    Summary Ferulic, caffeic, p-coumaric and chlorogenic acids are classified as hydroxycinnamic acids, presenting anticarcinogenic, anti-inflammatory and antioxidant properties. In this work, enzymatic extraction has been studied in order to extract high value-added products like hydroxycinnamic acids from Coffee Pulp. A commercial pectinase and enzyme extract produced by Rhizomucor pusillus strain 23aIV in solid-state fermentation using olive oil or Coffee Pulp (CP) as an inducer of the feruloyl esterase activity were evaluated separately and mixed. The total content (covalently linked and free) of ferulic, caffeic, p-coumaric and chlorogenic acids was 5276 mg per kg of Coffee Pulp. Distribution was as follows (in %): chlorogenic acid 58.7, caffeic acid 37.6, ferulic acid 2.1 and p-coumaric acid 1.5. Most of the hydroxycinnamic acids were covalently bound to the cell wall (in %): p-coumaric acid 97.2, caffeic acid 94.4, chlorogenic acid 76.9 and ferulic acid 73.4. The content of covalently linked hydroxycinnamic acid was used to calculate the enzyme extraction yield. The maximum carbon dioxide rate for the solid-state fermentation using olive oil as an inducer was higher and it was reached in a short cultivation time. Nevertheless, the feruloyl esterase (FAE) activity (units per mg of protein) obtained in the fermentation using CP as an inducer was 31.8 % higher in comparison with that obtained in the fermentation using olive oil as the inducer. To our knowledge, this is the first report indicating the composition of both esterified and free ferulic, caffeic, p-coumaric and chlorogenic acids in Coffee Pulp. The highest yield of extraction of hydroxycinnamic acids was obtained by mixing the produced enzyme extract using Coffee Pulp as an inducer and a commercial pectinase. Extraction yields were as follows (in %): chlorogenic acid 54.4, ferulic acid 19.8, p-coumaric acid 7.2 and caffeic acid 2.3. An important increase in the added value of Coffee Pulp was mainly due to the extraction of chlorogenic acid.

    Summary Hydroxycinnamic acids (HAs) have a potential application in the food and pharmaceutical industry because they are rich in phenolics. Feruloyl esterases release phenolic compounds from plant cell walls. Coffee Pulp is rich in HAs linked to polysaccharides. A solvent extraction of free HAs was performed with aqueous methanol (80 %). A response surface methodology was applied to optimise the extraction of these compounds from Coffee Pulp, and the best results were obtained at 56 °C for 34 min. Alkaline and acid hydrolyses were performed to evaluate the content of linked HAs. Treated (extracted) Coffee Pulp was used to produce feruloyl esterases in solid-state fermentation by Aspergillus tamarii V12307, previously selected by a hydrolysis plate assay. Different dilutions of a culture medium were added to the Coffee Pulp, and the diluted medium with half the nutrients allowed for higher CO2 production. A specific growth rate (mCO2 ) of 0.25 h –1 and a lag phase (tlag) of 14.3 h were observed under the selected conditions. Finally, enzymatic activities were 14.0 and 10.8 nkat per g of dried matter when methyl and ethyl ferulate were used as substrates, respectively. Productivities (9.3 and 7.2 nkat per g of dried matter per day, respectively) were higher when compared to other studies carried out in solid-state fermentation. Utilisation of Coffee Pulp for enzyme production improves the added value of this abundant by-product of the Coffee industry.

    Fresh and 3-day-old Coffee Pulp of the Arabica variety were analyzed for polyphenol composition followed by characterization by two different methods. The first method consisted in subjecting Coffee Pulp powder to direct thiolysis. For the second method, Coffee Pulp was subjected to successive solvent extractions, followed by thiolysis. Quantification of phenolic compounds was then achieved by high-performance liquid chromatography (HPLC) analysis of thiolysis products. Four major classes of polyphenols were identified: flavan-3-ols (monomers and procyanidins), hydroxycinnamic acids, flavonols, and anthocyanidins. Differences in concentration of procyanidins were observed between fresh and 3-day-old Coffee Pulp. Constitutive units were mainly epicatechin, representing more than 90% of the proanthocyanidin units, with average degrees of polymerization in the range of 3.8-9.1. Monomer to hexamer units of flavan-3-ols from fresh Coffee Pulp were separated by normal-phase HPLC. Molecular size of oligomeric proanthocyanidins was obtained by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS). Results obtained confirm the presence of oligomers of the flavan-3-ol (-)-epicatechin.

    Advances in industrial biotechnology offer potential opportunities for economic utilization of agro-industrial residues such as Coffee Pulp and Coffee husk. Coffee Pulp or husk is a fibrous mucilagenous material (sub-product) obtained during the processing of Coffee cherries by wet or dry process, respectively. Coffee Pulp/husk contains some amount of caffeine and tannins, which makes it toxic in nature, resulting the disposal problem. However, it is rich in organic nature, which makes it an ideal substrate for microbial processes for the production of value-added products. Several solutions and alternative uses of the Coffee Pulp and husk have been attempted. These include as fertilizers, livestock feed, compost, etc. However, these applications utilize only a fraction of available quantity and are not technically very efficient. Attempts have been made to detoxify it for improved application as feed, and to produce several products such as enzymes, organic acids, flavour and aroma compounds, and mushrooms, etc. from Coffee Pulp/husk. Solid state fermentation has been mostly employed for bioconversion processes. Factorial design experiments offer useful information for the process optimization. This paper reviews the developments on processes and products developed for the value-addition of Coffee Pulp/husk through the biotechnological means.

    The results of the cultivation of six strains of Pleurotus (P. djamor (2), P. ostreatus (2) and P. pulmonarius (2)) on Coffee Pulp and wheat straw are presented. Metabolic activity associated with biomass of each strain was determined, as well as changes in lignin and polysaccharides (cellulose and hemicellulose), phenolic and caffeine contents in substrate samples colonized for a period of up to 36 days. Analysis were made of changes during the mycelium incubation period (16 days) and throughout different stages of fructification. Greater metabolic activity was observed in the wheat straw samples, with a significant increase between 4 and 12 days of incubation. The degradation of polysaccharide compounds was associated with the fruiting stage, while the reduction in phenolic contents was detected in both substrates samples during the first eight days of incubation. A decrease was observed in caffeine content of the Coffee Pulp samples during fruiting stage, which could mean that some caffeine accumulates in the fruiting bodies.

    Abstract One of the compounds generally found in the residues of the Coffee and tea industries is caffeine, which in high concentration is toxic to various organisms, making it necessary to find an adequate treatment for these residues. Biotechnological treatments using enzymes can be an alternative to valorize and detoxify these residues. However, mixtures of substrates have not been evaluated to improve production. Therefore, the present investigation aimed to study the effect of different proportions of sorghum-Coffee Pulp mixtures as a substrate in solid-state fermentation with the fungus Rhizopus oryzae (MUCL 28168) for the production of n-demethylases. To evaluate the synergistic and antagonistic effects of Coffee Pulp and sorghum mixtures on n-demethylase enzyme production, a simplex-centroid design, using four levels: 1 (100%), 1/4 (25%), 1/2 (50%), 3/4 (75%). Results obtained were favorable, achieving a caffeine demethylase activity of 18.762 U/g, and reducing the caffeine content in the Coffee Pulp.

    Abstract Biotechnological advances offer potential opportunities for economic utilization of agro-industrial residues such as Coffee Pulp. Coffee Pulp is a fibrous and mucilaginous residual material obtained during the processing of Coffee cherries by the wet or dry process. The Coffee Pulp contains some amount of caffeine and tannins, which makes it toxic in nature, resulting in the disposal problem, however, the Coffee Pulp is characterized by its nutritional high-value. To increase the use of the Coffee Pulp (Arabica or Robusta) in animal feed, these compounds were removed by biodetoxification through solid-state fermentation using Rhizopus oryzae (MUCL 28,168). Temperature, moisture, and inoculum size were those studied parameters that had a significant influence on the decrement of the antiphysiological compounds. It was found that the highest decrease of caffeine (52%) and condensed tannins (52%) was achieved at pH 6.0, temperature 30 °C, initial moisture 70%, and inoculum 1 × 106 spores/g. The results show that with this process a reduction of these compounds can be achieved, allowing efficient use of the Coffee Pulp in different industries.

    Coffee Pulp is an excellent source of natural antioxidants, most of which are difficult of removing due to its chemical interactions into cell walls. These bioactive compounds can be released by fermentation-assisted extraction (FAE) process. In this study, the antioxidant properties of Coffee Pulp extracts, after FAE using several bacteria in a solid-state lactic fermentation were evaluated by ABTS and DPPH radical scavenging assays. Released bioactive compounds were identified by HPLC-MS. Lactobacillus casei enhanced the antioxidant capacity of Coffee Pulp extracts due to release of high amounts of phenolics like chlorogenic acid and resorcinol.

    Background: Coffee Pulp has a high content of antioxidants capable of modifying the oxidative status in small ruminants. However, high amounts for a prolonged time can reduce fertility. Objective: To determine the effect of two inclusion levels of Coffee Pulp during estrous synchronization on reproductive variables and oxidative status of primiparous ewes. Methods: Sixty Suffolk x Dorset primiparous ewes were distributed into three treatments in a completely randomized design; T0: (n=20) 1.5 kg balanced diet, Tl: (n=20) 1.5 kg balanced diet and 5% Coffee Pulp, T2: (n=20) 1.5 kg balanced diet and 10% Coffee Pulp. Supplementation was given for 16 days before estrus synchronization and until the beginning of the breeding season. A progestogen (CIDR®) was inserted for ll days and a dose of PGF2a was applied two days prior to its withdrawal. Estrus detection started 12 hours after CIDR withdrawal. Blood samples were obtained during the supplementation period to measure oxidative status, antioxidant capacity, glucose and insulin, and up to 9 days after breeding to determine progesterone concentration. Pregnancy diagnosis was performed at 30 and 60 days post-breeding. An analysis of repeated measures of mixed effects and frequency analysis were carried out. Results: Inclusion of Coffee Pulp for a short period prior to breeding did not affect reproductive parameters, nor progesterone, glucose or insulin concentrations (p>0.05); however, antioxidant capacity increased, while lipid oxidation showed an opposite trend (p<0.05). Conclusion: Inclusion of up to 10% Coffee Pulp in the diet of ewe lambs for 16 days prior to breeding improves oxidative status without causing adverse effects on pregnancy, estrus or prolificacy.

    Abstract: The objective of this study was to evaluate the productivity and health of fattening lambs fed different levels of Coffee Pulp in their diets. Thirty-five crossbred lambs with an average weight of 21.8 kg were fed isoproteic and isoenergetic diets with different percentages of Coffee Pulp (T0: control; T1: 7%; T2: 14%, T3: 21% and T4: 28%), the base diet was formulated with corn, soybean meal, alfalfa stubble, molasses, urea and mineral salt. The fattening period was 98 days. Productivity was measured by weight gain, feed intake and feed conversion. At the beginning and end of the study, blood samples were taken to determine the health status using a biochemical profile and blood count. Analysis of variance was performed using the initial weight as the covariate for the productive variables and the initial values of the analytes in the blood chemistry and hematological analysis tests. The Coffee Pulp did not affect productivity, although the amount of neutrophils decreased (P<0.05) as the Coffee Pulp in the diet increased. In all treatments, urea exceeded the reference values, whereas creatinine was below the reference values. We concluded that the inclusion of up to 28% Coffee Pulp in the diet did not affect the productive parameters, decreases the neutrophil count without affect health status of lambs during fattening.

    Summary Background: it is known that Coffee Pulp can modify the oxidative status and fertility in dairy cows. Objective: to evaluate the effect of dietary supplementation with Coffee Pulp on the antioxidant capacity, lipid oxidation and reproductive characteristics of ewes during estrous synchronization and early gestation. Methods: forty Dorset-Suffolk crossbred ewes with 3 or 4 parturitions were allocated to two treatments: T0 (n = 21), ewes supplemented with 450 g of a control feed; and T1 (n = 19), ewes supplemented with 450 g of the feed with 25% Coffee Pulp. Supplementation began 14 days before estrous synchronization and ended 25 days after breeding. During estrous synchronization, progestogen (CIDR, Controlled Internal Drug Release) was inserted and left in situ for 11 days. Eighteen hours later, estrous detection began with the aid of rams. Blood samples were collected at different times of synchronization and during early pregnancy to determine antioxidant capacity, lipid oxidation and blood progesterone concentration. Pregnancy diagnosis was performed 30 and 60 days after CIDR removal. Results: supplementation with Coffee Pulp did not affect estrous onset, estrous response or progesterone concentration, but fertility decreased from 100 to 78.95%. The antioxidant capacity measured using the FRAP technique was greater in Coffee Pulp supplemented ewes only before progestogen insertion. Coffee Pulp did not modify lipid oxidation; however, this variable was affected by sampling time, decreasing after progestogen removal to its lowest values at 22 days into pregnancy. Conclusion: although supplementation with Coffee Pulp at 25% in the concentrate increased antioxidant capacity of ewes before insertion of progestogen, it is not recommended to use this percentage during synchronization or early pregnancy since it can negatively affect gestation rate. Objetivo : evaluar los efectos del suministro dietario de Pulpa de cafe y su capacidad antioxidante, oxidacion lipidica, y caracteristicas reproductivas en ovejas durante la sincronizacion del estro y la gestacion temprana. Metodos: cuarenta ovejas cruzadas Suffolk x Dorset de 3 y 4 partos fueron asignadas a dos grupos: T0 (n = 21), suplementacion con 450 g de alimento (grupo testigo); y T1 (n = 19), suplementacion con 450 g de alimento con 25% de Pulpa de cafe. Resultados: la suplementacion con Pulpa de cafe no afecto el inicio del estro, la respuesta al estro ni la concentracion de progesterona. Sin embargo, la fertilidad disminuyo de 100 a 78,95%. La capacidad antioxidante, medida mediante la tecnica FRAP, fue mayor en ovejas suplementadas con Pulpa de cafe, pero solo antes de la insercion del progestageno. La Pulpa de cafe no modifico la oxidacion lipidica; sin embargo, si fue modificada por el tiempo de muestreo, decreciendo desde la remocion del progestageno hasta los 22 dias de prenez. Conclusion : aunque la suplementacion del concentrado con 25% de Pulpa de cafe incremento la capacidad antioxidante antes de la insercion del progestageno, no se recomienda ese porcentaje durante la sincronizacion y gestacion temprana, ya que redujo el porcentaje de gestacion de las ovejas.     Abstract Productive performance, rumen fermentation and oxidative status of sheep fed diets supplemented with Coffee Pulp ensiled with 5% molasses were evaluated. Thirty-four Blackbelly cross-breed sheep, 6 months old, mean weight 21.2 ± 0.63 kg, were assigned to three treatments T0: control treatment ( n  = 11), T1: treatment with 8% Coffee Pulp in the diet ( n  = 12), and T2: treatment with 16% Coffee Pulp in the diet ( n  = 11). Coffee Pulp at the levels evaluated did not affect daily weight gain, feed intake, and feed conversion ( P  > 0.05). However, water intake, acetic acid, butyric acid, and total volatile fatty acids significantly increased ( P P  > 0.05) were observed in the antioxidant capacity of the sheep's plasma; however, lipid peroxidation was lower for sheep fed with 16% Coffee Pulp. The present study concluded that a supplemented sheep diet with Coffee Pulp up to 16% Coffee Pulp did not affect their productive parameters but reduced oxidative stress.

    Abstract:

    The purpose of this study was to determine how feeding sheep Coffee Pulp affects carcass characteristics and what changes occur in physicochemical, antioxidant capacity and oxidation of the meat during refrigerated storage. The experiment was carried out in 15 Blackbelly lambs weighing an average 22.86±0.76kg. The animals were assigned to three treatments: T0=control diet, T1=diet with 8% Coffee Pulp, and T2=diet with 16% Coffee Pulp. After fattening for 56 days, the sheep were slaughtered and the carcasses assessed. The inclusion of 16% Coffee Pulp in the diet increased carcass dressing from 48.19 to 50.83% and decreased the amount of fat in rumen and intestines from 3.43 to 2.53% (P<0.05). The inclusion of Coffee Pulp in the diet did not alter the amount of crude protein or fat in meat or its oxidation and antioxidant capacity during refrigerated storage. However, the inclusion of Coffee Pulp in the diet decreased fat in the rumen and intestines, and thus increased the amount of usable meat.
