    Taylorella equigenitalis can be transmitted during artificial insemination. This report describes clinical T. equigenitalis transmission by cryopreserved stallion semen. T. equigenitalis isolates from a mare's vaginal discharge and semen from the same batch of the cryopreserved semen used for the insemination gave identical API ZYM, antibiotic susceptibility, and multilocus sequence typing results (ST-46); furthermore, the multilocus sequence typing lineage ST-46 is known to circulate in the country of semen collection. These results support the need for strict Contagious Equine Metritis screening of processed semen before use for artificial insemination.

    Abstract:

    The performance of culture and PCR methods routinely used to diagnose Contagious Equine Metritis (CEM) was evaluated and compared by two interlaboratory trials involving a total of 24 European laboratories, including 22 National Reference Laboratories for CEM. Samples were swab specimens artificially contaminated with bacteria present in the genital tract of Equidae, some with and some without Taylorella equigenitalis, the causative agent of CEM, and T asinigenitalis, responsible for possible misidentification as T equigenitalis Throughout both interlaboratory trials, PCR performed better in terms of specificity and sensitivity than the culture method, supporting the assertion that PCR should be accepted for CEM diagnosis. However, the culture performance during the second interlaboratory trial was better than during the first one, suggesting that the expertise of participants improved. This reveals the advantage of regular interlaboratory trials to constantly improve the expertise of laboratories. It also shows the need to develop new culture media that are more selective and/or better geared to the metabolism of T equigenitalis in order to improve the bacteriological diagnosis of CEM.

    Contagious Equine Metritis (CEM) is a sexually transmitted disease of equids caused by Taylorella equigenitalis , a Gram-negative microaerophilic coccobacillus of the Taylorella genus. The acute form of the disease is characterised by mucopurulent vaginal discharge and variable degrees of vaginitis, endoMetritis and cervicitis, leading to temporary infertility (Timoney 2011). It is assumed that disease dissemination results from the transfer of carrier stallions and mares, but is also linked to artificial breeding (Schulman and others 2013). After its discovery in 1977, CEM spread rapidly worldwide, causing international concern within the breeding horse industry (Matsuda and Moore 2003; Timoney 2011). It became one of the most regulated Equine diseases and is a notifiable disease of the World Organisation for Animal Health. Despite this, many countries are still considered to have endemic status for CEM within their non-Thoroughbred populations (Schulman and others 2013), probably due to several factors, for example, the absence or the shortcomings of national monitoring, surveillance and reporting programmes, and lack of knowledge about the ecology of T equigenitalis .     Objective—To determine the impact of antimicrobial-containing semen extender on the growth of Taylorella equigenitalis in semen culture-positive for Contagious Equine Metritis (CEM) and the development of CEM after artificial insemination with CEM-positive semen extended with antimicrobial-containing semen extender. Design—Prospective study. Animals—21 mature mares free of CEM, 1 mature stallion experimentally infected with CEM, and semen from a stallion naturally infected with CEM. Procedures—CEM-positive semen was incubated in semen extender with and without antimicrobials (amikacin [final concentration, 1 g/L] and penicillin G potassium [0.63 g/L]) followed by determination of the number of colony-forming units of T equigenitalis.     Contagious Equine Metritis (CEM) is a highly Contagious venereal infection of equids caused by Taylorella equigenitalis, a bacterium with fastidious growth requirements. A disease of major international concern, CEM can be the cause of short-term infertility and, very rarely, abortion in mares. Unlike the mare, stallions exposed to T. equigenitalis do not develop clinical signs of disease. CEM is transmitted by direct or indirect venereal contact. The carrier state occurs in the mare and the stallion and carrier animals are frequently the source of infection for new outbreaks of the disease. There are streptomycin-sensitive and -resistant biotypes of T. equigenitalis, and diagnosis is based primarily on culture of the bacterium from its predilection sites in the reproductive tract of the mare and the stallion. Treatment modalities are available for elimination of the carrier state. Prevention and control of CEM is achievable through a comprehensive programme of breeding farm management that includes early detection and treatment of carrier mares and stallions.

    In the present review article, recent molecular advances relating to studies with Taylorella equigenitalis, as well as the recently described second species of the genus Taylorella, namely Taylorella asinigenitalis, have been described. Molecular genotyping of T. equigenitalis strains by pulsed-field gel electrophoresis (PFGE) after digestion with the suitable restriction enzyme(s) enabled the effective discrimination of strains, thus allowing the examination of the scientific mechanism(s) for its occurrence and transmission of Contagious Equine Metritis (CEM). Alternatively, polymerase chain reaction (PCR) amplification and nucleotide sequencing of the 16S ribosomal DNA sequence and/or the other species specific sequence(s) as targets were confirmed to be effective for identification of T. equigenitalis. These new analytical methods at the genomic DNA level also enabled the discrimination of the newly discovered donkey-related T. asinigenitalis from T. equigenitalis, and moreover, the performance of phylogenetic analysis of genus Taylorella organisms with other closely related genera. Furthermore, detailed analysis of the genes responsible for CEM within the T. equigenitalis genome would be useful to help elucidate the pathogenic virulence and transmission mechanisms associated with the important Equine pathogen associated with CEM.

    The genomic DNA of eight strains of Taylorella equigenitalis, isolated from seven Norwegian Trotters and a Norwegian pony with Contagious Equine Metritis in Norway, was examined by pulsed-field gel electrophoresis after separate digestions with two restriction enzymes, namely, ApaI and NotI. The respective electrophoretic profiles of the fragments were essentially identical but differed from those of T. equigenitalis NCTC11184T and Kentucky 188. They also exhibited slight differences from profiles obtained from Japanese isolates. These results may possibly suggest a common genotype and a common source of infection for all these eight isolates in Norway.

    Profiles of the genomic DNA of 104 strains ofT. equigenitalis isolated from brood mares with Contagious Equine Metritis in Hokkaido during the breeding seasons from 1980 to 1993, as well as those of five strains (SS28, EQ56, EQ59, EQ70 and HH139) previously isolated in Japan were examined after restriction digestion and crossed-field gel electrophoresis. These profiles were essentially identical to each other and the various isolates and strains appeared to have a common genotype, designated ‘genotype J’, with respect to two restriction enzymes,ApaI andNotI. These results suggest a common source for all these isolates obtained over the course of more than 10 years in Japan.

    Background Taylorella equigenitalis is the causative agent of Contagious Equine Metritis, a sexually-transmitted infection of Equidae characterised in infected mares by abundant mucopurulent vaginal discharge and a variable degree of vaginitis, cervicitis or endoMetritis, usually resulting in temporary infertility. The second species of the Taylorella genus, Taylorella asinigenitalis, is considered non-pathogenic, although mares experimentally infected with this bacterium can develop clinical signs of endoMetritis. To date, little is understood about the basic molecular virulence and persistence mechanisms employed by the Taylorella species. To clarify these points, we investigated whether the host-pathogen interaction model Acanthamoeba castellanii was a suitable model for studying taylorellae.

    A direct-PCR assay was developed for the rapid detection of Taylorella equigenitalis, a Gram-negative bacterium responsible for Contagious Equine Metritis (CEM) in Equidae. The bacteria may be detected in Equine genital swabs without need for a preliminary step of DNA extraction or bacterial isolation. Specificity was determined with 125 isolates of T. equigenitalis, 24 isolates of Taylorella asinigenitalis, five commensal bacteria of the genital tract and a facultative intracellular pathogen of foals found in large concentration in soil. Our PCR is specific and amplified a 413-bp 16S ribosomal DNA product only in all T. equigenitalis.
