Actually, tracheal homografts can induce long-term stenosis and their growth potential is not known. Thus, in this work porcine tracheal matrices have been obtained by a detergent-Enzymatic Method. The treatment decreased the antigenicity of matrices which were able to support the in vitro adhesion of both chondrocytes and tracheal epithelial cells. On the contrary, only few cells were observed in tracheal matrices prepared with formalin, Thimerosal, and acetone, suggesting that the long-term stenosis occurring in vivo is probably because of an insufficient cell ingrowth. In summary, our results indicate that the detergent-Enzymatic Method allows us to obtain tracheal matrices which can function as a promising support to achieve an in vitro tissue-engineered cell-matrix construct.

Determination of Urinary L-citrulline by Enzymatic Method

A simple, accurate, and rapid Enzymatic Method is proposed for the determination of L -citrulline for its bioconversion by ornithine transcarbamoylase (OTC, EC 2.1.3.3) and carbamate kinase (CK, EC 2.7.2.2). The optimized conditions for the bioconversion were 30–35°C and pH 6–7. The reserved special activity of the immobilized enzymes was up to 95% after these were kept for 60 days at 4°C. The linear range was 2–10 μg/ml (R=0.9987), and the recovery of the samples was more than 99%.

A Novel Enzymatic Method for Production of L-theanine

An Enzymatic Method for optically pure L-theanine production with a new L-aminoacylases-production fungi Cunnighamella echinulata 9980 was developed. The optimum conditions of reaction were: 0.05 M N-Acyl-DL-theanine, 0.04 g/ml pellets, 5×10-4CoCl2, pH7. This was the first report that Cunnighamella echinulata was the source of aminoacylases, and aminoacylase, regardless of its source, was applied in DL-theanine resolution.

We studied where and how many fructosyl groups bind to albumin, and which glycation sites are measured by the Enzymatic Method for GA. We also studied the basic performance of the Enzymatic Method for GA.Methods:Glycated albumin was measured using an Enzymatic Method (Lucica®GA-L, Asahi Kasei Pharma) on a biochemical autoanalyzer. Molecular weights of purified GA and nonglycated albumin were measured by a mass spectrometry system. Two hundred one healthy volunteers with normal results of oral glucose tolerance testing were recruited to determine the reference range in Americans.Results:The present Method measured only glycated amino acids from albumin in serum protein. Native and gelatinized starch were subjected to Enzymatic hydrolysis (pullulanase, 40 U/g/10 h), autoclaved (121 °C/30 min), stored under refrigeration (4 °C/24 h), and lyophilized. The Enzymatic hydrolysis and thermal treatment of starch increased the formation of RS which showed an increase in water absorption and water solubility indexes and a decrease in swelling power due to hydrolytic and thermal process. The process for obtaining RS changed the crystallinity pattern from C to B and increased the crystallinity due to the retrogradation process. RS obtained from hydrolysis showed a reduction in viscosity, indicating the rupture of starch molecules. The viscosity was found to be inversely proportional to the RS content in the sample. The thermal properties of RS increased due to the retrogradation and recrystallization (P<0.05).
