    Abstract:

    Wolbachia is an intracellular alpha-proteobacteria Rickettisial endosymbiont present in most of the arthropods. Wolbachia a parasite in insects invades its host biology in many ways, including cytoplasmic incompatibility, feminization, male killing, parthenogenesis and reproductive fitness advantage. The whole genome sequence of Wolbachia reveals the function of several genes like wsp (Wolbachia surface protein) FtsZ (Wolbachia cell division protein) and ANK (ankarin domain protein). Among these, FtsZ (Filamenting temperature sensitive) protein plays a vital role in the cell division of Wolbachia .In the present study, we characterized the Wolbachia FtsZ gene infected uzifly, Exorista sorbillans a serious menace to silkworm Bombyx mori L. The study revealed the possible implications of FtsZ gene to combat the uzifly E. sorbillans an endoparasite of B. mori L.

    Abstract:

    The reproductive alterations induced by maternally inherited α-proteo-bacteria Wolbachia to their hosts is a well-document- ed phenomenon. In Nesolynx thymus, a gregarious hymenopterous ecto-pupal parasitoid of the uzifly, Exorista sorbillans, diagnostic PCR assay using specific primers revealed the presence of Wolbachia. Following genetic crossing experiments, we observed a female biased sex ratio of 1 : 9.5 at 25°C and 1 : 3 male to female ratio when the populations were exposed to heat shock 33°C for six hours. Furthermore, we found infection polymorphism, where female parasitoids are infected by Wolbachia but males are not infected. In- fected eggs develop into females, whereas uninfected eggs develop parthenogenetically into males. The results are discussed in the context of the possible mechanism of sex-ratio bias caused by Wolbachia.

E. bombycis Louis), a tachinid endoparasitoid of silkworm, Bombyx mori L. is known to exhibit patterns of geographic variation among populations and suspected to consists of several very closely related cryptic species. The Uzi fly harbours Wolbachia which induces cytoplasmic incompatibility, sex ratio distortion, fecundity reduction and abnormalities in oogenesis of uninfected female, irrespective of whether the males are infected or not. In our study we have investigated the effects of ecological factors on Wolbachia induced reproductive incompatibility and fecundity. Crossing experiments suggest high impact of ecological variations onrninduction of cytoplasmic incompatibility and diversification of Uzi fly, E. sorbillans among different populations.

    Abstract:

    Wolbachia are a group of obligate intracellular endosymbionts of a wide range of arthropod species. This microorganism is known for its manipulations of host reproduction that include cytoplasmic incompatibility, parthenogenesis, feminization and male killing. Wolbachia harbours a temperate bacteriophage named phage WO which may be contribute to the reproductive alterations induced in their hosts. Wolbachia and phage WO infections were characterized using polymerase chain reaction (PCR) on the Wolbachia ftsZ (Wolbachia cell division protein) gene and on minor capsid protein genes (orf 2 and orf 7 markers) of the phage WO. This paper results reports that in Exorista sorbillans individuals are infected by Wolbachia and its phage WO. Further, construction of molecular phylogeny of Wolbachia cell division protein (ftsZ) gene revealed double infection of Wolbachia strain A and B in Uzi fly Exorista sorbillans. Thus by evaluating, Wolbachia induced fitness attributes in Uzi fly and its potential role in control of Uzi fly Exorista sorbillans a serious menace of silkworm Bombyx mori L are discussed.

    The uzifly, Exorista sorbillans (=E. bombycis) a tachinid endoparasitoid of silkworm, Bombyx mori L., known to cause considerable damage to the silk industry. Earlier studies on various aspects of the uzifly indicated for possible presence of a number of cryptic species. Though the fly entered into South India 25 years ago, the basis for the existence of cryptic species was not known. To ascertain whether the genome of this parasitoid is differentiating into discrete gene pools in contrasting eco and geo-climatic conditions, an attempt has been made in the present study the molecular profiling of 8 populations (Chitoor, Kuppam, Erode, Bangarapete, Sira, Mandya, Chitradurga, and Kanakapura) from south India with 16 RAPD primers and also to explore the possibility of its endosymbiont Wolbachia as a causative agent as it known to cause cytoplasmic incompatibility in its host uzifly. The hierarchical clustering done on the basis of RAPD data and the Euclidean distance matrix put Chitradurga populations at the maximum distance from other south Indian populations studied and reveals the occurrence of spectacular region-wise pairing with separate clusters. Further, when we have crossed within and between selected populations for the Wolbachia induced reproductive isolation, the results showed the lower level (28–32%) of reproductive isolation within and between populations. The RAPD and crossing data clearly indicates the micro-level divergence in the uzifly, E. sorbillans. The significance of this study with a tachinid fly pest is discussed in the context of understanding the diversification of uzifly and also establishing this pest as a relevant biological material for studying microevolution in nature.

    Cytogenetic mechanism of Wolbachia-induced cytoplasmic incompatibility is investigated in the uzifly, Exorista sorbillans, a parasitoid of silkworm, Bombyx mori. The results re-vealed the condensation of chromatin to form the paternal pronucleus upon the sperm entering in to the egg, and subsequently condensed paternal and maternal pronuclei fused to form the diploid zygote in compatible crosses. In fertilized eggs from incompatible crosses, only the maternal pronucleus had individual chromosomes. The uncondensed paternal pronucleus scattered near maternal pronucleus, and in some eggs reappear as a diffused tangled chromatin mass and tend to get fragmented during the first mitotic division. Moreover, the abnormalities in the first mitotic division extend with the fragmentation and nonorientation of chromosomes on equatorial plane of first metaphase plate. The initiation of chromosome separation in late metaphase was an indication of beginning of anaphase in compatible crosses. In case of incompatible crosses, the chromosome separation had not occurred even at late metaphase/early anaphase leading the failure of development of viable embryo. However, abnormal behavior of paternal chromatin does not interfere with mitotic division of maternal chromosomes. The mechanism is discussed with applied strategy for the management of uzifly, Exorista sorbillans.

  • Tetracycline: Boon for silkworm and bane for uziflies

    Abstract:

    An experiment has been conducted on the dietary supplementation of antibiotic tetracycline to silkworms to understand the effect of the antibiotic on the fitness of silkworms infected from uzifly, Exorista sorbillans, which harbors Worbachia, an alpha-proteo-bacterium. The total reproductive fitness of the uziflies emerged from tetracycline treated silkworm hosts decreases significantly over the flies emerged from untreated silkworm hosts. It was found that the tetracycline treatment has no effect on longevity of the uziflies. The administration of tetracycline can cure the pylariasis in human by eliminating the Wolbachia from nematodes, causing infertility. The results suggest that the tetracycline administration helps in enhancing the fitness of silkworm and decreasing the reproductive fitness of the pest uzifly

The total reproductive fitness of the uziflies emerged from tetracycline treated silkworm hosts decreases significantly over the flies emerged from untreated silkworm hosts. It was found that the tetracycline treatment has no effect on longevity of the uziflies. The administration of tetracycline can cure the pylariasis in human by eliminating the Wolbachia from nematodes, causing infertility. The results suggest that the tetracycline administration helps in enhancing the fitness of silkworm and decreasing the reproductive fitness of the pest uzifly

    In laboratory experiments, pupae of the uzi fly, Exorista sorbillans Wiedemann (Diptera: Tachinidae), an endoparasitoid of the silkworm, Bombyx mori L. (Lepidoptera: Bombycidae) were subjected to UV-irradiation for different durations. Upon adult emergence, irradiated and control flies were crossed in all possible combinations and allowed to parasitise healthy silkworms. Adult emergence, fecundity and longevity of the resulting progeny were recorded. Progeny of irradiated E. sorbillans showed significantly reduced adult emergence, oviposition period and hatchability. The reduction in fecundity was negatively correlated with the duration of exposure to UV radiation. The longevity of uzi flies of both the sexes was also significantly reduced. Au cours des essais en laboratoire, des pupes de la mouche Exorista sorbillans (Diptera: Tachinidae), un endoparasitoïde du ver à soie, Bombyx mori L. (Lepidoptera: Bombycidae), ont été soumises à une irradiation aux rayons ultra-violets (UV) pour des durées différentes d’exposition. A leur émergence, les adultes des mouches irradiées et ceux du témoin étaient croisés par toutes les combinaisons possibles, puis laissés parasiter les vers à soie bien portants. On a pris note du taux d’émergence des adultes, de la fécondité et de la longévité des descendants issus des croisements. Il a été constaté que les descendants nés des sujets irradiés de E. sorbillans montraient une réduction significative du taux d’émergence des adultes, de la durée de ponte et du taux d’éclosion d’oeufs. La réduction de fécondité était négativement corrélée avec la durée d’irradiation aux rayons ultra-violets. La longévité était aussi significativement réduite chez les deux sexes.
