J Periodont Res 2010; 45: 557–563. © 2010 John Wiley & Sons A/S Background and Objective:  The increasing rate of resistance of microorganisms to penicillin and other antibiotics has generated concern among health authorities in Latin America. The present investigation determined the in vitro susceptibility of Porphyromonas gingivalis, Fusobacterium nucleatum, black-pigmented Prevotella spp. and Aggregatibacter actinomycetemcomitans to metronidazole, amoxicillin, amoxicillin/clavulanic acid, clindamycin and moxifloxacin in patients with Chronic Periodontitis. Material and Methods:  Subgingival plaque samples from patients with Periodontitis were collected and cultured on selective and nonselective culture media. The antimicrobial susceptibility of periodontopathogenic isolates was studied in Chronic Periodontitis patients in Colombia. Metronidazole, amoxicillin, amoxicillin/clavulanic acid, clindamycin and moxifloxacin were tested on all bacterial isolates and the percentage of resistant strains was calculated. Results:  Of the 150 bacteria identified, 51 were P. gingivalis, 45 were black-pigmented Prevotella spp., 36 were F. nucleatum and 18 were A. actinomycetemcomitans. All the isolates were sensitive to amoxicillin/clavulanic acid and to moxifloxacin, but exhibited variable susceptibility patterns to the other antimicrobial agents tested. Conclusion:  The results of the present study suggest that periodontal microorganisms in patients with Chronic Periodontitis can be resistant to the antimicrobial agents commonly used in anti-infective periodontal therapy. We suggest that the indiscriminate use of antimicrobials could result in the appearance of more highly antibiotic-resistant strains of bacteria associated with periodontal diseases in our population compared with the populations of other countries.

  • Distribution of rag genotypes of Porphyromonas gingivalis in patients with Chronic Periodontitis

    OBJECTIVE: To investigate the distribution of rag genotypes of Porphyromonas gingivalis (P. gingivalis) in Chronic Periodontitis patients. METHODS: Subgingival plaque samples were collected from 50 Chronic Periodontitis patients. The occurrence of P. gingivalis was determined by polymerase chain reaction (PCR) using 16S rDNA-specific primers. Distribution of rag genotypes was assessed in P. gingivalis positive samples by PCR. RESULTS: The occurrence of P. gingivalis was 70.7%, and the distribution of four rag genotypes among P. gingivalis positive samples was as follows: rag-1 60.4%; rag-2 23.6%; rag-3 44.3%; rag-4 15.1%, respectively. CONCLUSION: P.gingivalis with various rag genotypes was present in subgingival plaque samples from Chronic Periodontitis patients, and P. gingivalis with rag-1 and rag-3 were more predominant in Chronic Periodontitis patients, which may be associated with the development of Periodontitis.

    Abstract Aim Chronic Periodontitis may influence systemic cytokines in type 2 diabetes. This study aimed to evaluate the cytokine ratios in type 2 diabetes with, and without Chronic Periodontitis. Materials & methods Gingival status, periodontal, glycemic parameters and serum cytokines were evaluated in participants grouped as healthy, Chronic Periodontitis, and type 2 diabetes with, and without Chronic Periodontitis. Results and conclusions Cytokine ratios showed significant differences in type 2 diabetes and Chronic Periodontitis, were highest in participants having both type 2 diabetes and Chronic Periodontitis, with a statistically significant cut-off point and area under curve by receiver operating characteristic.

    BACKGROUND: Resistin is a putative adipocyte derived signalling polypeptide which belongs to the group of adipokines. It acts as a proinflammatory molecule and stimulates the synthesis and secretion of proinflammatory cytokines. The aim of this study is to evaluate the levels of resistin in gingival crevicular fluid (GCF) before and after non surgical periodontal therapy in systemically healthy Chronic Periodontitis patients and in Chronic Periodontitis patients with type 2 diabetes mellitus. MATERIALS AND METHODS: The study includes a total of 37 patients with 20 systemically healthy Chronic Periodontitis patients and 17 Chronic Periodontitis patients with type 2 diabetes mellitus. The patients were selected from the outpatient ward who visited the Department of Periodontology, KSR Institute of Dental Science and Research, Tiruchengode, Tamilnadu. The study includes 2 groups: GROUP I (20 patients) : Systemically healthy Chronic Periodontitis patients and GROUP II (17 patients) : Chronic Periodontitis patients with type 2 diabetes mellitus. The clinical parameters including plaque index (PlI), gingival index (GI), probing depth (PD) and clinical attachment level (CAL) were recorded for all the patients at baseline and at 3 months following non surgical periodontal therapy. GCF samples were collected from the tooth with deepest periodontal pocket at baseline and at 3 months following non surgical periodontal therapy. The levels of resistin in GCF samples were analysed using an Enzyme Linked Immunosorbent Assay (ELISA).
