Hydrolysis of Ginseng Saponins and Quantifications of Saponins, Prosapogenins and Sapogenins in Crude Drug Extracts for Quality Control

However ginseng saponins are easily hydrolyzed in acidic solutions of Crude Drug preparations. Due to the hydrolysis of saponins in acidic condition, it is generally difficult to determine ginseng saponins in Crude Drug preparations. Ginseng saponins, prosapogenins and sapogenins of Crude Drug extracts were quantified by HPLC. Ginseng saponins were quantified by HPLC on Lichrosorb-NH₂ column with acetonitrile/water/1-butanol(80:20:10, v/v). Ginseng prosapogenin-Rg₃ and -Rg₂ were extracted with ethyl acetate from 50% acetic acid hydrolyzates of saponin fractions and quantified by HPLC on Lichrosorb-NH₂ column with acetonitrile/water(90:10, v/v). Ginseng sapogenins, panaxadiol and panaxatriol, were extracted with diethyl ether from 7%-sulfuric acid hydrolyzates of saponin fractions and quantified by HPLC on μ-Bondapak C18 column with acetonitrile/methanol/chloroform(83:10:7, v/v). These methods of analyses of sapogenins and prosapogenins were more useful for quality control than those of ginseng saponins in some of Crude Drug preparations.

The formulation displayed inhibitory action against the test organism in vitro. Subsequently mice were challenged with a virulent strain of Salmonella typhi (Ty2) and the protective effect of the formulation was evaluated with post-infective, pre-infective, single and multiple dose schedules, administered either orally or subcutaneously. A schedule that included multiple divided doses prophylactically administered had a significant therapeutic effect.

In the traditional system of medicine in India, the formulation has been prescribed for intestinal disorders. It comprises of five medicinal herbs, namely, Boerhavia diffusa, Berberis aristata, Tinospora cordifolia, Terminalia chebula and Zingiber officinale . The dried and pulverized plants were extracted in ethanol together and individually. In vitro amoebicidal activity was studied to determine the minimal inhibitory concentration (MIC) values of all the constituent extracts as well as the whole formulation. The formulation had a MIC of 1000 μg/ml as compared with 10 μg/ml for metronidazole. In experimental caecal amoebiasis in rats the formulation had a curative rate of 89% with the average degree of infection (ADI) reduced to 0.4 in a group dosed with 500 mg/kg per day as compared with ADI of 3.8 for the sham-treated control group of rats. Metronidazole had a cure rate of 89% (ADI = 0.4) at a dose of 100 mg/kg per day and cured the infection completely (ADI = 0) when the dosage was doubled to 200 mg/kg per day. There were varying degrees of inhibition of the following enzyme activities of Crude extracts of axenically cultured amoebae: DNase, RNase, aldolase, alkaline phosphatase, acid phosphatase, α-amylase and protease.

a case of Drug eruption caused by the Crude Drug boi sinomenium stem sinomeni caulis et rhizoma

A 41-year-old female patient had been diagnosed with chronic rheumatoid arthritis in the department of internal medicine. After ingestion of a decoction of the Crude Drug Boi® for the alleviation of arthralgia, a slight fever developed, which was followed by systemic edematous erythema with itching. HPLC showed that the main components of the Crude Drug Boi® are sinomenine and magnoflorine. The results of patch tests were negative for all oral Drugs that the patient had been taking. Oral ingestion tests showed that the patient showed positive reactions to the as-is Boi® boiling-water decoction and 1/10-volume sinomenine. Based on this, the Drug eruption was judged to be caused by sinomenine. It is considered the first time that the causative component of a Drug eruption was confirmed by oral ingestion tests with components of a Crude Drug of Kampo medicine (Sino-Japanese traditional medicine).
