Maillard reaction products in sun-dried coffee Cascara have never been reported. “Instant Cascara” (IC) extract was obtained by aqueous extraction and freeze-drying. Proteins, amino acids, lipids, fatty acid profile, sugars, fiber, minerals, and vitamins were analyzed for its nutritional characterization. Acrylamide and caffeine were used as chemical indicators of safety. Colored compounds, also called melanoidins, their stability under 40 °C and in light, and their in vitro antioxidant capacity were also studied. A safe instant beverage with antioxidant properties was obtained to which the following nutritional claims can be assigned: “low fat”, “low sugar” “high fiber” and “source of potassium, magnesium and vitamin C”. For the first time, Cascara beverage color was attributed to the presence of antioxidant melanoidins (>10 kDa). IC is a potential sustainable alternative for instant coffee, with low caffeine and acrylamide levels and a healthy composition of nutrients and antioxidants.

Sensory Acceptance, Appetite Control and Gastrointestinal Tolerance of Yogurts Containing Coffee-Cascara Extract and Inulin.

In this context, we aimed to optimize the concentration of inulin in combination with 10 mg/mL of coffee-Cascara extract in yogurt while considering their effect on appetite control, gastrointestinal wellbeing, and their effect on the sensory and technological properties of the product. For this purpose, we tested four coffee-Cascara yogurt treatments in a blind cross-over nutritional trial with 45 healthy adults: a coffee-Cascara yogurt without inulin (Y0) and coffee-Cascara yogurts containing 3% (Y3), 7% (Y7), and 13% (Y13) of inulin. The ratings on sensory acceptance, satiety, gastrointestinal tolerance, and stool frequency were measured. Surveys were carried out digitally in each participant’s cellphone. Yogurt pH, titratable acidity, syneresis, and instrumental texture were analyzed. Inulin addition increased the yogurt’s firmness and consistency. Y13 achieved significantly higher overall acceptance, texture, and taste scores than Y0 (p < 0.05). Y3 presented similar gastrointestinal tolerance to Y0. However, 7% and 13% of inulin produced significant (p < 0.05) bloating and flatulence when compared to Y0. The appetite ratings were not significantly affected by the acute intake of the different yogurts. Overall, Y3 was identified as the formulation that maximized nutritional wellbeing, reaching a “source of fiber” nutritional claim, without compromising its technological and sensory properties.

Spray-Dried Cascara sagrada Extract for Direct Compression: Tablet Formulation and a Simple HPLC Method for Tablet Performance Evaluation

Despite its wide diffusion, Cascara sagrada dried extracts with good flow and compactability properties for tablet formulation by direct compression and simple methods for the determination of Cascara sagrada extract release from tablets are not available. To overcome this lack of information, this article is focused on the design of tablets based on a Cascara sagrada extract obtained by spray-drying and on the quantification of its active constituents using a novel and isocratic HPLC-UV technique, which was validated. Flow properties of the Cascara sagrada spraydried extract and of the physical mixtures of it with conventional excipients were evaluated. The different formulated powders were compacted and their properties (i.e., hardness, friability, disintegration and dissolution) were studied. The proposed tablet formulations, based on the Cascara sagrada spray-dried extract, reached the pharmacopoeial requirements for immediate release tablets. In particular, the formulas containing a superdisintegrant showed the best pharmaceutical performance. Nevertheless, the use of excipients to improve the powder flow and/or the tablets properties was not strictly necessary. Besides, the HPLC-UV method used to monitor the extract release from tablets was validated, proving to be linear within the concentration range adequate for therapeutic purposes and appropriate for routine analysis due to its simplicity, good sensitivity, accuracy and precision.

The experiments were carried out in a Buchi B-290 Mini Spray Dryer using colloidal silicon dioxide as carrier agent. A 2 5 − 1 fractional factorial statistical design was used to find adequate spray-drying operating conditions to produce Cascara sagrada powders with good flow, good compactability properties, high process yield, low moisture content, low hygroscopicity, and caking tendency. Morphology, size, structural and thermal properties of the particles were also evaluated. The operating variables studied were air inlet temperature, atomization air flow rate, pump feed rate, aspiration capacity, and feed solids concentration. The 2 5 − 1 factorial experimental design used was a good strategy to establish spray-drying operating conditions to produce Cascara sagrada extract powders with good properties for direct compression and high process yields. Colloidal silicon dioxide was an effective drying adjuvant. Its use in relatively high concentrations together with low atomization air flow rates improved powder flowability, increased the product's glass transition temperature, and decreased its moisture content and hygroscopicity.

Coffee cherry pulp contains considerable amounts of phenolic compounds and caffeine. An attempt to produce Cascara, a refreshing beverage, has been made. Six dried coffee pulp samples and a beverage called Cascara produced in Switzerland out of one of those samples were investigated. Aqueous extraction of coffee pulps revealed a content of total polyphenols between 4.9 and 9.2 mg gallic acid equivalents (GAE)/g DM. The antioxidant capacity was between 51 and 92 μmol Trolox equivalents (TE)/g DM as measured by the assay with ABTS radical. Bourbon variety from Congo and maragogype variety showed highest caffeine contents with 6.5 and 6.8 mg/g DM. In all samples chlorogenic acid, protocatechuic acid, gallic acid and rutin were present. The beverage Cascara contained 226 mg/L of caffeine and 283 mg GAE/L of total polyphenols whereas antioxidant capacity amounted to 8.9 mmol TE/L.
