The influence of lactic acid bacteria (LAB)-fermented soy−skim milk on mucosal integrity in a Gastric mucosal Lesion rat model was also investigated. After 24 h, cell densities of L. paracasei subsp. paracasei NTU 101 and L. plantarum NTU 102 fermented in 75% soy milk and 25% milk (optimal condition) were 1.2 × 109 and 2.5 × 109 CFU/mL. After 180 days at 4 °C, the cell densities of the freeze-dried powders of the fermented soy−skim milks were 1 × 109 CFU/g; slight variations in pH and acidity were observed. Pylorus ligation with acidified ethanol treatment was used as the Gastric Lesion animal model. LAB-fermented soy−skim milk reduced the Gastric Lesion index and the lipid peroxides (LPO) of Gastric mucosa and serum. The extract inhibited Gastric Lesions induced by HCl/ethanol in female rats. A dose–response effect study showed that the ED50 was 150 mg/kg b.w. Extract doses higher than 100 mg/kg b.w. were more effective than 30 mg/kg of lansoprazol in inhibiting Gastric Lesions. A methanolic fraction (257.12 mg/kg) which reduced Gastric Lesion at 88.20% is likely to contain the active principle of the antiulcer effect. No signs of acute toxicity were observed when mice were treated with extract dose up to 2000 mg/kg b.w. A chemical analysis of the extract allowed the identification of phenolic compounds as the major components. Glycosylated quercetin, amentoflavone derivate and a tetramer of proanthocyanidin were identified by liquid chromatography coupled to mass spectrometry. The level of total phenolics in the extract was evaluated at 35.5% and flavonoid content was 2.58%.

Per os (p.o.) administration of PLE (300, 1000 mg/kg) exhibited significant anti-inflammatory activity in acute (carrageenin, histamine, 5-hydroxytryptamine and prostaglandin E 2 -induced hind paw edema), subcute (kaolin-carrageenin and formaldehyde-induced hind paw edema) and chronic (cotton pellet granuloma) models of inflammation. PLE did not show any sign of toxicity and mortality up to a dose level of 10.125 g/kg, p.o. in mice. Both acute as well as chronic administration of PLE (100, 300 and 1000 mg/kg, p.o.) did not produce any Gastric Lesion in rats. These results indicate that PLE possesses significant anti-inflammatory activity without ulcerogenic activity suggesting its potential as an anti-inflammatory agent for use in the treatment of various inflammatory diseases.

The extract did not show any sign of toxicity up to a dose of 10.125 g/kg, p.o. in mice. Significant anti-inflammatory activity was observed in carrageenin (acute), kaolin-carrageenin (subacute), formaldehyde (subacute)-induced paw oedema and cotton pellet granuloma (chronic) tests in rats. The extract did not induce any Gastric Lesion in both acute and chronic ulcerogenic tests in rats. Thus, the present study demonstrated that S. cumini bark extract has a potent anti-inflammatory action against different phases of inflammation without any side effect on Gastric mucosa.
