  • Effect of Cucumis sativus L on Glucose Absorption through Intestinal Mucosal Membrane of Wistar Rat Models

    Abstract:

    Background: The Cucumis sativus L has been used in traditional medicine for several conditions, with one of them being diabetes mellitus. The purpose of this experiment is to observe the anti-hyperglycemic effect of Cucumis sativus L on glucose absorption through the intestinal mucosal membrane of Wistar rats. Methods : The research was conducted on September–November 2012 at Biochemistry Laboratory, Faculty of Medicine, Universitas Padjadjaran. Using in situ perfusion method, the rat’s epithelial mucosa in the intestine was bathed in a simple glucose solution in control, and bathed in glucose and Cucumis sativus L infusion in treatment. After admission, a sample was taken from the digestive tract and the glucose level was measured through a spectrophotometer. Result: There is no significant difference in the absorption of control and treatment solution. Conclusion : The Cucumis sativus L does not have any significant effect on the glucose absorption through the intestinal mucosal membrane. Penelitian ini dilakukan pada September–November 2012 di Laboratorium Biokimia, Fakultas Kedokteran, Universitas Padjadjaran Hasil : Tidak terdapat perbedaan bermakna antara penyerapan glukosa pada larutan kontrol dengan larutan perlakuan. Simpulan : Cucumis sativus L tidak memiliki pengaruh yang signifikan terhadap penyerapan glukosa melalui membran mukosa usus. Penelitian lebih lanjut masih diperlukan untuk mengetahui mekanisme anti hyperglycemic Cucumis sativus L.   • Effect of arsenic species on the growth and arsenic accumulation in Cucumis sativus.

    Abstract:

    The effects of arsenic (As) species, such as As(III), As(V) and dimethylarsinic acid (DMA), on the accumulation of As in cucumber (Cucumis sativus), as well as on its growth in a soil mesocosm were evaluated. When Cucumis sativus was cultivated in soils contaminated with 20 and 50 mg/kg of As(III), As(V) or DMA for 40 days, the growth was markedly inhibited by the inorganic As (As(III) and As(V)) rather than the organic As (DMA). Irrespective of the As species, the As concentrations accumulated in Cucumis sativus increased with increasing As concentration in the soil. The As bioaccumulation factors from soil into the tissue of Cucumis sativus were 17.5–35.4, 29.3–42.7 and 17.6–25.7 for As(III), As(V) and DMA, respectively. In addition, the As translocation factors from the roots to shoots were 0.025–0.031, 0.018–0.032 and 0.014–0.026 for As(III), As(V) and DMA, respectively. In conclusion, Cucumis sativus mainly accumulated As in its roots rather than its shoots and easily accumulated inorganic rather than organic As from the soil into its tissue.

    Abstract:

    Phytoextraction is a remediation technology with a promising application for removing arsenic (As) from soils and waters. Several plant species were evaluated for their As accumulation capacity in hydroponic culture amended with As. Cucumis sativus (cucumber) displayed the highest tolerance against As among 4 plants tested in this study (corn, wheat, sorghum and cucumber). The germination ratio of Cucumis sativus was more than 50% at the high concentration of 5,000 mg-As/l. In Cucumis sativus grown in a solution contaminated with 25 mg-As/l, the accumulated As concentrations in the shoot and root were 675.5 ± 11.5 and 312.0 ± 163.4 mg/kg, respectively, and the corresponding values of the translocation and bioaccumulation factors for As were 1.9 ± 0.9 and 21.1 ± 8.4, respectively. These results indicate Cucumis sativus is to be a candidate plant for phytoextraction of As from soils and water.

    Abstract:

    Cucumis sativus L. (cucumber), from the family Cucurbitaceae, is a therapeutic plant with various pharmacological benefits, broadly utilized as a part of complementary medicine (e.g., Unani, Ayurveda, Siddha, and Traditional Chinese). In light of past research discoveries, this plant had been chosen to consider its potential antibacterial action. Extracts were purified by dialysis and ion exchange chromatography strategy and then assayed for antibacterial activity against four standard pathogenic bacterial strains known to cause foodborne infections and spoilage of food and herbal drugs. Antimicrobial peptides were extracted from seeds using a sodium phosphate citrate (pH 7.2) - CTAB cradle (pH 6.0). The highest protein concentration was seen with elute fractions 1 and 3 (370 mg/mL) compared with elute fractions 2 and 4 (340 mg/mL). Among the bacteria utilized, E. coli was clearly the most sensitive out of selected four strains. Our results suggest that Cucumis sativus L seeds extracts have significant potentials as new antimicrobial agents.
