    Abstract:

    Abstract Functional and medicinal beverages have been used to treat and cure diseases since the beginning of civilization. Among these beverages, coffee (Coffea arabica), black/green tea (Camellia sinensis), and yerba mate (Ilex paraguariensis), have been studied due to their important functional activities. These plants present several bioactive molecules in their Chemical Matrix that are responsible for therapeutic properties. For instance, coffee is mainly composed of caffeine, chlorogenic acid (CGA), cafestol, and kahweol, while black/green tea present principally polyphenols, caffeine, and theobromine. Moreover, yerba mate is predominantly composed of caffeine, theobromine, theophylline, aglycones, ursolic acid, oleoanolic, and CGA. Some studies reported important functional properties of these plants. Previous investigations described that these plants are able to increase neural activity and to promote health, besides presenting antiinflammatory, antitumor, thermogenic, and antioxidant capacities. In this sense, the frequent intake of these functional beverages can contribute to a healthier life.

    Abstract:

    Acai (Euterpe oleracea, Mart.) is fruit broadly consumed in the world. From its Chemical Matrix is possible that acai could has some cytotoxic effect against prostate cancer (PCa). To test this hypothesis using an in vitro PCa model DU145 cell. Additionally, potential synergism between acai and docetaxel (DO), a chemotherapic drug used to treat advanced PCa was also evaluated. Cells were exposed an acai hydro alcoholic extract at different concentrations (1 to 1000 μg/mL) and its effect on viability, apoptosis and cellular proliferation was determined by MTT assay, growth cell, clonogenic assays and cell cycle analysis by flow cytometry. Differential modulation of Bcl-2 and BAX genes was also determined by Pcr quantitative in real time (qRT-PCR) analysis. Acai at lower concentrations (1-10 μg/mL) presented significant cytotoxic and antiproliferative action against PCa cells decreasing frequency of S phase cycle. Probably, this effect was associated with its strong down-regulation of Bcl-2 gene. However, acai did not contribute to improve Docetaxel effect´s on PCa cells. Acai’s PCa antitumor effects could be related to elevate concentrations of orientin plus vitexin, p-coumaric acid, apigenin and catechins present its Chemical Matrix, which are molecules with antitumor effect previously described in the literature.

Despite this, there are reports of adverse events and previous studies associating the use of ZIP the inflammatory response. It is possible to infer that bioactive molecules present in some foods could attenuate peripheral inflammatory and oxidative stress potentially triggered ZIP. This is the case of guarana xanthine-catechin Chemical Matrix (XC-Mix) that presents caffeine, theobromine, and catechin. The in vitro protocols using murine RAW 264.7 cell macrophages were ZIP-exposure in culture medium supplemented with Chemical isolated and admixture of Caf, The, and Cat. Main results showed that supplementation with isolated and XC-mix had a lowering effect on 72 h macrophages proliferation. XC-mix with 1:1:1 proportion at 25 μg/mL of each caffeine, theobromine, and catechin, molecules present lowering effect on nitric oxide levels, oxidative stress markers (DNA oxidation quantified by 8-hydroxy-2' -deoxyguanosine), lipoperoxidation, and protein carbonylation. XC-mix also decreased protein levels and downregulated genes of proinflammatory cytokines (IL-1β, IL-6, TNF-α). At contrary, XC-Mix increased levels and upregulated gene of anti-inflammatory IL-10 cytokine. The results suggest that XC-Matrix could present some beneficial action on peripheral proinflammatory effects ZIP-triggered. Complementary in vivo studies could be useful to confirm these in vitro findings described here.
