    Abstract:

    To study the α-toxin zymotechnique of type A Clostridium perfringens by using automatic mechanical stirring fermenter.Three factors,including temperature,pH value,and fermentation time were utilized in L9(34) orthogonal experiment in order to ascertain which zymotechnique was the most important factor in producing α-toxin and the growth and toxin production curves of Clostridium perfringens were drawn.The results indicated that pH value was significant for toxin producing(0.01P0.05),the fermentation time and temperature were not(P0.05).So,the optimal fermentation process of type A Clostridium perfringens in producing α-toxin was in pH7.0,at 43℃,and cultivated for 6 h.

    Emphysematous cystitis is a rare disease and is usually caused by aerobic bacteria, most commonly Escherichia coli. Only rarely have anaerobic bacteria been associated with this condition. We report a case of emphysematous cystitis due to Clostridium perfringens with bacteremia in an elderly diabetic woman.

  • Research on contrast of SDS-PAGE identifying toxin types of Clostridium perfringens

    Abstract:

    This experiment was carried out by using SDS-PAGE method to determine the molecular weight of exotoxin protein produced by 20(16 of type A,3 of type C and 1 of type D) airborne Clostridium perfringens strains from cow stable that the toxin types were examined by ELISA and 57 Clostridium perfringens strains selected from Shandong Province had been identified by multi-PCR to confirm the kind of toxin,then their toxin types were obtained.The detecting results showed that the quality concentrations of exotoxin by coarse distill of Clostridium perfringens standard strains,airborne Clostridium perfringens samples from cow stable in German and Clostridium perfringens strains separated from Shandong Province were 3.261,2.238-3.333 and 1.451-3.109 mg/mL,respectively;the quality concentrations of exotoxin by extractive distill were 0.803,0.521-0.999 and 0.530-0.812 mg/mL,respectively.The detecting results of 20 airborne Clostridium perfringens strains from cow stable were 17 of type A,2 of type C and 1 of type D,and the accordance with ELISA was 95.0%;the detecting results of 57 Clostridium perfringens strains separated from Shandong Province were all of type A,and the accordance with multi-PCR was 100%.The results showed that the difference existed between ELISA method and SDS-PAGE method and there was coherence between multi-PCR method and SDS-PAGE method.
