vein disease is one of the most important diseases of Blackberry in the United States. Several viruses are found associated with the symptomology but Blackberry yellow vein associated virus (BYVaV) appears to be the most prevalent of all, leading to the need for a better understanding of its epidemiology. Efficient detection protocols were developed using end-point and quantitative reverse-transcription polymerase chain reaction. A multi-state survey was performed on wild and cultivated Blackberry to assess the geographical distribution of the virus. Two whitefly species, Trialeurodes abutilonea and T. vaporariorum, were identified as vectors and 25 plant species were tested as potential BYVaV hosts. The information obtained in this study can be used at multiple levels to better understand and control Blackberry yellow vein disease.

    Abstract:

    Symptoms of leaf vein yellowing and bush decline in Blackberry were attributed to infection by a novel crinivirus named Blackberry yellow vein associated virus (BYVaV). The disease is an emerging threat to Blackberry production because it can cause substantial yield loss. The objective of this study was to identify the source and means of spread of BYVaV. A survey of Blackberry plants for BYVaV from wild, cultivated, and nursery stocks was conducted. Insect traps and healthy Blackberry sentinel plants were placed among symptomatic plants in a production field throughout two growing seasons to monitor the occurrence of potential vectors and virus spread. Virus indicator plants were grafted with BYVaV-infected Blackberry because this virus was latent in some Blackberry cultivars, but indicator plants failed to express symptoms when infected with BYVaV. Reverse-transcription polymerase chain reaction detection revealed the occurrence of BYVaV in Blackberry nurseries in the United States, in production fields in Arkansas, South Carolina, and North Carolina, and in wild Blackberry populations in Arkansas. Whiteflies (Trialeurodes packardii and T. ruborum), potential vectors of BYVaV, were observed on sticky traps placed in Blackberry fields and were found colonizing Blackberry plants; however, transmission studies failed to produce whitefly-mediated transmission of BYVaV. Further understanding of the disease etiology is needed to devise viable management strategies for this disease.

Blueberry and Blackberry pomace showed excellent antioxidant potential that was not diminished after the film production. Chitosan matrix was not significantly changed to influence permeability to oxygen and mechanical properties, while water vapour permeability slightly decreased. Only the film stiffness increased with the addition of extract. Visible and significant colour changes of dry pH indicator films occurred with changing pH. The film colour was visibly transformed from pale yellow for control film, to blue-green and purple (with negative and positive a* values, respectively) with the addition of blueberry and Blackberry pomace extracts, respectively. With changing pH from 2 to 10, films with blueberry changed from rose to blue green and with Blackberry from red to dark violet. The most significant change was observed in the pH range from 4 to 7 that is important for determination of pH change due to the food spoilage in real foodstuff. Blackberry pomace extract had 4× more polyphenols than blueberry one. As expected, when extracts were added to chitosan films an increase in polyphenol content was also determined and antioxidant activity significantly increased. Films with Blackberry pomace extract showed the highest antioxidant capacity probably due to the fact that already pure Blackberry pomace extract was better antioxidant than the blueberry one. This result pointed high antioxidant activity of all produced films.
