    Abstract:

    Soak and rinse methods were compared for the recovery of Salmonella from whole cantaloupes. Cantaloupes were surface inoculated with Salmonella cell suspensions and stored for 4 days at 2 to 6 degrees C. Cantaloupes were placed in sterile plastic bags with a nonselective preenrichment Broth at a 1:1.5 cantaloupe weight-to-Broth volume ratio. The cantaloupe Broths were shaken for 5 min at 100 rpm after which 25-ml aliquots (rinse) were removed from the bags. The 25-ml rinses were preenriched in 225-ml portions of the same uninoculated Broth type at 35 degrees C for 24 h (rinse method). The remaining cantaloupe Broths were incubated at 35 degrees C for 24 h (soak method). The preenrichment Broths used were buffered peptone water (BPW), modified BPW, lactose (LAC) Broth, and Universal Preenrichment (UP) Broth. The Bacteriological Analytical Manual Salmonella culture method was compared with the following rapid methods: the TECRA Unique Salmonella method, the VIDAS ICS/SLM method, and the VIDAS SLM method. The soak method detected significantly more Salmonella-positive cantaloupes (P < 0.05) than did the rinse method: 367 Salmonella-positive cantaloupes of 540 test cantaloupes by the soak method and 24 Salmonella-positive cantaloupes of 540 test cantaloupes by the rinse method. Overall, BPW, LAC, and UP Broths were equivalent for the recovery of Salmonella from cantaloupes. Both the VIDAS ICS/SLM and TECRA Unique Salmonella methods detected significantly fewer Salmonella-positive cantaloupes than did the culture method: the VIDAS ICS/SLM method detected 23 of 50 Salmonella-positive cantaloupes (60 tested) and the TECRA Unique Salmonella method detected 16 of 29 Salmonella-positive cantaloupes (60 tested). The VIDAS SLM and culture methods were equivalent: both methods detected 37 of 37 Salmonella-positive cantaloupes (60 tested).

    The relative effectiveness of Rappaport-Vassiliadis (RV) medium, selenite cystine (SC) Broth, and tetrathionate (TT) Broth for the recovery of Salmonella spp. from foods with a low microbial load was determined. RV medium made from its individual ingredients and incubated at 42°C was compared with a commercial preparation of SC Broth, incubated at 35°C, and TT Broth incubated at 35 and 43°C, for the recovery of Salmonella spp. Twenty-one artificially contaminated food types that included dairy foods, spices, and egg products, as well as other low-microbial-load foods, were analyzed. The foods were inoculated with single Salmonella serovars at target levels ranging from 0.04 to 0.4 CFU/g. No significant differences (P ≤ 0.05) among the selective enrichment Broths for the recovery of Salmonella spp. from 18 of the foods were observed. Significantly fewer Salmonella-positive test portions of gelatin, guar gum, and nonfat dry milk were recovered with RV medium than with SC Broth incubated at 35°C and TT Broth...

    Abstract:

    Rapid detection of methicillin-resistant Staphylococcus aureus (MRSA) by PCR can be performed directly from nasal specimens with the IDI-MRSA assay. To improve the efficiency of screening, we evaluated the performance of the IDI-MRSA assay for the detection of MRSA from pooled and unpooled specimens cultured in a selective Broth. Of the 287 specimens evaluated, 71 were culture and PCR positive, 203 were culture and PCR negative, 3 were culture positive and PCR negative, 8 were culture negative and PCR positive, and 2 remained inhibited. A methicillin-susceptible Staphylococcus aureus isolate was recovered from five of the eight specimens with false-positive PCR results. Compared to the results of culture, the sensitivity, specificity, and negative and positive predictive values of the IDI-MRSA assay for detection of MRSA from Broth were 96%, 96%, 90%, and 98%, respectively. Following implementation of the IDI-MRSA assay, PCR-positive Broths were subcultured for evaluation of assay performance. Of the 298 IDI-MRSA assay-positive Broths, the results for 103 could not be confirmed by culture. A methicillin-susceptible S. aureus (MSSA) isolate was recovered from 77 of these 103 Broths. Repeat testing by the IDI-MRSA assay directly with the MSSA isolates confirmed the original positive PCR result. The positive predictive value of the IDI-MRSA assay fell from 90% during the evaluation phase to 65% postimplementation. The IDI-MRSA assay performed well for the detection of MRSA from a selective Broth compared to the performance of the detection of MRSA from culture. However, because of the burden associated with implementation of infection control precautions, cultures remain essential in confirming positive IDI-MRSA results.

    Abstract:

    Although radial impellers, especially the Rushton turbine, are widely used in stirred bioreactors, their applicability is limited by the high apparent viscosities of the Broth. Beside the intensification of Broth circulation, the energetic efficiency and the shear effects on biocatalysts must be taken into account to select the optimum impeller or impellers combination. In this context, the paper presents a comparative study on the efficiency of seven different radial impellers for simulated Broth mixing in a stirred bioreactor. The analysis was made by means of the mixing time values obtained by vertically changing the position of the pH-sensor in the Broths, in correlation with the energy consumption needed for a certain level of mixing time or for the uniform distribution of the mixing intensity into the bulk volume of the Broths.

    Abstract:

    The previous works on simulated Broths are continued and developed for Propionibacterium shermanii Broths. The obtained results indicated the considerable increase of k _L a in presence of n -dodecane as oxygen-vector and the existence of a certain value of hydrocarbon concentration that corresponds to the maximum mass transfer rate of oxygen. The magnitude of the positive effect of the oxygen-vector strongly depends on operational conditions of the bioreactor, on Broth characteristics and on P. shermanii concentration.

    Detection of diarrheagenic Escherichia coli (DEC) typically depends on identification of virulence genes from stool cultures, not on stool itself. We developed a multiplex polymerase chain reaction (PCR) assay that detects key DEC virulence genes (stx1, stx2, eae, bfpA, ipaH, LT, STh, aaiC, aatA). The assay involved a multiplex PCR reaction followed by detection of amplicon(s) using Luminex beads. The assay was evaluated on over 100 colony and Broth specimens. We then evaluated the assay using DNA extracted from stool, colony pools, and Gram-negative Broths, using stool spiked with known quantities of DEC. Performance of the assay on stool DNA was most quantitative, while stool Broth DNA offered the lowest limit of detection. The assay was prospectively evaluated on clinical specimens in Tanzania. Stool DNA yielded higher sensitivity than colony pools compared with Broth DNA as the standard. We propose using this assay to screen for DEC directly in stool or stool Broths.

    Broth cubes are packed spices highly prized for their low cost and for the flavor given to dishes, usually carbohydrate-based. The objective of this work was to establish compressed Broth cubes, exploiting the nutritional characteristics of pirambeba ( Serrasalmus brandtii ) and tilapia ( Oreochromis niloticus ), seeking a new product to the spice market, and contribute to a proper waste disposal of the fishing industry. A formulation of this pattern was prepared, where 30% was replaced by different forms of processing of the species used, resulting in different fish Broth, to which analysis of interest were performed. From the analysis, it was concluded that the fish Broth using ground tilapia presented the best characteristics when compared to commercial Broth. Chemical composition of fish bouillon cubes with species and tilapia pirambeba showed no major differences, which proves that other waste of fish or the fishing industry may also contribute to the production of Broths. The Broth commercial was less variety of polyunsaturated fatty acids, and low contents of calcium and phosphorus minerals, and more lipids compared to fish Broth prepared.
