    Abstract:

    ABSTRAK Minyak atsiri bunga kenanga (Cananga odorata ( Lmk ) Hook.f & Thoms) mempunyai aktvitas sebagai repelan terhadap nyamuk Aedes aegypti. Penggunaaan secara langsung pada kulit kurang efektif dan khasiatnya kurang maksimal karena sifat minyak atsiri yang mudah menguap, sehingga perlu diformulasikan dalam bentuk gel. Penelitian ini bertujuan untuk mengetahui pengaruh variasi konsentrasi CMC Na terhadap sifat fisik dan efektifitas minyak atsiri bunga kenanga sebagai repelan nyamuk Aedes aegypti. Minyak atsiri diperoleh dengan destilasi uap air. Dalam penelitian ini dibuat tiga formula gel minyak atsiri bunga kenanga dengan konsentrasi CMC Na 3%, 4%, 5%. Gel minyak atsiri bunga kenanga ini digunakan untuk uji sifat fisik gel ( pH, viskositas, homogenitas, daya sebar, daya lekat ) dan uji daya repelan dilakukan dengan memasukan tangan naracoba dalam sangkar dengan interval waktu 5 menit dan waktu pemaparan 30 detik. Data analisis hasil uji daya sebar, daya lekat dan daya repelan nyamuk dianalisis dengan anova satu arah, data hasil uji viskositas dianalisis dengan uji anova dua arah dengan taraf kepercayaan 95% dan dilanjutkan dengan uji BNT. Analisis uji korelasi dilanjutkan dengan regresi pada taraf kepercayaan 95% untuk mengetahui hubungan viskositas gel dengan daya repelan gel minyak atsiri bunga kenanga. Hasil penelitian menunjukkan bahwa konsentrasi CMC Na berbanding terbalik dengan daya sebar tapi berbanding lurus dengan daya lekat, viskositas dan daya repelan gel minyak atsiri bunga kenanga. Kata kunci : Minyak atsiri bunga kenanga, gel, CMC Na, repelan, Aedes aegypti. ABSTRACT Volatile oil of Cananga (Cananga odorata (Lmk ) Hook.f & Thoms) flower has an activity as repelant against Aedes aegypti. It is less effective if it is used immediately because it will be vapor easily in the air, so it is formulated into gel. This research was aimed to know the effect of concentration variation of CMC Na toward its physically characteristic and repellency against Aedes aegypti. Volatile oil was taken by distillation method. This research was held by making three formulas with different concentration of CMC Na, they are 3%, 4%, 5%. Analyzed parameters are physically characteristic ( pH, viscosity, homogeneity, dispersive and adhesive) and repellency. The result of dispersive, adhesive and repellency was analysed by one way anova test, viscosity was analysed by univariate with 95% significance level continued by least significant defference (LSD Test ). Corelation and regresion with 95% significance level was done to know the relation of gel, viscosity with repellency gel of Cananga flower volatile oil. The result of this research showed that concentration of CMC Na opposite- proportional with dispersive ability but directly proportional with adhesive ability, viscosity and repellancy of Cananga flower volatile oil gel.     ABSTRAK Tujuan penelitian ini adalah untuk mengetahui pengaruh konsentrasi carbopol terhadap sifat fisik gel dan daya repelan gel minyak atsiri bunga kenanga. Minyak atsiri bunga kenanga diperoleh dengan cara penyulingan uap air. Dibuat tiga formula gel minyak atsiri bunga kenanga dengan konsentrasi carbopol yang berbeda (0,5 g, 1,0 g dan 1,5 g). Gel yang dihasilkan dievaluasi sifat fisiknya meliputi uji pH, viskositas, homogenitas, daya sebar, daya lekat dan identifikasi minyak atsiri bunga kenanga. Uji daya repelan dilakukan dengan memasukkan tangan naracoba ke dalam sangkar dengan interval waktu diluar sangkar nyamuk selama 5 menit. Ada lima kelompok yang diujikan, meliputi tangan tanpa intervensi, kontrol negatif berupa tiga formula gel tanpa minyak atsiri bunga kenanga, minyak atsiri bunga kenanga, tiga formula gel minyak atsiri bunga kenanga dan kontrol positif berupa lotion merk “X”. Hasil penelitian menunjukkan bahwa konsentrasi carbopol berbanding terbalik dengan nilai pH dan daya sebar tetapi berbanding lurus dengan daya lekat dan viskositas gel minyak atsiri bunga kenanga. Daya repelan gel minyak atsiri bunga kenanga berbanding lurus dengan konsentrasi carbopol.     Abstract:

    Cananga oil has high economical value because of its utilisation as fragrance and food flavour. In the present work, the oil was isolated by steam distillation and organic solvent extraction from dry mature flowers of Cananga odorata Hook. fil. et Thomson, forma macrophylla. Chloroform and hexane were used as solvents with different polarities. The yield of volatile oil and its grouped compounds (LOC, sesquiterpenes hydrocarbons and HOC) was investigated as a function of time. LOC consists of oxygenated monoterpenes and other light oxygenated compounds whereas HOC comprises oxygenated sesquiterpenes and other heavy oxygenated compounds. The oil yields of 5.1, 3.8 and 2.5 % dry matter were obtained with chloroform (8 h), hexane (8 h) and steam distillation (16 h), respectively. The isolation rate was described by a first-order kinetics equation. The rate was lowest with steam distillation, intermediate with hexane extraction and best with chloroform extraction. The oil composition was analyzed by gas chromatography coupled to mass spectrometry. The steam-distilled oil and chloroform oil had a similar composition, with a high content of oxygenated compounds (61.7% and 58.3%), while hexane oil was rich in sesquiterpenes hydrocarbons (66.5%).

    The isolation of Indonesian Cananga oil was carried out by a new process, instantaneous controlled pressure drop (DIC). Dry Cananga flowers (Cananga odorata Hook. fil. et Thomson, forma macrophylla) were exposed for a short time to saturated steam and then the pressure was abruptly decreased to a vacuum level (about 5 kPa). This abrupt pressure drop provokes auto-vaporization of the superheated volatile compounds, expansion and breaking of the cell walls and instantaneous cooling. In this study, the effects of two parameters of DIC process namely pressure-drop rate and flowers moisture content on the yield of essential oil and its grouped compounds such as oxygenated monoterpenes, sesquiterpenes hydrocarbons, oxygenated sesquiterpenes and other light and heavy oxygenated compounds were studied. The oil composition was analyzed by GC coupled with MS. The availability of the volatile compounds in the residual flowers was analyzed using organic solvent extraction. The quantity of compounds obtained by DIC treatment increased with pressure-drop rate. At a rate of 1.43MPa/s the total amount of compounds in the direct and residual oils was greater than in the reference oil obtained by chloroform extraction (2 h) from untreated flowers. The hydration of dry Cananga flowers had no effect on the essential oil yield.

    The isolation of Cananga oil by a new process, Instantaneous Controlled Pressure Drop (DIC), was investigated. This process consists in heating the dry Cananga (Cananga odorata Hook. fil. et Thomson,forma macrophylla) flowers for a short time period by steam, followed by an abrupt pressure drop into a vacuum (about 5 kPa). This pressure drop provokes auto-vaporization of the volatile compounds, puffing of flowers, breaking of cell walls and cooling. The effect of the process parameters, namely number of DIC cycles (1-9), saturated steam pressure (0.2-0.6 MPa), and heating time (0.5-20 min) on the oil yield and oil composition was examined. The results indicated a significant increase of oil yield with increasing processing pressure and number of DIC cycles, however the total heating time was not a significant parameter. The DIC oil was compared with the oil obtained using steam distillation (SD). DIC exhibited better results than SD concerning rapidity (4 min versus 24 h), oil yield (2.74% versus 2.60%) and also oil quality.

This pressure drop provoked auto-vaporization of the volatile compounds, puffing of flowers, breaking of cell walls, and cooling. The yields of major volatile compounds of the Cananga oil obtained from the condensate were studied as functions of heating time (0.5 to 20 min) and cycle number (1 to 8) at different steam pressures (0.2 to 0.6 MPa). The results of DIC isolation and steam distillation (SD) were compared. A satisfactory yield of oil without thermal degradation was obtained using several DIC cycles with a high steam pressure (0.6 MPa) and short heating time (<1 min). The quantity of the oil obtained by SD in 12 h was isolated by DIC in 4 min. The quality of DIC oil was better because it contained more light and heavy oxygenated compounds (LOC and HOC) than the SD oil. The DIC isolation kinetics leads to the conclusion that sesquiterpenes and LOC are located in the exogenous site and a majority of HOC in the endogenous site.

    Java kenanga or kenanga (Cananga odorata f. macrophylla) is a tropical and sub-tropical tree species that grows naturally in Indonesia and produces essential oil. This species has been cultivated in the past by community in Indonesia, mainly on their yard. The population of kenanga has decreased significantly, but in Blitar district the community is still doing cultivating the species although in a small scale. The purpose of this research is to observe the cultivation efforts by the community and also kenanga flower production in Blitar district. This research was conducted by interviewing farmers and observation of kenanga trees in the field. The results showed that cultivation efforts being made to improve the production of kenanga flower is by pruning, fertilizing, flowering routine and retrieval or emoval of kenanga fruit. The average kananga flower production reached 381 kg / year / tree in the age group above 11 years. Factors that affect flower production were tree age (years), total tree height (m), diameter at breast height (cm) and the width tree canopy (m2). The most significant factors affecting flower production were tree diameter and width of tree canopy. Cultivation efforts being made to improve the production of kenanga flower is by pruning, fertilizing, flowering routine and retrieval or removal of kenanga fruit.     Abstract:

    The suitable seed storage method is important to maintain seed viability. This study aimed to examine the effect of periods and media storage on the viability of Cananga seed (Cananga odorata (Lam.) Hook.f. & Thomson forma genuina). The treatments used in the seed viability test were 0, 2, 4, and 6 weeks as period storage and cocopeat, ash, and charcoal as media storage. The seeds were stored in the plastic at room temperature (± 25 ° C). The variables observed in this study were seed moisture content, percentage of germination, maximum growth potential, average days of germination, T 50 , germination value, and the number of seeds that germinate during storage. The percentage of C. odorata seeds germination without storage is 54%. The seed storage for 2 – 6 weeks in ash and charcoal hush caused their viability was drop. The seeds storage of C. odorata up to 6 weeks in cocopeat could maintain seeds viability with a percentage of germination that has no significant difference compared to control.     Abstract:

    Cananga is one of the forestry plant that has high economic value and benefits. The provision of high-quality seed is necessary for establishy Cananga plantation. The aim of this study was to observe the fruit morphology and analyze the physical quality of Cananga seeds. This research method consisted of (1) morphological observation of seed (fruit size, number of seeds per fruit, and their correlation); (2) physical quality testing of seed (the weight of 1 000 seeds and seed moisture content). The observations showed that the fruit length were 7.40-27.5 mm, width 0.43-16.63 mm and weight 0.36-2.88 grams. The size of fruit have significant effect on the number of seeds in the fruit, there are 1-12 seeds in one Cananga fruit. There are approximately 16 000 seeds in 1 kg of Cananga seed. Seed moisture content of fruit that taken from tree (51.20-59.20%) is higher than seed water content of fruits that fall due to ripe physiologically (34.57-37.82%). Key words: Cananga, morphological, physical quality, seed
