novel epidithiodiketopiperazines as anti viral zinc ejectors of the Feline Immunodeficiency Virus fiv nucleocapsid protein as a model for hiv infection
Bioorganic & Medicinal Chemistry, 2019Co-Authors: Regina Hofmannlehmann, Christopher R M Asquith, Tuomo Laitinen, Antti Poso, Graham J Tizzard, Simon J Coles, Stephen T HiltonAbstract:Abstract Focused libraries of multi-substituted epidithiodiketopiperazines (ETP) were prepared and evaluated for efficacy of inhibiting the nucleocapsid protein function of the Feline Immunodeficiency Virus (FIV) as a model for HIV. This activity was compared and contrasted to observed toxicity utilising an in-vitro cell culture approach. This resulted in the identification of several promising lead compounds with nanomolar potency in cells with low toxicity and a favorable therapeutic index.
-
investigation of the pentathiepin functionality as an inhibitor of Feline Immunodeficiency Virus fiv via a potential zinc ejection mechanism as a model for hiv infection
ChemMedChem, 2019Co-Authors: Christopher R M Asquith, Lidia S Konstantinova, Tuomo Laitinen, Antti Poso, Oleg A Rakitin, Graham J TizzardAbstract:A small diverse library of pentathiepin derivatives were prepared to evaluate their efficacy against the nucleocapsid protein function of the Feline Immunodeficiency Virus (FIV) as a model for HIV, using an in vitro cell culture approach. This study led to the development of nanomolar active compounds with low toxicity.
-
evaluation of substituted 1 2 3 dithiazoles as inhibitors of the Feline Immunodeficiency Virus fiv nucleocapsid protein via a proposed zinc ejection mechanism
ChemMedChem, 2016Co-Authors: Christopher R M Asquith, Lidia S Konstantinova, Tuomo Laitinen, Marina L Meli, Antti Poso, Oleg A Rakitin, Regina HofmannlehmannAbstract:A diverse library of 5-thieno-, 5-oxo-, and 5-imino-1,2,3-dithiazole derivatives was synthesized and evaluated for efficacy against the Feline Immunodeficiency Virus (FIV) as a model for HIV in cells. Several diverse compounds from this series displayed nanomolar activity and low toxicity, representing a potential new class of compounds for the treatment of FIV and HIV.
