    Abstract:

    Chlamydia psittaci causes psittacosis in humans, mainly in people in contact with birds in either the setting of occupational or companion bird exposure. Infection is associated with a range of clinical manifestations from asymptomatic infection to severe atypical pneumonia and systemic disease. C. psittaci has also been associated with ocular adnexal lymphoma in human patients. The current paper describes successful doxycycline treatment of a male patient suffering from C. psittaci chronic unilateral conjunctivitis, most probably linked to the visit of a South African wildlife reserve. Increased awareness among general and occupational physicians, ophthalmologists, clinicians, and the public on the potential of C. psittaci to cause ocular infection is needed.

  • Full Genome Sequences of All Nine Chlamydia psittaci Genotype Reference Strains

    Chlamydia psittaci primarily infects birds, but zoonotic transmission occurs in people in close contact with infected birds. The clinical outcome ranges from inapparent disease to pneumonia. Here we report the genome sequences of all 9 Chlamydia psittaci genotype reference strains. Chlamydia psittaci is a Gram-negative, obligate intracellularbacterium causing respiratory disease in birds but also in mammals. Sequencing of the C. psittaci major outer membrane protein (ompA) gene identified 9 genotypes (A to F, E/B,M56, and WC) (4). Strainswith genotypesA to F are isolated frombirds, and genotypes WC and M56 represent mammalian isolates (1, 7, 8). The genotypes cluster with host species (5). Genotypes A andB are associated with psittacine birds (cockatoos, parrots, parakeets, and lories) and pigeons, respectively. Genotype C has been iso-lated fromducks and geese, whereas genotypeDwas foundmainly in turkeys. The host range of genotype E is more diverse, since it has been isolated from pigeons, ratites, ducks, turkeys, and occa-sionally humans. Genotype F was isolated from psittacine birds and turkeys.     Abstract Chlamydia psittaci is a zoonotic pathogen with a wide range of avian hosts and worldwide geographical spread. Zoonotic transfer occurs by inhalation or direct contact and may cause psittacosis or parrot disease. Host reservoirs of particular epidemiological interest include gregarious or migratory species, as colonial behavior facilitates microbial spread amongst conspecifics and a migratory ecology permits disperse over a wide geographic region. The current study detected C. psittaci antibodies in 76 of 81 (93.8%) feral Canada geese (Branta canadensis) using a species-specific enzyme-linked immunosorbent assay. Viable C. psittaci were isolated from pharyngeal swabs of 47 of 81 (58%) birds, and subsequent ompA-based genotyping revealed genotypes E, B, and E/B. Absence of clinical symptoms in these geese could reflect a strong natural protection or a persistent infection. Canada geese are considered an important alien species in Europe.     Ocular adnexa MALT-lymphomas represent approximatively 5-15 % of all extranodal lymphomas. Almost 75 % of OAMLs are localized in orbital fat, while 25 % of cases involves conjunctive. MALT-lymphomas often recognize specific environmental factors responsible of lymphoma development and progression. In particular as Helicobacter pylori in gastric MALT lymphomas, other bacterial infections have been recognized related to MALT lymphomas in specific site. Recently Chlamydia psittaci has been identified in Ocular Adnexa MALT lymphomas, with variable frequence dependently from geographic areas. Thus bacterial infection is responsible of clonal selection on induced MALT with subsequent lymphoma development. Moreover Chlamydia psittaci could promote chromosomal aberration either through genetic instability as a consequence of induced proliferation and probably through DNA oxidative damage. The most common translocation described in MALT lymphomas affects NF-kB pathway with a substantial antiapoptotic effect. Several therapeutic approaches are now available, but the use of antibiotic-therapy in specific cases, although with conflicting results, could improve the treatment of ocular adnexa MALT lymphomas.     ABSTRACT The role of polymorphonuclear neutrophils (PMNs) in the development of the specific immune response against Chlamydophila abortus (Chlamydia psittaci serotype 1) infection was studied in a pregnant mouse model involving treatment with RB6-8C5 monoclonal antibody. PMN depletion significantly affected the immune response in the liver, in which the T-lymphocyte and F4/80+cell populations decreased, particularly the CD8+ T-cell population. A Th1-like response, characterized by high levels of gamma interferon without detectable levels of interleukin 4 (IL-4) in serum, was observed in both depleted and nondepleted mice, although an increased production of IL-10 was detected in the depleted group. Our results suggest that PMNs play a very important role in the recruitment of other leukocyte populations to the inflammatory foci but have little influence in the polarization of the immune specific response toward a Th1-like response.

    Abstract The immune mechanisms in response to Chlamydophila abortus (Chlamydia psittaci serotype 1) infection were studied in C57BL/6 and CBA mice. The infection was monitored and the following aspects of the immune response were evaluated: the nature of the leucocyte infiltrate in the liver, the percentages of polymorphonuclear neutrophils (PMNs), macrophages and lymphocytes in the spleen, and the concentrations of cytokines in serum. In addition, the serum concentrations of IgG1 and IgG2a were determined. Both mouse strains showed a Th1-like immune response, with high concentrations of IFN-γ and minimal levels of IL-4; however, C57 mice differed from CBA mice in showing milder clinical signs and earlier resolution of infection. The greater ability of C57 mice than CBA mice to eliminate chlamydophilae was related to the establishment of an earlier innate immunity, based on a more pronounced PMN response, and on a greater presence of CD8+T cells.

    Strain-specific primer sequences derived from the helicase gene of an ovine abortifacient strain (S26/3) of Chlamydophila abortus (Chlamydia psittaci) were evaluated for the diagnosis of enzootic abortion in ewes (EAE) by polymerase chain reaction (PCR). C. abortus DNA was amplified from tissues submitted from ovine abortion cases using genus-specific and strain-specific primers in a standard thermal cycler. Amplification was followed by Southern blotting and hybridisation with a strain-specific probe. Real-time PCR was also evaluated using strain-specific primers in a microvolume fluorimeter-based thermal cycler (LightCycler™). Detection using both PCR methods was compared with other diagnostic methods against the standard of McCoy cell culture isolation. In this paper we report the application of strain-specific PCR as a fast, sensitive, specific method for the detection of EAE.

    Enzootic abortion in ewes (EAE) is caused by strains of Chlamydia psittaci which have the ability to invade and colonise the placenta of sheep. In an attempt to improve diagnostic methods for the detection of EAE, subtraction hybridisation was used to isolate unique fragments of the genome of an abortifacient strain (S26/3) of C. psittaci. One S26/3 strain-specific sequence identified was shown to encode a putative helicase which is repeated throughout the EAE genome. The labelled strain-specific helicase gene fragment was used in a dot-blot hybridisation test for the detection of EAE DNA in ovine placental samples. We report the identification of C. psittaci S26/3 strain-specific sequences with potential as diagnostic probes for the detection of EAE.
