dolabrane type diterpenes from the mangrove plant Ceriops tagal with antitumor activities
Fitoterapia, 2015Co-Authors: Yi Yang, Yang Zhang, Dong Liu, Min Liweber, Bing Shao, Wenhan LinAbstract:Chemical examination of the barks of mangrove plant Ceriops tagal resulted in the isolation of six new dolabrane-type diterpenes with the trivial names of tagalenes A-F (1-6), together with 10 known analogues. The structures of new compounds were elucidated by extensive spectroscopic data analyses. Tagalenes A-C (1-3) are characterized by 18-nordolabrane scaffold, while tagalene F (6) featured by a 2,3-seco dolabrane derivative. Antitumor assay revealed that seven compounds exhibited potent inhibitory effects against a panel of tumor cell lines with IC50<10 μM, while tagalsin C (8) exerted the most potent activities in comparison with the IC50 values of the rest compounds. The primary structure-activity relationship is discussed. Tagalsin C also exerted the potent effects against a panel of drug-resistant human tumor cell lines, indicating it to be a promising molecule for further evaluation as an antitumor lead compound.

METHODS Column chromatography techniques including HPLC were used for the separation and purification, and extensive spectral analysis including various 2D NMR spectra were employed for structure elucidation. RESULTS Nine compounds, namely, tagalsins A (1), ent-5alpha-dolabr-4 (18) -ene-15S,16-diol (2), squalene (3), betulinic acid (4), lup-20 (29) -en-3-on-28-oic acid (5), betulin (6), lup-20 (29) -en-3-on-28-ol (7), beta-sitosterol (8), n-hexacosanylferulate (9) were obtained. Of which 1 and 2 belong to dolabrane diterpene. CONCLUSION Compound 1 is a new compound, and 2 to 9 are isolated from this species for the first time.

Tagalsins I and J, Two Novel Tetraterpenoids from the Mangrove Plant, Ceriops tagal

The stereochemistry of 1 was further determined by single-crystal X-ray diffraction.

Struktur Vegetasi dan Zonasi Mangrove di Wilayah Pesisir Kecamatan Kwandang Kabupaten Gorontalo Utara Provinsi Gorontalo

Dikatakan kompleks karena ekosistemnya dipenuhi oleh vegetasi mangrove, juga merupakan habitat berbagai satwa dan biota perairan. Ekosistem mangrove adalah suatu sistem di alam tempat berlangsungnya kehidupan yang mencerminkan hubungan timbal Balik antara makhluk hidup dengan lingkungannya dan diantara makhluk hidup itu sendiri, terdapat pada wilayah pesisir, terpengaruh pasang surut air laut, dan didominasi oleh spesies pohon atau semak yang khas dan mampu tumbuh dalam perairan asin/payau. Penelitian ini bertujuan untuk mengetahui struktur vegetasi dan zonasi mangrove di wilayah pesisir Kecamatan Kwandang Kabupaten Gorontalo Utara Provinsi Gorontalo. Pengambilan data struktur vegetasi mangrove dengan cara membuat transek segi empat dengan panjang transek dibuat dengan ukuran 30 m x30 m (total transek sebanyak 31 transek).Hasil penelitian menemukan 16 spesies yang mendiami hutan mangrove di wilayah pesisir Kecamatan Kwandang Kabupaten Gorontalo Utara yakni spesies Rhizophora mucronata Blume, Rhizophora Apiculata Lamk, Ceriops decandra (Griff.) Ding Hou, Ceriops tagal (Perr.) C.B.Rob, Brugueira gymnorrhiza (L) Lamk, Bruguiera paviflora (Roxb) W&A, Sonneratia alba J.E. Smith, Soneratia caseolaris (L) Eng, Xylocarpus mulocensis (Lamk) Roem, Xylocarpus granatum Koen (niri), Avecennia alba Blume, Avecennia marina (Forsk) Vierh, Avicennia officinalis (L) Lamk, Acanthus ilicifolius L, Heritiera littoralis Dryand. Ex W.Ait, Aegiceras corniculatum (L.) Blanco.Profil zonasi di wilayah ini masuk pada zonasi sederhana (satu zonasi atau zonasi campuran) dimana dari pinggir pantai sampai pedalaman daratan setiap spesies saling berasosiasi dalam satu lapisan. Profil zonasi di wilayah pesisir Kecamatan Kwandang tidak terdiri atas beberapa zonasi, karena tidak ada zonasi yang murni satu genus saja, yang ditemukan hanya satu zonasi yang merupakan campuran (mixel), dimana setiap spesies tumbuh berulang sampai kearah daratan, dan tumbuh saling bercampur antara 16 spesies tersebut.Keseluruhan spesies yang ditemukan terdapat spesies Ceriops decandra yang merupakan spesies langka secara global sehingga diperlukan perhatian khusus dalam pengelolaan hutan mangrove di wilayah pesisir Kecamatan Kwandang

The constitution and the relative configuration of 1 were determined by HRMS (ESI) and extensive NMR investigations, and the absolute configuration by circular dichroism (CD) and optical-rotatory dispersion (ORD) spectroscopy in combination with quantum-chemical calculations. Decandrinin is the first 7,8-seco-ent-abietane.

The structures of these compounds were established on the basis of spectroscopic data (new compounds) or comparison with data in the literature (known compounds). This is the first report of abietane and podocarpane diterpenoids from C. decandra.

ChemInform, 2012Co-Authors: Hui WangAbstract:In this review, the literature data on phytochemical and biological investigations of the genus Ceriops are compiled. The Ceriops species are mangrove plants widely distributed along the sea coasts of Africa, Madagascar, South Asia, and South Pacific islands. To date, 43 diterpenes and 29 triterpenes have been reported from the embryos, fruits, hypocotyls, roots, stems, and twigs of C. tagal and C. decandra. Diterpenoids and triterpenoids are the main constituents of this genus. The isolated terpenes showed an enormous structural diversity and exhibited various biological properties, such as antitumor, antibacterial, and larvicidal activities.
