    Abstract:

    Insect larvae and adult insects found on human corpses provide important clues for the estimation of the postmortem interval (PMI). Among all necrophagous insects, flesh flies (Diptera: Sarcophagidae) are considered as carrion flies of forensic importance. DNA variations of 17 Malaysian, two Indonesian and one Japanese flesh fly species are analysed using the mitochondrial COI and COII. These two DNA regions were useful for identifying most species experimented. However, characterisation of the species was not sufficiently made in the case of Sarcophaga javanica. Seventeen Malaysian species of forensic importance were successfully clustered into distinct clades and grouped into the six species groups: peregrina, albiceps, dux, pattoni, princeps and ruficornis. These groups correspond with generic or subgeneric taxa of the subfamily Sarcophaginae: Boettcherisca, Parasarcophaga, Liosarcophaga, Sarcorohdendorfia-Lioproctia, Harpagophalla-Seniorwhitea and Liopygia. The genetic variations found in COI and COII can be applied not only to identify the species of forensic importance, but also to understand the taxonomic positions, generic or subgeneric status, of the sarcophagine species.

    Abstract:

    Die Power-Velocity-Analyse ist eine neue ultraschallbasierte Methode zur Quantifizierung von Insuffizienzflussen bei Herzklappenerkrankungen. Sie bestimmt den Regurgitationsfluss direkt aus dem Dopplerspektrum des reflektierten Schalls, da die Energie des reflektierten Schalls bei laminarem Fluss proportional zum Blutvolumen ist. Um Turbulenzen zu vermeiden erfolgte daher bislang die Analyse des Dopplerspektrums am schmalsten, proximalsten Abschnitt des Regurgitationsjets, da hier der Fluss noch weitgehend laminar ist. In vivo stellen sich jedoch haufig Dopplerspektren dar, bei denen die Geschwindigkeiten relativ gleichmasig uber das gesamte Geschwindigkeitsspektrum verteilt sind. Diese Art des Flusses ist nicht eindeutig laminar und nicht eindeutig turbulent. Wir bezeichneten diese Form des Flusses als nicht-laminaren isodirektionalen Fluss. Bislang bestanden keine Daten uber den Einfluss dieses Flussverhaltens auf die Power-Velocity-Analyse. Mit Hilfe eines Flussphantoms quantifizierten wir daher unter kontrollierten Bedingungen unterschiedliche Flussformen mittels Power-Velocity-Analyse. Analysiert wurden neben dem laminaren Fluss zwei Auspragungen des isodirektionalen Flusses. Unsere in vitro Ergebnisse zeigten, dass die PVI-Analyse des gesamten, breiten Dopplerspektrums bei teil-turbulentem, isodirektionalem Fluss keinen Effekt im Sinne einer Uberschatzung der Doppler-Power ergab. Zur Validierung der Daten erfolgte bei 25 Patienten ein Vergleich der Power-Velocity-Integral-Analyse mit der Regurgitationsflussquantifizierung mittels MRT. Bei 5 Patienten stellte sich ein rein laminarer Fluss in der Vena contracta dar, bei 20 Patienten ein isodirektionaler Fluss. Analog zu den in vitro Resultaten zeigt sich auch in vivo bei isodirektionalem Fluss kein Effekt auf die Doppler-Power-Analyse. Zusammenfassend ergab die Auswertung unserer in vivo und in vitro Daten, dass der isodirektionale Fluss bezuglich seines Flussverhaltens eher dem laminaren Fluss entspricht und somit nicht von der Power-Velocity-Analyse ausgeschlossen werden muss.
