The natural course of the Hormone in relationship to the microadenoma and ovarian findings, however, remains largely unknown. CASE: A 40-year-old woman with enlarged multicystic ovaries was nonsurgically diagnosed with an FSH-producing pituitary microadenoma. During her subsequent 12-month follow-up, the serum concentration of E2, but not FSH, and the size of the multicystic ovaries fluctuated dramatically. Both the E2 level and ovarian size were transiently normalized. CONCLUSION: Because of disease-related fluctuations, a reproductive-aged woman with an FSH-producing adenoma did not always present with laboratory values characteristic of ovarian hyperstimulation. This finding points out a possible pitfall in diagnosis and clinical management.

5 alkylated thiazolidinones as Follicle Stimulating Hormone fsh receptor agonists

ShenAbstract:Abstract We prepared analogs of potent thiazolidinone-based Follicle-Stimulating Hormone (FSH) agonists 1 , that is, 3 that contained an additional 5-alkyl substituent. This extra substituent was added to reduce synthetic problems that arose during preparation of analogs of 1 . These compounds ( 3 ) were evaluated in a Chinese hamster ovary (CHO) cell line that expressed recombinant human FSH receptor (FSHR) and a luciferase reporter gene regulated by a cAMP response element (CRE). Selected compounds were also tested on a CHO-cell line that over expressed the FSHR for the ability to induce cAMP production. When the 5-alkyl substituent was a methyl group as in analog 16a , similar FSH activity (i.e., EC 50 = 51 nM, 100% efficacy relative to hFSH) to the analogous 5-hydrogen series compound (e.g., 2 ) was observed; thus, proving that a small 5-alkyl substituent was well tolerated. New derivatives of 3 , in which the potentially hydrolytically labile secondary amide function of 1 (–CONH–) was modified to other moieties (e.g., –CH 2 NH–, –CH 2 S–, and –CH 2 OCONH–), were also prepared and evaluated. These congeners (namely 21 , 22 , and 24 ) also displayed good potency in the CRE-luciferase assay.

Preparation of highly substituted γ-lactam Follicle Stimulating Hormone receptor agonists

ShenAbstract:An unusual combination of Weinreb amidation and Mitsunobu lactam formation was used to prepare highly substituted γ-lactam analogues of a thiazolidinone Follicle Stimulating Hormone receptor agonist. The analogue synthesis was stereoselective and the final products were chemically stable. Biological properties of the target molecules were nearly identical to those of the lead compound.

Several trials have compared the two preparations, but none had sufficient power to detect a clinically meaningful difference in pregnancy rates. The purpose of this study was to determine the clinical pregnancy rates per started cycle by pooling data from randomized trials which compared the use of rFSH and uFSH in treatment cycles using in-vitro fertilization (IVF) or intracytoplasmic sperm injection (ICSI). A thorough search of the literature identified 12 trials which met the inclusion criteria. In four trials, both IVF and ICSI were performed, in seven trials only IVF was performed and in one trial only ICSI was performed. Data were extracted and pooled using the principles of meta-analysis. There was no significant heterogeneity of treatment effect across the trials. The common odds ratio and the risk difference (and their 95% confidence intervals), obtained by pooling the data using a fixed effects model, were 1.20 (1.02-1.42) and 3.7% (0.5-6.9%) respectively, in favour of rFSH. The pregnancy rate with the alpha preparation of rFSH was statistically significantly higher than with uFSH in IVF cycles. The overall conclusion from this meta-analysis is that the use of rFSH in assisted reproduction is preferred over uFSH.

In-vitro and in-vivo biological activity of recombinant yellowtail kingfish (Seriola lalandi) Follicle Stimulating Hormone

Incubation of ovarian and testicular fragments with the recombinant Hormone stimulated E2 and 11-KT secretion, respectively. In-vivo trial in immature female YTK resulted in a significant increase of plasma E2 levels and development of oocytes. In males at the early stages of puberty, advancement of spermatogenesis was observed, however plasma 11-KT levels were reduced when administered with rytkFsh.
