Strictinin and seven other isomers, tellimagrandin I, and Ellagic Acid were identified from tea infusions. The ellagitannin content in tea infusions was determined after Acid hydrolysis and ranged from 0.15 to 4.46 mg of Ellagic Acid equivalent/g of tea in the infusions. The intake of Ellagic Acid after drinking a cup of tea brewed with 4 g of tea could range between 0.59 and 17.89 mg. These results indicate that tea can be a significant contributor to the dietary intake of ellagitannins. Urolithins, the gut microbiota metabolites produced in vivo from Ellagic Acid and ellagitannins, were detected in human urine after dietary tea beverage intake. Urolithin metabotypes A, B, and 0 were identified in volunteers after tea intake. These results suggest that the daily intake of ellagitannins from tea can have a role in tea health effects.

For this reason their potential effects preventing oxidative related diseases, such as cardiovascular diseases, have been largely studied. In vitro studies show that ellagitannins, at concentrations in the range 10–100 μM, show some relevant anti-atherogenic, anti-thrombotic, anti-inflammatory and anti-angiogenic effects, supporting the molecular mechanisms for the vascular health benefits. While there is good evidence supporting the vascular effects in vitro , the evidence on animal models or humans is much scarcer. The in vitro results often do not match the findings in the in vivo studies. This could be explained by the low bioavailability of the antioxidant ellagitannins and Ellagic Acid. The main ellagitannin metabolites circulating in plasma are Ellagic Acid microbiota metabolites known as urolithins, and they have lost their free-radical scavenging activity. They are present in plasma as glucuronide or sulphate conjugates, at concentrations in the nM range. Future studies should focus in the bioavailable metabolites, urolithins, and in the form (conjugated with glucuronic Acid or sulphate) and concentrations (nM range) in which they are found in plasma. In this review we critically discuss the role of ellagitannins and Ellagic Acid on vascular health.

Free radical studies of Ellagic Acid, a natural phenolic antioxidant.

Its antioxidant capacity has been estimated using the 1,1-diphenyl-2-picrylhydrazyl radical assay. To understand the actual mechanisms involved in antioxidant activity and the free radical scavenging ability,a nanosecond pulse radiolysis technique has been employed. The rate constants for the reactions of several reactive oxygen species and reactive nitrogen species such as hydroxyl, peroxyl, and nitrogen dioxide radicals have been found to be in the range of 10(6)-10(9) M(-1) s(-1). The Ellagic Acid radicals have been characterized by the absorption spectra and decay kinetics. Studies on the reactions of Ellagic Acid with the 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonate) radical and the radicals of Ellagic Acid with ascorbate have been used to estimate its one-electron reduction potential. Ellagic Acid has also been found to be a good scavenger of peroxynitrite. Using stopped-flow reaction analyzer with absorption detection, the rate constant for this reaction has been determined to be 3.7 x 10(3) M(-1) s (-1). The electron spin resonance spectra of the oxidized Ellagic Acid radicals have been recorded by horseradish peroxidase and hydrogen peroxide method.

Ellagic Acid (20 and 40 mg/kg, p.o.) and diazepam (0.5 mg/kg, i.p.) per se were administered once for acute treatment and 14 successive days for chronic treatment. Pentylenetetrazole (80 mg/kg, i.p.) induced tonic and clonic convulsions, but picrotoxin (6 mg/kg, i.p.) induced only tonic convulsions. Acute administration of higher dose (40 mg/kg) and chronic administration of both the doses of Ellagic Acid significantly delayed onset of convulsions, decreased duration of clonic and tonic convulsions; and reduced mortality as compared to pentylenetetrazole and picrotoxin treated control. Ellagic Acid also reversed pentylenetetrazole and picrotoxin-induced decrease in brain GABA levels. Acute and chronic administration of diazepam showed significant anticonvulsant activity and increased brain GABA levels. Thus, Ellagic Acid showed significant antiepileptic activity in mice probably through increase of GABAergic transmission in brain.

Antidepressant-like activity of Ellagic Acid in unstressed and acute immobilization-induced stressed mice
Pharmacological Reports, 2012Co-Authors: Dinesh Dhingra, Ritu ChhillarAbstract:Background The aim of present study was to evaluate antidepressant-like activity of Ellagic Acid in Swiss young male albino mice; and to explore the possible underlying mechanisms for this activity. Methods Mice were immobilized for 150 min once only for induction of stress. Ellagic Acid (8.75, 17.5, 35 mg/kg, po ) and fluoxetine (20 mg/kg, ip ) per se were administered to unstressed and stressed mice; and immobility periods were recorded using tail suspension test and forced swim test. The plasma nitrite levels were also estimated in unstressed and stressed mice. Effects of 7-nitroindazole (nNOS inhibitor), aminoguanidine (iNOS inhibitor), prazosin (a_1-adrenoceptor antagonist), sulpiride (selective D2-receptor antagonist), and p-chlorophenylalanine (p-CPA - tryptophan hydroxylase inhibitor) on antidepressant-like activity of Ellagic Acid were also evaluated. Results Ellagic Acid (17.5 and 35 mg/kg, po ) and fluoxetine per se significantly decreased immobility periods of unstressed and stressed mice, indicating significant antidepressant-like activity. There was no significant effect on locomotor activity of the mice. Ellagic Acid significantly decreased the plasma nitrite levels in stressed mice only. Aminoguanidine significantly potentiated antidepressant-like activity and plasma nitrite decreasing effect of Ellagic Acid (35 mg/kg) in stressed mice. 7-Nitroindazole did not enhance antidepressant-like activity and plasma nitrite decreasing effect of Ellagic Acid in unstressed mice. Prazosin and p-CPA significantly attenuated antidepressant-like effect of Ellagic Acid (35 mg/kg) in unstressed mice only. Conclusion Thus, Ellagic Acid showed antidepressant-like activity in unstressed mice probably by interaction through adrenergic and serotonergic systems. On the other hand, antidepressant-like activity of Ellagic Acid in stressed mice might be through inhibition of inducible NOS.
