  • osmotic stress and salt stress inhibition and gibberellin mitigation of leaf elongation associated with up regulation of genes controlling cell expansion
    Environmental and Experimental Botany, 2016
Co-Authors: Patrick Burgess, William A Meyer, Bingru Huang
    Abstract:

Leaf elongation rates (LER) is sensitive to salt stress and osmotic stress, and gibberellic acid (GA) may mediate stress-inhibition of leaf elongation by affecting genes controlling cell expansion. The objectives of this study were to examine the differential inhibitory effects of osmotic stress and salt stress on LER for fast- and slow-growing genotypes of tall fescue (Festuca arundinacea) and to determine whether GA could mitigate stress-induced decline in LER. Plants of fast-growing ‘K-31′ and slow-growing ‘Bonsai’ were grown hydroponically and treated with GA under either osmotic or salt stress. LER of both genotypes were inhibited by osmotic and salt stress (46–86%) due to reduced cell elongation and production rate. ‘Bonsai’ LER was reduced to a greater extent than that of ‘K-31′ due to greater reduction in cell production rate. Exogenous GA application significantly enhanced LER regardless of stress for both genotypes, which was more pronounced in ‘Bonsai’ than in ‘K-31′ caused by greater cell elongation and production rates. Quantitative PCR analysis revealed that four expansin genes (EXPA4, EXPA5, EXPA7 and EXPB7) and one XET gene (XET2) were down-regulated by osmotic stress and EXPA7 was down-regulated by salt stress in both genotypes. Three expansin genes (EXPA4, EXPA5 and EXPA7) were up-regulated by GA treatment in both genotypes under non-stress conditions, in which EXPA4 was also up-regulated by GA treatment in both genotypes under osmotic stress. The expression level of XET1 and XET3 increased in ‘K-31′ and ‘Bonsai’, respectively, with exogenous GA treatment under non-stress and salt stress. This study demonstrated the roles of GA regulation of genetic variations in leaf differential responses to osmotic and salt stress involving different expansins and XET genes.

  • gibberellin regulation and genetic variations in leaf elongation for tall fescue in association with differential gene expression controlling cell expansion
    Scientific Reports, 2016
Co-Authors: Sanalkumar Krishnan, Emily Merewitz, Bingru Huang
    Abstract:

Leaf elongation rate (LER) is an important factor controlling plant growth and productivity. The objective of this study was to determine whether genetic variation in LER for a fast-growing (‘K-31’), and a dwarf cultivar (‘Bonsai’) of tall fescue (Festuca arundinacea) and gibberellic acid (GA) regulation of LER were associated with differential expression of cell-expansion genes. Plants were treated with GA3, trinexapac-ethyl (TE) (GA inhibitor), or water (untreated control) in a hydroponic system. LER of ‘K-31’ was 63% greater than that of ‘Bonsai’, which corresponded with 32% higher endogenous GA4 content in leaf and greater cell elongation and production rates under the untreated control condition. Exogenous application of GA3 significantly enhanced LER while TE treatment inhibited leaf elongation due to GA3-stimulation or TE-inhibition of cell elongation and production rate in leaves for both cultivars. Real-time quantitative polymerase chain reaction analysis revealed that three α-expansins, one β-expansin, and three xyloglucan endotransglycosylase (XET) genes were associated with GA-stimulation of leaf elongation, of which, the differential expression of EXPA4 and EXPA7 was related to the genotypic variation in LER of two cultivars. Those differentially-expressed expansin and XET genes could play major roles in genetic variation and GA-regulated leaf elongation in tall fescue.

  • root physiological characteristics associated with drought resistance in tall fescue cultivars
    Crop Science, 2000
    Co-Authors: Bingru Huang, Hongwen Gao
    Abstract:

Knowledge of root traits associated with drought tolerance is important for further understanding drought tolerance mechanisms of the whole plant. The experiment was designed to investigate effects of drought stress on root physiological activities of six cultivars (Kentucky.31, Falcon II, Houndog V, Phoenix, Rebel Jr., and Bonsai) of tall fescue (Festuca arundinacea Schreb.) varying in drought resistance. Grasses were grown in well-watered or drying (nonirrigated) soil for 35 d in a greenhouse. Drying reduced root length and dry mass in the 0- to 20-cm layer for all six cultivars. Root length and dry mass in the 40- to 60-cm layer was enhanced for Houndog V, Falcon II, and Kentucky-31; was not affected for Phoenix and Bonsai; and was reduced for Rebel Jr. by soil drying. Water uptake rates for Falcon II and Kentucky-31 decreased with soil drying in the 0- to 20cm layer but increased in the 40- to 60-cm layer. Soil drying limited water uptake by Rebel Jr. in both layers. Drought stress increased root mortality in the 0- to 20-, 20- to 40-, and 40- to 60-cm layers, but the increase was most dramatic in the surface soil layer. The increase in root mortality in each soil layer was most severe for Rebel Jr. and least severe for Kentucky-31. Root death of tall fescue cultivars during drought was positively correlated with root desiccation, as evidenced by severe leakage of organic solutes from roots in drying soil. Carbohydrate supply to roots was not a contributor to root death during drought stress. This was supported by the increased or unaffected total nonstructural carbohydrates in both shoots and roots, and the increased C allocation to roots under soil drying conditions.

  • commodity risk assessment of black pine pinus thunbergii parl Bonsai from japan
    EFSA Journal, 2019
Co-Authors: Claude Bragard, Katharina Dehnenschmutz, Francesco Di Serio, Paolo Gonthier, Marieagnes Jacques, Josep Anton Jaques Miret, Annemarie Fejer Justesen, Alan Macleod, Christer Sven Magnusson, Panagiotis Milonas
    Abstract:

The EFSA Panel on Plant health was requested to deliver a scientific opinion on how far the existing requirements for the Bonsai pine species subject to derogation in Commission Decision 2002/887/EC would cover all plant health risks from black pine (Pinus thunbergii Parl.) Bonsai (the commodity defined in the EU legislation as naturally or artificially dwarfed plants) imported from Japan, taking into account the available scientific information, including the technical information provided by Japan. The relevance of an EU-regulated pest for this opinion was based on: (a) evidence of the presence of the pest in Japan; (b) evidence that P. thunbergii is a host of the pest and (c) evidence that the pest can be associated with the commodity. Sixteen pests that fulfilled all three criteria were selected for further evaluation. The relevance of other pests present in Japan (not regulated in the EU) for this opinion was based on (i) evidence of the absence of the pest in the EU; (ii) evidence that P. thunbergii is a host of the pest; (iii) evidence that the pest can be associated with the commodity and (iv) evidence that the pest may have an impact in the EU. Three pests fulfilled all four criteria and were selected for further evaluation (Crisicoccus pini, Sirex nitobei and Urocerus japonicus). For the selected 19 pests, the risk mitigation measures proposed in the technical dossier were evaluated. Limiting factors on the effectiveness of the measures were documented. For each of the 19 pests, an expert judgement is given on the likelihood of pest freedom taking into consideration the risk mitigation measures acting on the pest, including any uncertainties. For all evaluated pests, the median likelihood of the pest freedom is 99.5% or higher and within the 90% uncertainty range it is 99% or higher.
