Respons Dosis Biotip Rumput Belulang (Eleusine Indica L. Gaertn) Resisten-Glifosat Terhadap Glifosat, Parakuat Dan Indaziflam Dose Response of Goosegrass (Eleusine Indica L. Gaertn) Biotype Glyphosate-Resistance to Glyphosate, Paraquat, and Indazifla
2020Co-Authors: Satria Parlindungan Dalimunthe, Edison Purba, MeirianiAbstract:Goosegrass (Eleusine Indica) is one of weeds commonly found in oil palm plantation of which is getting more difficult to control with glyphosate during the last few years in Adolina Estate, PTPN IV. This research aims to determine the dose response and level of resistance of the glyfosatresisten population to paraquat, glyphosate, and indaziflam. The rates of glyphosate applied were 0, 60, 120, 240, 480, 960, 1920 g a.i. ha -1 ; paraquat at 0, 50, 100, 200, 400, 800, 1600 g a.i. ha -1 and indaziflam at 0, 12,5, 25, 50, 100, 200, 400 g a.i. ha -1 . The treatments were arranged in randomised block design (RBD) and each treatment was made in three replicates. The results showed that the putative glyphosate-resistant population collected from Adolina Estate was resistant to glyphosate and paraquat is the survival of resistant-glyphosate was 100 % at 480 g a.i glyphosate/ha and 86,7 % at 400 g a.i paraquat/ha. The level of resistance to glyphosate and paraquat were 7,5 and 5,5 fold of the susceptible population respectively. Meanwhile, indaziflam controlled satisfactorily this population

Indica populations from oil palm plantations in North Sumatra, Indonesia have never been reported to be glyphosate-resistant. This research was aimed to determine the survival percentage and to classify the resistance of E. Indica to glyphosate from oil palm plantations in 11 districts in North Sumatra Province. The research was conducted in the Weed Research Center Field Laboratory, Faculty of Agriculture, Universitas Sumatera Utara from October 2016 to August 2017. This research was arranged in a Randomized Block Design with the glyphosate-treatment using a recommended dose of two l ha-1 and three replications. The ESU0 population was collected in the soccer field of Politeknik Negeri Medan (Medan city) was used as a check, and has never been exposed to the herbicide. Parameters were analyzed using one-way ANOVA, and the means were compared using DMRT at a probability level of 5%. The results showed that 276 (65.56%) of 421 populations were classified as glyphosate-resistant, 88 populations (20.90%) were classified as glyphosate-resistant developing, and 57 populations (13.54%) were classified as glyphosate-susceptible. The highest range of dry weight of glyphosate-resistant E. Indica populations were found in 5 districts including South Labuhanbatu District followed by the Serdang Bedagai, Simalungun, Asahan, and North Labuhanbatu Districts. The observed resistance level and dry weight range in the present study Indicated the failure in controlling E. Indica due to continued use of glyphosate and the potency of E. Indica distribution into other oil palm estates due to its high biomass production capability.

Penelitian ini bertujuan untuk melaporkan sebaran populasi dan mengklasifikasi resistensi E. Indica terhadap herbisida glifosat pada perkebunan kelapa sawit di Kabupaten Batu Bara. Penelitian ini dilaksanakan di Lahan Pusat Penelitian Gulma Fakultas Pertanian Universitas Sumatera Utara pada Oktober 2016 - Agustus 2017. Penelitian ini menggunakan glifosat dengan dosis rekomendasi 720 g b.a.ha-1 dan tiga ulangan. Populasi ESU0 (dari Lapangan Bola Politeknik Negeri Medan) sebagai pembanding. Analisis data menggunakan software IBM SPSS Statistics 20. Hasil penelitian menunjukkan bahwa populasi E. Indica yang paling resisten (63,33%) pada perkebunan kelapa sawit di Kabupaten Batu Bara terdapat pada ESU4.11 (afdeling 4 Kebun Tanah Gambus). Populasi E. Indica yang tergolong resisten-glifosat 83,33% (10 populasi), tergolong moderat resisten-glifosat 16,67% (2 populasi) dan tidak ada populasi yang sensitif-glifosat pada dosis rekomendasi 720 g b.a.ha-1 pada perkebunan kelapa sawit di Kabupaten Batu Bara.

This study aims to distribution mapping and resistance classification of E. Indica population to glyphosate herbicide of oil palm plantations in Serdang Bedagai Regency. This research was conducted on Weed Research Center Land in Faculty of Agriculture, Universitas Sumatera Utara in October 2016 to August 2017. This research used glyphosate herbicide with the recommended dose at 720 g ai ha -1 and three replications. Population ESU 0 (from Politeknik Negeri Medan Ball Field) as a comparison. Data analysis using IBM SPSS Statistics 20 software. The results showed there E. Indica population classified as glyphosate-resistant amount 89.36% (42 population), classified as glyphosate-resistant moderate amount 10,64% (5 population) and there is no population glyphosate-susceptible on oil palm plantations in Serdang Bedagai Regency of recommended dose at 720 g ai ha -1 .

As a result of repeated application of dinitroaniline herbicides, resistant biotypes of goosegrass (Eleusine Indica) developed in previously susceptible wild-type populations. We have previously reported that α-tubulin missense mutations correlate with dinitroaniline response phenotypes (Drp) (Plant Cell 10: 297–308, 1998). In order to ascertain associations of other tubulins with dinitroaniline resistance, four β-tubulin cDNA classes (designated TUB1, TUB2, TUB3, and TUB4) were isolated from dinitroaniline-susceptible and -resistant biotypes. Sequence analysis of the four β-tubulin cDNA classes identified no missense mutations. Identified nucleotide substitutions did not result in amino acid replacements. These results suggest that the molecular basis of dinitroaniline resistance in goosegrass differs from those of colchicine/dinitroaniline cross-resistant Chlamydomonas reinhardtii and benzimidazole-resistant fungi and yeast. Expression of the four β-tubulins was highest in inflorescences. This is in contrast to α-tubulin TUA1 that is expressed predominantly in roots. Collectively, these results imply that β-tubulin genes are not associated with dinitroaniline resistance in goosegrass. Phylogenetic analysis of the four β-tubulins, together with three α-tubulins, suggests that the resistant biotype developed independently in multiple locations rather than spreading from one location.

These amino acid substitutions alter hydrophobicity; therefore, they may alter the dinitroaniline binding property of the protein. These mutations were correlated with the dinitroaniline response phenotypes (Drp). Plants homozygous for susceptibility possessed the wild-type TUA1 allele; plants homozygous for resistance possessed the mutant tua1 allele; and plants heterozygous for susceptibility possessed both wild-type and mutant alleles. Thus, we conclude that TUA1 is at the Drp locus. Using polymerase chain reaction primer-introduced restriction analysis, we demonstrated that goosegrass genomic DNA can be diagnosed for Drp alleles. Although not direct proof, these results suggest that a mutation in an alpha-tubulin gene confers resistance to dinitroanilines in goosegrass.

Single and Multiple Resistance of Eleusine Indica from Asahan Regency, Indonesia

This research was aimed to the resistance classification of E. Indica population to glyphosate from one of oil palm estate in the Asahan Regency and to find out of glyphosate-resistant Eleusine (GRE) biotype has been also resistant to paraquat. This research was conducted on Weed Research Center Land, Faculty of Agriculture, Universitas Sumatera Utara, Medan from September 2017 until May 2018. Single resistance evaluation of E. Indica population was performed using glyphosate at the dose 720 g a.i.ha-1. However the multiple resistance of GRE biotype was performed using paraquat at rates of 0, 50, 100, 200, and 400 g a.i.ha-1 and glyphosate at rates of 0, 720, 1440, 2880, and 5760 g a.i.ha-1. Susceptible-population (ESU-0) was taken at ball field of Politeknik Negeri Medan (Medan City) as a comparison. The data were analyzed by ANOVA and followed by Duncan test at level 5% using IBM SPSS Statistics software. The results showed that E. Indica populations from one of oil palm estate in the Asahan Regency had survival of 81.48% (classified as glyphosate-resistant) and the dry weight of 24.35 g. Glyphosate at the dose 2880 g a.i.ha-1 and paraquat at the dose 200 g a.i.ha-1 were effectively (100%) controlled survival, tillers.pot-1, fresh weight.pot-1, dry weight.pot-1 and mortality of GRE biotype. The resistance index value of GRE biotype to glyphosate and paraquat of 4.91- and 1.26-fold, respectively compared to susceptible plants.

Similarity and Phylogenetic Analysis of Herbicide-Resistant Goosegrass (Eleusine Indica) Biotypes

This review article is aimed to obtain the basic data of similarity and phylogenetic values among herbicide-resistant E. Indica biotypes. This research used a descriptive analytical method. The analysis involved 14 nucleotide sequences of herbicide-resistant E.Indica biotypes obtained from the National Center for Biotechnology Information. The nucleotide alignment of herbicide-resistant E. Indica biotypes was conducted with ClustaLW using the Molecular Evolutionary Genetics Analysis (MEGA) v. 5.05 software based on a method of neighbor-joining tree construct/test. The results showed that two nucleotides of 7,921 herbicide-resistant E. Indica biotypes were homologous (sequence 1,231 and sequence 1,408). The similarity values among herbicide-resistant E. Indica biotypes ranged from 0.00 to 1.19. The information of phylogenetic pattern is needed in the selection of the herbicides mode of action rotation in order to control herbicide-resistant E. Indica biotypes.

This research was arranged in a Randomized Block Design with the glyphosate-treatment using a recommended dose of two l ha-1 and three replications. The ESU0 population was collected in the soccer field of Politeknik Negeri Medan (Medan city) was used as a check, and has never been exposed to the herbicide. Parameters were analyzed using one-way ANOVA, and the means were compared using DMRT at a probability level of 5%. The results showed that 276 (65.56%) of 421 populations were classified as glyphosate-resistant, 88 populations (20.90%) were classified as glyphosate-resistant developing, and 57 populations (13.54%) were classified as glyphosate-susceptible. The highest range of dry weight of glyphosate-resistant E. Indica populations were found in 5 districts including South Labuhanbatu District followed by the Serdang Bedagai, Simalungun, Asahan, and North Labuhanbatu Districts. The observed resistance level and dry weight range in the present study Indicated the failure in controlling E. Indica due to continued use of glyphosate and the potency of E. Indica distribution into other oil palm estates due to its high biomass production capability.

Penelitian ini bertujuan untuk melaporkan sebaran populasi dan mengklasifikasi resistensi E. Indica terhadap herbisida glifosat pada perkebunan kelapa sawit di Kabupaten Batu Bara. Penelitian ini dilaksanakan di Lahan Pusat Penelitian Gulma Fakultas Pertanian Universitas Sumatera Utara pada Oktober 2016 - Agustus 2017. Penelitian ini menggunakan glifosat dengan dosis rekomendasi 720 g b.a.ha-1 dan tiga ulangan. Populasi ESU0 (dari Lapangan Bola Politeknik Negeri Medan) sebagai pembanding. Analisis data menggunakan software IBM SPSS Statistics 20. Hasil penelitian menunjukkan bahwa populasi E. Indica yang paling resisten (63,33%) pada perkebunan kelapa sawit di Kabupaten Batu Bara terdapat pada ESU4.11 (afdeling 4 Kebun Tanah Gambus). Populasi E. Indica yang tergolong resisten-glifosat 83,33% (10 populasi), tergolong moderat resisten-glifosat 16,67% (2 populasi) dan tidak ada populasi yang sensitif-glifosat pada dosis rekomendasi 720 g b.a.ha-1 pada perkebunan kelapa sawit di Kabupaten Batu Bara.

This study aims to distribution mapping and resistance classification of E. Indica population to glyphosate herbicide of oil palm plantations in Serdang Bedagai Regency. This research was conducted on Weed Research Center Land in Faculty of Agriculture, Universitas Sumatera Utara in October 2016 to August 2017. This research used glyphosate herbicide with the recommended dose at 720 g ai ha -1 and three replications. Population ESU 0 (from Politeknik Negeri Medan Ball Field) as a comparison. Data analysis using IBM SPSS Statistics 20 software. The results showed there E. Indica population classified as glyphosate-resistant amount 89.36% (42 population), classified as glyphosate-resistant moderate amount 10,64% (5 population) and there is no population glyphosate-susceptible on oil palm plantations in Serdang Bedagai Regency of recommended dose at 720 g ai ha -1 .

This study was aimed to report distribution and resistance classification of E. Indica population on oil palm plantations in Deli Serdang Regency. The research was conducted on Weed Research Center in Faculty of Agriculture, Universitas Sumatera Utara from October 2016 until August 2017. This research used Randomized Block Design with recommended dose at 720 g a.i.ha-1 of glyphosate and three replications. Population ESU0 (from Politeknik Negeri Medan Ball Field) as a comparison. Data on average survival, mortality, dry weight, resistance classification of E. Indica were analyzeds using IBM SPSS Statistics 20 software. The results showed that there were 12 E. Indica populations classified as glyphosate-resistant (56.52%), 7 glyphosate-resistant developing (30.43%), and 4 populations classified as glyphosate-susceptible (13.04%) with the recommended dose at 720 g a.i.ha-1 from oil palm plantations in Deli Serdang Regency.
